The anti-dengue virus properties of statins may be associated with alterations in the cellular antiviral profile expression

Dengue virus (DENV) susceptibility to cholesterol depleting treatments has been previously reported. There are numerous questions regarding how DENV seizes cellular machinery and cholesterol to improve viral production and the effect of cholesterol sequestering agents on the cellular antiviral response. The aim of the present study was to evaluate the mechanisms involved in the negative regulation of DENV replication induced by agents that diminish intracellular cholesterol levels. Cholesterol synthesis was pharmacologically (fluvastatin, atorvastatin, lovastatin, pravastatin and simvastatin treatment) and genetically (HMGCR-RNAi) inhibited, in uninfected and DENV2-infected hepatoma Huh-7 cells. The cholesterol levels, DENV titer and cellular antiviral expression profile were evaluated. A reduction in the DENV titer, measured as plaque forming units, was observed in DENV-infected cells following 48 h treatment with 10 mu M fluvastatin, 10 mu M atorvastatin, 20 mu M lovastatin and 20 mu M simvastatin, which achieved 70, 70, 65 and 55% DENV2 inhibition, respectively, compared with the untreated cells. In addition, the cytopathic effect was reduced in the statin-treated DENV-infected cells. Statins simultaneously reduced cholesterol levels at 48 h, with the exception of DENV2 infected cells. Genetic inhibition of cholesterol synthesis was performed using RNA interference for 3-hydroxy-3-methylglu-taryl-CoA reductase (HMGCR-siRNA), which indicated a slight reduction in DENV2 titer at 48 h post-infection, however, with no significant reduction in cholesterol levels. In addition, DENV2 infection was observed to augment the intracellular cholesterol levels in all experimental conditions. Comparison between the cellular antiviral response triggered by DENV2 infection, statin treatment and HMGCR-siRNA in infected, uninfected, treated and untreated Huh7 cells, showed different expression profiles for the antiviral genes evaluated. All downregulating cholesterol agents evaluated reduced the expression of genes associated with cellular immune and pro-inflammatory responses. These results indicate that statin-mediated downregulation of DENV2 infectious particles number is independent of cholesterol levels and it is partially mediated by the modulation of the cellular antiviral profile.

Summary: The study in Mexico on academic professionals during the COVID-19 quarantine showed significant statistical correlations among many variables, with perceived stress and anxiety falling mainly in the Absent to Mild category, while the danger section of the ACSS fell into the Moderate category. The high level of resilience generated during the quarantine appears to be a predictor of how well academic professionals have adapted to cope with stress.

Donors aged 18-49 were more likely to be seropositive, and females had a 1.46-fold higher prevalence compared to males. The majority of SARS-CoV-2 cases were recorded in Monterrey city.

CAR-NK Cells for Cancer Therapy: Molecular Redesign of the Innate Antineoplastic Response

Summary: The Chimeric Antigen Receptor (CAR) technology is a powerful tool in synthetic biology, with the potential to be applied in T and NK cells. While CAR T cell therapy has shown success in clinical trials, there are still challenges to address regarding safety and long-term efficacy. NK cells, with their intrinsic advantages over T cells, offer a promising alternative for treating tumors. Early clinical trials suggest that CAR NK cells may have improved safety profiles and can be used off-the-shelf due to their HLA independence. However, more data is needed in this field. Combining NK cell therapy with immunological checkpoint inhibitors and genetic circuits may further enhance its effectiveness. This review provides an overview of CAR technology for modifying NK cells and its translation from preclinical studies to early clinical trials in immunotherapy.

Genomic Characterization by Whole-Exome Sequencing of Hypermobility Spectrum Disorder

Summary: Currently, there is no established genetic basis to differentiate hypermobility spectrum disorders (HSD) from hypermobile Ehlers-Danlos syndrome (hEDS). This study used whole-exome sequencing to identify DNA mutations in patients diagnosed with generalized HSD. Three genes (MUC3A, RHBG, and ZNF717) were found to be mutated in all five patients evaluated. Functional enrichment analysis revealed the extracellular matrix (ECM) structural constituent as the primary overrepresented molecular function. Comparison with other studies supported the hypothesis of two distinct diseases sharing a common genetic background. Five potential biomarkers for HSD were identified.

Summary: This study describes an IRD patient with ABCA4 and USH2A pathogenic biallelic mutations resulting from paternal uniparental disomy on chromosome 1. This is the first known case of a patient with confirmed UPD for two recessively mutated IRD genes, expanding on the genetic heterogeneity of IRDs and highlighting the importance of UPD as a mechanism of autosomal recessive disease in non-consanguineous parents.

Summary: In this study, a biosensor based on gold nanoparticles and antibody was designed for the detection of dengue virus in vitro, showing significant differences in size and characteristics between different conjugates and naked AuNPs. The biosensor was further used in the development of a lateral flow assay for detecting flavivirus envelope protein in isolated DENV samples, with promising results for future dengue detection.

Summary: Maternal overnutrition and exposure to high-energy diets during pregnancy can lead to metabolic and immune alterations in offspring, including obesity and inflammation. This study suggests that maternal nutritional programming increases susceptibility to ghrelin-induced overeating in offspring, particularly under chronic stress conditions. The findings also indicate a relationship between cortisol accumulation and ghrelin sensitivity in the offspring.

Nursing Staff Mortality During the Covid-19 Pandemic, Scoping Review

Summary: During the COVID-19 pandemic, nurses face a higher risk of infection and mortality. This study reviewed information on the mortality of nursing personnel from different countries. The results showed that nursing personnel had a higher mortality rate compared to physicians and other healthcare workers. The average age of deceased nurses was 43 years, with higher rates among men. Inadequate planning and lack of personal protective equipment were identified as contributing factors to the contagion and deaths.
