Prostate inflammation has been suggested as an etiology for benign prostatic hyperplasia (BPH). We show that decreased expression of the androgen receptor (AR) in luminal cells of human BPH specimens correlates with a higher degree of regional prostatic inflammation. However, the cause-and-effect relationship between the two events remains unclear. We investigated specifically whether attenuating AR activity in prostate luminal cells induces inflammation. Disrupting luminal cell AR signaling in mouse models promotes cytokine production cell-autonomously, impairs epithelial barrier function, and induces immune cell infiltration, which further augments local production of cytokines and chemokines including Il-1 and Ccl2. This inflammatory microenvironment promotes AR-independent prostatic epithelial proliferation, which can be abolished by ablating IL-1 signaling or depleting its major cellular source, the macrophages. This study demonstrates that disrupting luminal AR signaling promotes prostate inflammation, which may serve as a mechanism for resistance to androgen-targeted therapy for prostate-related diseases.

Pathologic HIF1α signaling drives adipose progenitor dysfunction in obesity

Mengle Shao, Chelsea Hepler, Qianbin Zhang, Bo Shan, Lavanya Vishvanath, Gervaise H. Henry, Shangang Zhao, Yu A. An, Yibo Wu, Douglas W. Strand, Rana K. Gupta

Summary: Regional differences in adipose precursor cells' response to obesity are driven by HIF alpha-induced PDGFR beta signaling, leading to inhibitory phosphorylation of PPAR gamma. Suppression of HIF alpha promotes adipogenesis and improves metabolic health in obesity, with potential for pharmacological targeting of APCs to enhance metabolic health.

Progenitors in prostate development and disease

Diya B. Joseph, Anne E. Turco, Chad M. Vezina, Douglas W. Strand

Summary: The prostate develops through epithelial budding and branching processes during fetal and postnatal stages, showing remarkable regenerative capacity. Benign prostatic hyperplasia (BPH) and prostate cancer (PCa) may involve androgen-independent epithelial progenitors. Multiple castration-insensitive epithelial cell types exist in the proximal area of the prostate, with not all acting as progenitors.

Summary: Through single-cell RNA sequencing, different stromal cell types in the human prostate were identified, including three smooth muscle subtypes and two fibroblast subtypes. The research provides insight into the molecular identity and anatomy of discrete stromal cell types in the prostate, laying the foundation for new therapeutic targets in benign prostatic hyperplasia.

Summary: The void spot assay is a cost-effective method for evaluating and quantifying mouse urinary voiding phenotypes. It can differentiate voiding behaviors between different experimental groups and accurately classify mice based on voiding dysfunction etiology. The approach of using principal components analysis and hierarchical clustering can help identify specific urinary phenotypes in mice.

Diya B. Joseph, Gervaise H. Henry, Alicia Malewska, Jeffrey C. Reese, Ryan J. Mauck, Jeffrey C. Gahan, Ryan C. Hutchinson, James L. Mohler, Claus G. Roehrborn, Douglas W. Strand

Summary: Benign prostatic hyperplasia (BPH) is a common condition in aging men, treated with 5-alpha reductase inhibitors (SARI) to reduce prostate volume and improve lower urinary tract symptoms. However, the histologic response to SARI treatment is heterogeneous, with potential need for surgical intervention.

Summary: This study characterized the pathways associated with failure of medical therapy for BPH/LUTS. It found that 5-alpha reductase inhibitor (5ARI) therapy continued to inhibit androgenesis, but also increased tissue levels of glucocorticoids (GC). This GC activation served as a master regulator of prostatic branching and growth.

Summary: Prostate cancer is highly heritable and researchers have identified multiple risk alleles associated with the disease.
