Lipidomic Approaches towards Deciphering Glycolipids from Microalgae as a Reservoir of Bioactive Lipids

In recent years, noteworthy research has been performed around lipids from microalgae. Among lipids, glycolipids (GLs) are quite abundant in microalgae and are considered an important source of fatty acids (FAs). GLs are rich in 16- and 18-carbon saturated and unsaturated fatty acids and often contain polyunsaturated fatty acids (PUFAs) like n-3 -linolenic (ALA 18:3), eicosapentaenoic (EPA, 20:5) and docosahexaenoic (DHA, 22:6). GLs comprise three major classes: monogalactosyldiacyl glycerolipids (MGDGs), digalactosyl diacylglycerolipids (DGDGs) and sulfoquinovosyl diacylglycerolipids (SQDGs), whose composition in FA directly depends on the growth conditions. Some of these lipids are high value-added compounds with antitumoral, antimicrobial and anti-inflammatory activities and also with important nutritional significance. To fully explore GLs' bioactive properties it is necessary to fully characterize their structure and to understand the relation between the structure and their biological properties, which can be addressed using modern mass spectrometry (MS)-based lipidomic approaches. This review will focus on the up-to-date FA composition of GLs identified by MS-based lipidomics and their potential as phytochemicals.

Light modulates the lipidome of the photosynthetic sea slug Elysia timida

Summary: A study on the sea slug Elysia timida revealed that light intensity influences the degradation of stolen chloroplasts (kleptoplasts), with older kleptoplasts being targeted for degradation. The lipidome of E. timida showed differences under different light treatments, indicating a light-driven remodelling of the lipidome.

This study proposes a miniaturized automated method to measure NPs concentration, which is fast, simple, and suitable for research and quality control operations. The method utilizes the measurement of light scattering caused by NPs passing through an optical path, achieving accurate and efficient quantification of NPs concentration.

Summary: Phosphatidylinositols (PIs) are complex lipids that can be modified by reactive oxygen and nitrogen species (RNS) to produce a variety of modified lipid products. This study used mass spectrometry to characterize the fragmentation signature of nitrated and nitroxidized PIs, allowing for the identification of these modified lipid products accurately. The findings of this study provide new opportunities to understand the pathophysiological roles of nitrated and nitroxidized PI derivatives at the cellular and tissue levels.

Summary: This study aimed to characterize the chemical structure of water-soluble polysaccharides from Saccharina latissima and evaluate their immunostimulatory and hypocholesterolemic activities. It was found that laminarans and two fractions of fucose-containing sulphated polysaccharides showed immunostimulatory activity on B lymphocytes, and one of the fractions exhibited a significant effect in reducing cholesterol bioaccessibility.

Potato Chips Byproducts as Feedstocks for Developing Active Starch-Based Films with Potential for Cheese Packaging

Summary: The brownish water-soluble extract (BrE) from the potato chip industry, which is usually landfilled, was found to have the potential to be used in active packaging. When incorporated into starch-based films, BrE improved their properties such as color, transparency, traction resistance, elasticity, antioxidant activity, hydrophilicity, water vapor barrier, and UV-light barrier. Moreover, the packaging film containing BrE showed better preservation effects on sliced cheese compared to the commonly used polyamide/polyethylene film.

A Perspective on the Link between Mitochondria-Associated Membranes (MAMs) and Lipid Droplets Metabolism in Neurodegenerative Diseases

Summary: Neurodegenerative diseases affect millions of people worldwide, but currently there is no cure for these disorders. Research on the endoplasmic reticulum-mitochondria interaction, called mitochondria-associated membranes (MAMs), is gaining attention in understanding the role of lipid metabolism in neurodegeneration. MAMs are involved in lipid synthesis, transport, and degradation, and changes in lipid metabolism can contribute to the development of neurodegenerative diseases. This review highlights the importance of lipid metabolism and MAMs in neurodegenerative disorders.

Prevention of UVB Induced Metabolic Changes in Epidermal Cells by Lipid Extract from Microalgae Nannochloropsis oceanica

Summary: The lipid extract from Nannochloropsis oceanica effectively protects human keratinocytes from UVB-induced damage by inhibiting cell death, reducing oxidative enzyme activity and ROS levels, and enhancing antioxidant enzyme activity. It also silences overexpressed membrane transporters caused by UVB radiation and decreases inflammation levels, stabilizing the natural metabolism of skin cells.

Revealing the polar lipidome, pigment profiles, and antioxidant activity of the giant unicellular green alga, Acetabularia acetabulum

Summary: Marine algae are important sources of high-value compounds. This study focused on the lipidome and pigment profile of Acetabularia acetabulum, a macroscopic green single-celled alga. Analysis identified 16 polar lipid classes and 191 lipid species, as well as carotenoids and chlorophylls. The lipid extracts showed high antioxidant activity.

Summary: Autoimmune diseases like SLE and SS involve immune system dysregulation. Lipids have potential as biomarkers for improved diagnosis and prediction in these diseases. This study identified differences in lipid profiles in SLE and SS patients, including potential biomarkers like sphingomyelin and ceramide in SS and phosphatidylserine in SLE. Further research is needed to understand the role of lipids in the pathological mechanisms of these diseases.

Mitochondrial Metabolism Drives Low-density Lipoprotein-induced Breast Cancer Cell Migration

Summary: Most cancer-related deaths are caused by metastases. A study showed that low-density lipoprotein (LDL)-cholesterol, a systemic factor, promotes the migration and invasion of triple-negative breast cancer (TNBC) cells by inducing the accumulation of lipid droplets and the expression of CD36. LDL also increases mitochondrial mass and network spread in migrating cells, leading to the dependence on fatty acids for mitochondrial respiration. Furthermore, LDL treatment increases reactive oxygen species (ROS) production. Blocking CD36 or ROS reverses the LDL-induced cell migration and metabolic adaptations. This research reveals a new vulnerability in metastatic breast cancer driven by LDL-induced reprogramming of mitochondrial metabolism.

Summary: This study found that the lipid extract derived from Nannochloropsis oceanica microalgae can mitigate the detrimental effects of UVB radiation on skin cells by increasing the concentration of anti-inflammatory and antioxidant polyunsaturated fatty acids, and reducing the production of pro-inflammatory substances and the expression of receptors.
