Inhibitory effect of Gardenblue blueberry (Vaccinium ashei Reade) anthocyanin extracts on lipopolysaccharide-stimulated inflammatory response in RAW 264.7 cells

Blueberries are a rich source of anthocyanins, which are associated with health benefits contributing to a reduced risk for many diseases. The present study identified the functional Gardenblue blueberry (Vaccinium ashei Reade) anthocyanin extracts (GBBAEs) and evaluated their capacity and underlying mechanisms in protecting murine RAW 264.7 cells from lipopolysaccharide (LPS)-stimulated inflammation in vitro. Enzyme-linked immunosorbent assay (ELISA) kit results showed that GBBAEs significantly inhibited the production of nitric oxide (NO), prostaglandin E-2 (PGE(2)), interleukin-6 (IL-6), IL-1 beta, and interferon-gamma (INF-gamma). Real-time polymerase chain reaction (PCR) analysis indicated that the mRNA expression levels of IL-6, IL-1 beta, tumor necrosis factor-alpha (TNF-alpha), monocyte chemoattractant protein-1 (MCP-1), and cyclooxygenase 2 (COX-2) were suppressed in LPS-stimulated RAW 264.7 cells. Additionally, Western blot analysis was used to evaluate the relative protein expression levels of COX-2 and nuclear factor-kappa B p65 (NF-kappa Bp65). All these results suggested the potential use of GBBAEs as a functional food for the treatment of inflammatory diseases.

Enzymolysis peptides from Mauremys mutica plastron improve the disorder of neurotransmitter system and facilitate sleep-promoting in the PCPA-induced insomnia mice

Summary: Through experiments on PCPA-induced insomnia model, enzymolysis peptides from PMM were found to improve neurotransmitter system disorder, restore over-expression of 5-HT1A receptor and GABAA receptor alpha 1-subunit, and show a good sleep-promoting effect. The unique amino acid composition, sequence, and glycosylation modification of PMM peptides may be the key factors for their activity, providing a basis for the development of sleep-promoting peptide products.

Triterpenoid acids from medicinal mushroom Inonotus obliquus (Chaga) alleviate hyperuricemia and inflammation in hyperuricemic mice: Possible inhibitory effects on xanthine oxidase activity

Summary: The study explored the hypouricemic effect of triterpenoid acids from Inonotus obliquus on hyperuricemia mice and deciphered the underlying xanthine oxidase inhibitory mechanism. The triterpenoids were found to efficiently lower uric acid levels and hepatic XO in hyperuricemia mice, indicating potential benefits in controlling hyperuricemia and gout. The findings suggest that Inonotus obliquus could serve as a promising supplement for functional foods or medicines in the treatment of hyperuricemia and gout.

Inhibition on α-Amylase and α-Glucosidase of Polysaccharides from Inonotus obliquus and Effects on Delaying the Digestion of Polysaccharides-Dough system

Summary: In this study, three alcohol precipitation polysaccharides fractions (IOP40, IOP60, and IOP80) were isolated from Inonotus obliquus polysaccharides (IOP). In vitro starch digestion experiments showed that IOP40 and IOP60 had stronger inhibitory effects compared to crude polysaccharides. Inhibition kinetics revealed that IOP60 had a reversible and anti-competitive inhibitory effect on alpha-glucosidase. In vivo studies confirmed that IOP60 could inhibit the increase in postprandial blood glucose level. Further purification of IOP60 resulted in IOP-2S, which showed a triple helix structure. The findings suggest that I. obliquus polysaccharides may be used in functional foods for postprandial hyperglycemia control.

Diacylated anthocyanins from purple sweet potato (Ipomoea batatas L.) attenuate hyperglycemia and hyperuricemia in mice induced by a high-fructose/high-fat diet

Summary: Studies have shown that diacylated anthocyanins from purple sweet potato (diacylated AF-PSPs) have dual regulatory effects on hyperuricemia and hyperglycemia induced by a high-fructose/high-fat diet. The effects of diacylated AF-PSPs include alleviating hyperuricemia, regulating liver XO activity, lipid accumulation, relevant renal transporters, reducing body weight, improving lipid metabolism disorder, liver oxidative stress, enhancing insulin utilization and sensitivity, lowering blood sugar and reducing hyperglycemia, restoring mRNA levels related to renal fructose metabolism, reducing kidney injury and inflammation.

Effective Therapeutic Verification of Crocin I, Geniposide, and Gardenia (Gardenia jasminoides Ellis) on Type 2 Diabetes Mellitus In Vivo and In Vitro

Summary: Gardenia has been highly valued as a food herbal medicine due to its bioactive compounds, but the mechanism underlying its hypoglycemic effect is unknown. This study evaluated the effect of gardenia extracts on type 2 diabetes and compared the efficacy of different purified fragments. The results showed that geniposide had a better inhibitory effect on T2DM in animal experiments, while crocin I had a better effect in vitro. These findings suggest potentially different mechanisms between geniposide and crocin I in treating T2DM.
