Cleland, Kamyar Arasteh, Ellen Caniglia, Yu Chen, Gavin Bart, Jean Pierre Moles, Vinh Vu Hai, Roselyne Vallo, Catherine Quillet, Delphine Rapoud, Mai Le Sao, Laurent Michel, Didier Laureillard, Maria R. Khan

Summary: There has been a significant increase in methamphetamine use among persons who use drugs in Vietnam in the last 5-10 years. The study found that methamphetamine use is associated with unsuppressed HIV viral load among people who inject drugs (PWID) despite high levels of antiretroviral therapy (ART) adherence. Further research is needed to better understand these relationships, with emphasis on potential biological pathways that may interact with ART.

Differential effects of Usutu and West Nile viruses on neuroinflammation, immune cell recruitment and blood-brain barrier integrity

Orianne Constant, Ghizlane Maarifi, Jonathan Barthelemy, Marie-France Martin, Bachirou Tinto, Giovanni Savini, Philippe Van de Perre, Sebastien Nisole, Yannick Simonin, Sara Salinas

Summary: This study characterizes and compares the infectivity, innate immunity and inflammatory responses, and immune cell recruitment of Usutu and West Nile viruses in the blood-brain barrier. Both viruses can infect the blood-brain barrier, but with different consequences. Usutu virus does not strongly affect the endothelium integrity but induces neuroinflammation and immune cell recruitment. In contrast, West Nile virus infection results in endothelium impairment, potent neuroinflammation, and immune cell recruitment.

Summary: DFNA68 is a rare subtype of autosomal dominant nonsyndromic hearing impairment caused by alterations in the HOMER2 gene. A novel variation (c.1064 A > G) in the HOMER2 gene was identified in a Sicilian family suffering from progressive dominant hearing loss. In vivo studies using a zebrafish model and behavioral tests confirmed the deleterious impact of this variation on hearing function.

Boris Dmitrenko, Vincent Gatinois, Morgana D'Ottavi, Ahmed El Mouatani, Pauline Bouret, Segolene Debiesse, Michel Mondain, Mohamed Akkari, Maxime Dallemagne, Franck Pellestor, Philippe Vande Perre, Jean-Pierre Moles

Summary: This study aims to determine the quantity and persistence of maternal microchimeric cells using cytogenetic fluorescent in situ hybridization (FISH) and automated microscopy techniques. The results show that the presence and density of maternal microchimeric cells are influenced by two main factors: the age of the child and whether they were breastfed. Additionally, the study also found a relationship between breastfeeding and infant health.

Jean-Madeleine de Sainte Agathe, Mathilde Filser, Bertrand Isidor, Thomas Besnard, Paul Gueguen, Aurelien Perrin, Charles Van Goethem, Camille Verebi, Marion Masingue, John Rendu, Mireille Cossee, Anne Bergougnoux, Laurent Frobert, Julien Buratti, Elodie Lejeune, Eric Le Guern, Florence Pasquier, Fabienne Clot, Vasiliki Kalatzis, Anne-Francoise Roux, Benjamin Cogne, David Baux

Summary: SpliceAI-visual is a free online tool based on the SpliceAI algorithm that complements the traditional SpliceAI analysis by addressing its drawbacks. It manipulates raw scores instead of delta scores, provides user-friendly graphical presentation, and can handle complex variants such as delins. Using SpliceAI-visual, we demonstrate its relevance in the assessment/modulation of the PVS1 classification criteria and its ability to elucidate complex splicing defects.
