Simultaneous extraction and determination of free and conjugated phytosterols in tobacco

Acid hydrolysis and alkaline saponification were incorporated into a microwave-assisted extraction process for the simultaneous extraction of free and conjugated phytosterols from tobacco. The crude extract of the microwave-assisted extraction was purified by C-18 solid-phase extraction and then determined by high-performance liquid chromatography. Phytosterols of cholesterol, ergosterol, stigmasterol, campesterol, and beta-sitosterol were determined by chromatographic quantification. The multiple parameters of microwave-assisted extraction were optimized by a uniform design method. The optimal ratio of extraction ethanol solvent to tobacco mass was 30 mL/g. The microwave-assisted extraction acid hydrolysis was carried out in sulfuric acid medium by heating for 10min at 55 degrees C. The microwave-assisted extraction alkaline saponification was performed after adding excessive sodium hydroxide by heating another 10min. The repeatability of the proposed method was acceptable with recoveries from 69.68 to 88.17% for the phytosterols. Five target phytosterols were all found in the tobacco samples, and the contents were significantly different in samples from different producing areas.

Uncovering the Metabolic Mechanism of Salidroside AlleviatingMicroglial Hypoxia Inflammation Based on Microfluidic Chip-MassSpectrometry

Summary: This study elucidated a new mechanism of Salidroside in reducing hypoxia-induced inflammation in microglia by regulating cellular energy metabolism. The hypoxic microenvironment of BV2 microglial cells was simulated in vitro using a cell microfluidic chip-mass spectrometry (CM-MS) system, and the real-time change of cell metabolites was investigated. The levels of hypoxia-related factors were also detected to confirm the metabolic mechanism of BV2 cells under hypoxia. The study found that Salidroside could invert the changes in cellular energy metabolism and identified two metabolites of Salidroside.

Viscosity-Based Flow Sensor on Paper for Quantitative and Label-Free Detection of α-Amylase and Its Inhibitor

Isomer-specific biomarker discovery in multiple myeloma with dual-derivatized N-glycans

Summary: Protein glycosylation, an important post-translational modification, plays a vital role in physiological processes. N-glycans exhibit structural diversity in linkages and branching styles and structure-specific glycan profiling can provide more potential biological information. In this study, N-glycans released from human serum samples were profiled using nanoLC-ESI-MS with PGC as a stationary phase. Alpha 2-3 neuraminidase was applied for distinguishing sialic acid linkage types. Significant differences in abundance were observed for 20 isomeric N-glycans between multiple myeloma cases and healthy controls. ROC analysis showed high diagnostic accuracy for MM, with 6 AUCs exceeding 0.80. PCA was used to establish differences among sample sets. Furthermore, these specific isomers have important clinical application value for early detection of multiple myeloma.

In-situ monitoring calcium signaling through tumor microtubes for single cell-cell communication via an open microfluidic probe
