Summary: In this research, the role of the SENP1-Sirt3 signaling pathway and its correlation with mitochondrial dysfunction in acute kidney injury (AKI) was investigated. It was found that activating the AMPK pathway protected proximal tubular epithelial cells (PTECs) from apoptosis. Additionally, reducing mtROS levels using NAC or MitoQ alleviated AKI. Metabolomics analysis revealed increased antioxidants and metabolic shifts in Sirt3 KR mice during AKI.

Summary: Metabolic associated fatty liver disease is a major cause of chronic liver disease worldwide. In this study, we found that perillartine derived from Perilla frutescens (L.) improved fat deposition and glucose homeostasis in hepatocytes. In vitro, perillartine reduced lipid synthesis and transport, and increased mitochondrial content in hepatocytes. In vivo, perillartine reduced body weight gain, improved glucose metabolism, and altered gut microbial composition in mice fed a high-fat diet. Perillartine was also identified as a potential inhibitor for ROR gamma, a transcriptional activator involved in lipid accumulation and glucose metabolism. These findings suggest that perillartine could be a new candidate for the treatment of obesity and metabolic associated fatty liver disease.

Psyllium enriches the high-fat diet and provides strong protection against gut inflammation and metabolic consequences induced by obesity.

Summary: This study investigates the mechanism of toxicity induced by clioquinol (CQ) and its related analogues, and suggests that oxidative stress is the core mechanism of CQ-induced toxicity. The expression of the antioxidant protein NQO1 plays a crucial role in protecting against CQ-induced toxicity. These findings were replicated in zebrafish, indicating that the geographic restriction of subacute myelo-optic neuropathy (SMON) cases to Japan could be explained by the higher prevalence of an inactivating NQO1 polymorphism in the Japanese population.

Summary: In this study, isoginkgetin (IGK) was found to protect against obesity-induced metabolic cardiomyopathy by activating the Nrf2 signaling pathway. IGK stabilized Nrf2 protein to protect against oxidative stress and mitochondrial damage induced by obesity. These findings highlight the potential therapeutic use of IGK in treating obesity-induced cardiomyopathy.
