Summary: The brush border is a specialized structure on the apical surface of epithelia that greatly increases absorptive surface area, but the transcriptional regulatory networks controlling brush border genes are not fully understood. This study identifies HNF4 as a conserved regulator of the brush border gene program in multiple organs, such as intestine, kidney, and yolk sac, and reveals its role in chromatin looping between promoters and enhancers in the intestine.

Summary: Researchers used live cell imaging to study the interaction between polarized epithelial cells and Salmonella. They found that microvilli effacement involves two mechanisms, namely F-actin depolymerization mediated by villin and G-actin consumption by formation of membrane ruffles. Furthermore, they demonstrated the dual functions of SopE in F-actin remodeling, where SopE triggers F-actin polymerization and ruffle formation through Rac1 activation, and depolymerizes F-actin through PLC gamma and villin activation.

Summary: This study reveals the cellular mechanisms underlying the activation of PKC beta II through loss-of-function experiments and mutant analysis. The phosphorylation of PKC beta II at the T500 site mediated by PDK1 is required for its nuclear entry and subsequent ubiquitination by Mdm2 in response to phorbol ester stimulation. Ubiquitination of PKC beta II in turn is required for inducible phosphorylation at the T500 site mediated by PDK1 in the nucleus. After translocation out of the nucleus, PKC beta II interacts with actin, undergoes inducible phosphorylation at the T641 site mediated by mTORC2, interacts with clathrin, and translocates to the plasma membrane.
