Summary: In this study, the potential protective effect of caffeic acid phenethyl ester (CAPE) on ototoxicity caused by gentamicin in a rat model was investigated. The results showed that CAPE can ameliorate hearing deterioration caused by gentamicin ototoxicity and protect the cochlear cells from apoptosis due to its strong antioxidant effect.

Summary: The chromatographic behavior of mixed-mode and hydrophilic interaction liquid chromatography (HILIC) was assessed in this study for the separation of metabolites from glycolysis and pentose phosphate pathways. Optimization of chromatographic conditions allowed for selective targeted analysis of glycolytic phosphorylated carbohydrate metabolites using liquid chromatography with tandem mass spectrometry (MS/MS). The separation of pentose phosphate isomers was achieved using mixed-mode HILIC/SAX and HILIC BEH Amide columns. HILIC-MS/MS showed challenges in separating hexose monophosphate metabolites, but the BEH Amide column provided the best results for this separation.

Summary: Ion-pairing reversed-phase liquid chromatography (IP-RPLC) is commonly used for the analysis of nucleic acid related compounds. However, the high concentration of ion-pairing reagent in IP-RPLC is not suitable for coupling with mass spectrometry (MS). This study explored the use of a PBT-bonded silica-based stationary phase for the separation of Patisiran without the need for ion-pairing reagents. The method allowed for MS and MS/MS characterization without contamination or related issues.

Summary: In this work, new polymeric chiral stationary phases were prepared and compared with brush-type chiral stationary phases. The polymeric CSPs showed similar enantioselectivity but lower chromatographic efficiencies compared to the brush phases. Oxidation of the polymeric CSPs reduced retention times and allowed for lower counterion concentrations in liquid chromatography-electrospray ionization-mass spectrometry applications. In addition, cryogenic cooling of the column enabled baseline separation of estradiol.

Summary: In this study, seven commercially available UHPLC columns were systematically evaluated for their retention and isomer selectivity of branched chain fatty acids (BCFAs). It was found that IG-U column showed the best isomer selectivity for short and medium-chain BCFA isomers, while RP C18 column had good selectivity for long-chain BCFA isomers. Furthermore, an untargeted isomer-selective BCFA profiling method was developed using UHPLC-ESI-MS/MS without pre-column derivatization.

The interactions, conformational flexibilities, and XB donor capacities of CF2X moieties are analyzed. Furthermore, the implementation of CF2X acetamides in molecular design and their binding modes in JNK3 are investigated.

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.

The optimized experimental parameters result in low dissolved organic carbon content and residual acidity concentration. The accuracy of the method is confirmed through the analysis of certified reference materials, and its environmental friendliness is validated using the AGREE metric. Principal component analysis is used to classify the medicinal herbs based on their inorganic constituent concentrations.

The results showed that PEG600, PEG2K, PEG4K and PEG10K exhibited low toxicity and had limited cellular entry at concentrations of 5-1200 μg/mL. Additionally, the environmental friendliness of the developed analytical methods was evaluated.

Summary: This study employed network pharmacology-based analysis and reverse molecular docking to investigate the molecular targets and pathways associated with gynecological disorders and the potential therapeutic effects of fenugreek constituents. The results highlighted the potential of steroidal saponins, isoflavones, and flavones as active constituents of fenugreek with implications for lactation, reproductive processes, and estrogenic activity. Animal model study further supported the presence of these bioactive compounds and their effects on body weight, serum prolactin levels, serum estradiol levels, and ovarian histological structure.
