This work deals with the preparation of single-labeled bioconjugates of the antibiotic Gentamicin (GT) with the sulforhodamine-derived fluorescence dye Texas Red (R)-X (TR), its purification by high-performance liquid chromatography (HPLC) and its characterization by high-resolution mass spectrometry. Aminoglycosides such as GT are efficient antibiotics, but also problematic due to severe side effects such as nephro- and ototoxicity. Fluorescent labeled GT is used to visualize cellular uptake and distribution of the antibiotic to finally understand the mechanisms of serious adverse drug reactions. Pharmaceutically administered GT is a mixture of mainly four different components, which exhibit three (GT(C1)) or four (GT(C1a), GT(C2), GT(C2a)) primary amino functional groups which can be coupled with the labeling reagent TR. Thus, multiple labeling could be envisaged which was assumed to be detrimental for uptake studies by fluorescence imaging. The proposed synthesis aimed at preparation of single labeled product and together with the employed purification strategy indeed yielded single labeled GT as product. Analytical control of the reaction product was carried out by means of mass spectrometry (UHPLC-ESI-QTOF-MS/MS) to rule out over-labeling of GT, which would alter the physicochemical characteristics of GT and its cellular uptake significantly. Moreover, LC-MS/MS analysis gave valuable insights into structural diversity of single labeled products. Further, high-resolution MS and MS/MS spectra of underivatized GT are provided as well. The analytical information on preparation strategy and structure diversity is valuable for studies with a clinical focus on research of aminoglycoside toxicity. Furthermore, it is deemed to be useful for the development of LC-MS/MS assays for the determination of aminoglycosides or the fast screening of synthetic biology samples from biotechnological drug discovery. Isomer selectivity of one- and two-dimensional approaches of mixed-mode and hydrophilic interaction liquid chromatography coupled to tandem mass spectrometry for sugar phosphates of glycolysis and pentose phosphate pathways

Min Su, Kristian Serafimov, Peng Li, Cornelius Knappe, Michael Laemmerhofer

Summary: The chromatographic behavior of mixed-mode and hydrophilic interaction liquid chromatography (HILIC) was assessed in this study for the separation of metabolites from glycolysis and pentose phosphate pathways. Optimization of chromatographic conditions allowed for selective targeted analysis of glycolytic phosphorylated carbohydrate metabolites using liquid chromatography with tandem mass spectrometry (MS/MS). The separation of pentose phosphate isomers was achieved using mixed-mode HILIC/SAX and HILIC BEH Amide columns. HILIC-MS/MS showed challenges in separating hexose monophosphate metabolites, but the BEH Amide column provided the best results for this separation.

Polybutylene terephthalate-based stationary phase for ion-pair-free reversed-phase liquid chromatography of small interfering RNA. Part 1: Direct coupling with mass spectrometry

Feiyang Li, Shenkai Chen, Sylwia Studzinska, Michael Laemmerhofer

Summary: Ion-pairing reversed-phase liquid chromatography (IP-RPLC) is commonly used for the analysis of nucleic acid related compounds. However, the high concentration of ion-pairing reagent in IP-RPLC is not suitable for coupling with mass spectrometry (MS). This study explored the use of a PBT-bonded silica-based stationary phase for the separation of Patisiran without the need for ion-pairing reagents. The method allowed for MS and MS/MS characterization without contamination or related issues.

Cross-linked polysiloxane-coated stable bond O-9-(2,6-diisopropylphenylcarbamoyl)quinine and quinidine chiral stationary phases as well as application in enantioselective cryo-HPLC

Ulrich Woiwode, Adrian Sievers-Engler, Michael Laemmerhofer

Summary: In this work, new polymeric chiral stationary phases were prepared and compared with brush-type chiral stationary phases. The polymeric CSPs showed similar enantioselectivity but lower chromatographic efficiencies compared to the brush phases. Oxidation of the polymeric CSPs reduced retention times and allowed for lower counterion concentrations in liquid chromatography-electrospray ionization-mass spectrometry applications. In addition, cryogenic cooling of the column enabled baseline separation of estradiol.

Summary: In this study, seven commercially available UHPLC columns were systematically evaluated for their retention and isomer selectivity of branched chain fatty acids (BCFAs). It was found that IG-U column showed the best isomer selectivity for short and medium-chain BCFA isomers, while RP C18 column had good selectivity for long-chain BCFA isomers. Furthermore, an untargeted isomer-selective BCFA profiling method was developed using UHPLC-ESI-MS/MS without pre-column derivatization.
