An increasing corpus of research has shown that membrane shape, generated by the external environment or intrinsic mechanisms, is an important parameter in controlling cellular processes. Recent findings demonstrate how membrane curvature affects protein function on nanometer to micron length-scales. Experimental modulation of membrane curvature reveals its role as a signal that controls protein affinity and enzymatic activity, ensuring the progression of key cellular processes.
An increasing corpus of research has demonstrated that membrane shape, gen-erated either by the external environment of the cell or by intrinsic mechanisms such as cytokinesis and vesicle or organelle formation, is an important parameter in the control of diverse cellular processes. In this review we discuss recent find-ings that demonstrate how membrane curvature (from nanometer to micron length-scales) alters protein function. We describe an expanding toolkit for experi-mentally modulating membrane curvature to reveal effects on protein function, and discuss how membrane curvature - far from being a passive consequence of the physical environment and the internal protein activity of a cell - is an important sig-nal that controls protein affinity and enzymatic activity to ensure robust forward pro-gression of key processes within the cell.

Summary: Fusion-associated small transmembrane (FAST) proteins are a diverse family of nonstructural viral proteins that drive cell-cell fusion, increasing viral spread and pathogenicity. Unlike viral fusogens with tall ectodomains, FAST proteins have short fusogenic ectodomains that cannot bridge the intermembrane gap between neighboring cells. This work suggests that localized mechanical pressure on the plasma membrane coupled to a membrane-disruptive ectodomain is sufficient to drive cell-cell fusion.

A preferred sequence for organelle inheritance during polarized cell growth

Summary: Organelle inheritance in budding yeast follows a specific order, starting with the inheritance of cortical endoplasmic reticulum and peroxisomes, followed by vacuoles, mitochondria, and finally the nucleus and perinuclear endoplasmic reticulum. Even when the cell cycle is disrupted, organelle inheritance still occurs, indicating that organelle inheritance is independent of the completion of S-phase.

Branched actin networks are organized for asymmetric force production during clathrin-mediated endocytosis in mammalian cells

Summary: This study investigates the organization of actin networks at clathrin-mediated endocytosis (CME) sites and how actin assembly is regulated. The findings show that actin assembles preferentially at stalled CME sites, where it exerts an asymmetric force to pull vesicles into the cell. N-WASP and the Arp2/3 complex are recruited to one side of CME sites for stimulating asymmetric actin assembly.

Summary: During mitosis, microtubules make attachments to chromosomes via a protein complex called the kinetochore, facilitating accurate chromosome segregation. This study used TIRF microscopy to observe the translocation of kinetochores on the lateral surface of microtubules and found that it is dependent on the kinesin-8 protein Kip3. The findings provide insights into the mechanism of cell division.

Cik1 and Vik1 accessory proteins confer distinct functions to the kinesin-14 Kar3

Summary: The study shows that the binding partners of the yeast kinesin-14 Kar3, Cik1, and Vik1, regulate its localization and function in a cell cycle-dependent manner. Specifically, Kar3-Vik1 induces MT catastrophes in S phase and metaphase while limiting MT polymerization in G1 and anaphase. On the other hand, Kar3-Cik1 promotes catastrophes and pauses in G1 and increases catastrophes in metaphase and anaphase. Additionally, Cik1 is necessary for Kar3 to track MT plus-ends in S phase and metaphase.

Self-organizing actin networks drive sequential endocytic protein recruitment and vesicle release on synthetic lipid bilayers

Summary: This research demonstrates the sequence recruitment of core endocytic proteins and regulatory proteins, as well as the assembly of actin networks in live cells. The study shows that supported lipid bilayers coated with purified yeast WASP recruit downstream endocytic proteins and assemble actin networks. The actin-driven vesicle generation could be a precursor for various vesicle forming processes in different cellular environments.

Induced nanoscale membrane curvature bypasses the essential endocytic function of clathrin

Summary: Cail et al. demonstrate that induced nanoscale membrane curvature can recruit endocytic proteins and produce cargo-laden vesicles without the coat protein clathrin. However, the induced curvature does not bypass AP2 and FCHo1/2 functions. This work suggests that the essential function of clathrin is to physically stabilize the curvature of the evolving vesicle.
