Summary: An uncommon enzymatic-fingerprinting workflow was proposed for the characterization and discrimination of mushroom polysaccharides using HILIC-ESI-MS. The study revealed the structural features of polysaccharides in Hericium erinaceus fruiting bodies and visually classified different polysaccharides through similarity analysis and other methods. This enzymatic-fingerprinting workflow provides an effective example for quality evaluation of fungi polysaccharides using a structural-fingerprinting strategy.

Summary: The present study investigated the structural characteristics of an acid-extracted polysaccharide fraction from mountain tea. The fraction was found to be predominantly composed of uronic acids, with smaller amounts of galactose and glucose, and mainly consisted of highly methyl-esterified homogalacturonan. Enzymatic fingerprinting techniques revealed the presence of unsaturated oligomers, indicating large blocks of highly methyl-esterified GalA residues in the polysaccharide.

Summary: This study compared the differences in Lycium barbarum polysaccharides from different regions of China. The results showed significant variations in monosaccharide composition and content of the polysaccharides among different geographical origins. Using chemical fingerprinting and chemometrics analysis, it was possible to differentiate L. barbarum samples from different regions. These findings provide a reliable method for tracing the origin of L. barbarum samples and improving the quality control of therapeutic polysaccharides.

Summary: Advancements in metabolomics have allowed for the identification and characterization of saliva metabolites as biomarkers, but discrepancies in biomarker content for a given disease have been observed. Differences in saliva collection, storage, and treatment methods could contribute to these discrepancies, as there is currently no standardized method. This study investigated the effects of different collection methods on saliva metabolite analysis, and found that saliva obtained through spitting and aspiration yielded comparable results, while the Salivette (R) method did not appear suitable for untargeted analysis of the salivary metabolome.

Summary: Liver disease is a major cause of global mortality, and identifying biomarkers for diagnosing its progression is crucial for improving outcomes. Targeted mass spectrometry is a powerful tool for verifying biomarker candidates and clinical applications, particularly for glycoproteins translation. However, the limitation of analyzing only one sample per run has become apparent. In this study, a high-throughput intact N-glycopeptides quantification strategy was developed, allowing the validation of 20 samples per run with an average analysis time of 3 minutes per sample. The strategy was applied in a cohort of 461 serum samples and identified a panel of 10 IgG N-glycopeptides that have strong clinical utility in evaluating the severity of liver disease.
