An evaluation of the Luminex NxTAG Respiratory Pathogen Panel was performed on 404 clinical respiratory specimens. Clinical sensitivities and specificities of the assay compared to those of the reference methods were 80.0% to 100.0% and 98.9% to 100.0%, respectively. Correct genotyping information was provided for 95.5% of influenza virus A specimens. The closed-tube format of the assay simplified the workflow and minimized carryover contamination.

Chemerin contributes to inflammatory responses and suppresses osteogenic differentiation in chronic periodontitis

Xinling Wang, Yuejun Tang, Rui Xiao

Summary: This study found that levels of chemerin and inflammatory cytokines were higher in the gingival crevicular fluid and serum of chronic periodontitis (CP) patients, and were positively correlated with each other. Periodontal ligament stem cells (PDLSCs) isolated from CP patients had higher expressions of chemerin and these cytokines compared to healthy individuals. Furthermore, chemerin dose-dependently increased inflammatory responses and inhibited osteogenic differentiation of PDLSCs.

Response of Microbial Communities on Cathode with Different Potentials in a Single-Chamber Reactor

Liyun An, YingChun Yan, Yong Nie, Min Gou, YueQin Tang, Xiao-Lei Wu

Summary: Microbes can use both organic and inorganic compounds as electron donors to produce energy for growth. Previous studies have mainly focused on the effects of different electron donors on microbial community under a set cathode potential, while their response to different redox potentials in different environments remains under-researched. In this study, lake sediment was incubated in a reactor equipped with three working electrodes at different potentials. The results showed that the structure of bacterial communities was similar for closed-circuit electrodes, but significantly different for open-circuit electrodes. Specific bacteria were also preferentially enriched by different electrode potentials.

The responses of mesophilic and thermophilic anaerobic digestion of municipal sludge to periodic fluctuation disturbance of organic loading rate

Zong-Lin Wu, Quan Zhang, Zi-Yuan Xia, Min Gou, Zhao-Yong Sun, Yue-Qin Tang

Summary: This study investigated the effects of fluctuation disturbance of organic loading rate (OLR) on anaerobic digestion (AD) of municipal sludge. The performance of the AD reactors remained similar before and after disturbance, indicating their buffer capacity to OLR periodic fluctuation. Different microbial community at RNA level was observed in the two reactors, with greater changes in the thermophilic AD. The results showed that the thermophilic microbial community was more vulnerable, but the reactor performance could be maintained using the functional redundancy strategy under OLR fluctuation disturbance.

Thermodynamic restrictions determine ammonia tolerance of methanogenic pathways in Methanosarcina barkeri

Yue Yi, Jan Dolfing, Ge Jin, XiaoYu Fang, WenHao Han, LaiYan Liu, YueQin Tang, Lei Cheng

Summary: In this study, the influence of ammonia on different methanogenic pathways was evaluated by culturing Methanosarcina barkeri with acetate, H2/CO2, and methanol. Aceticlastic methanogenesis was more sensitive to increased ammonia concentrations than hydrogenotrophic and methylotrophic methanogenesis. The results reveal the varying ammonia tolerances of M. barkeri under different methanogenic pathways, providing insights into the role of the thermodynamic potential of methanogenesis in ammonia stress tolerance and shedding light on the mechanism of ammonia inhibition on anaerobic digestion.

Article Exosomal miR-27b-3p secreted by visceral adipocytes contributes to endothelial inflammation and atherogenesis

Yan Tang, Li-Jie Yang, Hao Liu, Yan-Jue Song, Qi-Qi Yang, Yang Liu, Shu-Wen Qian, Qi-Qun Tang

Summary: Obesity, especially increased visceral fat, is associated with metabolic challenges such as atherosclerosis, but the exact mechanism is not fully understood. This study aims to investigate the role of visceral-fat-derived exosomes in endothelial cells and atherosclerosis. The research shows that obesity alters the miRNA profile of adipose exosomes, and exosomal miR-27b-3p can enter endothelial cells and activate the NF-kB pathway by suppressing PPARa. In ApoE-deficient mice, administration of miR-27b-3p mimic increases inflammation and atherogenesis, while overexpression of PPARa protects against atherosclerosis. Therefore, obesity-induced exosomal miR-27b-3p promotes endothelial inflammation and facilitates atherogenesis through PPARa suppression.

Proteiniphilum and Methanothrix harundinacea became dominant acetate utilizers in a methanogenic reactor operated under strong ammonia stress

Gao Feng, Yan Zeng, Hui-Zhong Wang, Ya-Ting Chen, Yue-Qin Tang

Summary: Microorganisms in anaerobic digestion are affected by ammonia, especially methanogens. This study investigated the effect of ammonia on microbial community and metabolism in a mesophilic chemostat. Results showed that even at high ammonia concentrations, methanogenesis and acetate degradation remained normal. The dominant methanogen changed with increased ammonia, and Proteiniphilum showed a positive correlation with ammonia concentration. Proteiniphilum and M. harundinacea had strong ammonia tolerance and played critical roles in acetate degradation. These findings contribute to the regulation and management of anaerobic digestion processes.

Valorizing kitchen waste to produce value-added fertilizer by thermophilic semi-continuous composting followed by static stacking: Performance and bacterial community succession analysis

Xiang-Yu Peng, Shi-Peng Wang, Xiu-Lin Chu, Zhao-Yong Sun, Zi-Yuan Xia, Cai-Yun Xie, Min Gou, Yue-Qin Tang

Summary: This study investigated the performance and bacterial community succession of a thermophilic semi-continuous composting (TSC) followed by static stacking (SS) for decentralized kitchen waste (KW) treatment. The TSC using an integrated automatic reactor achieved a stable performance with an organic matter degradation efficiency of 80.88% and a seed germination index (GI) of 78.51% by a daily feeding ratio of 10%. The subsequent SS for seven days further improved the compost quality with a GI of 91.58% by promoting the rapid proliferation of Proteobacteria and Bacteroidetes. The results offer new perspectives for practical applications of decentralized KW treatment using TSC.
