Development and validation of an enantioselective SFC-MS/MS method for simultaneous separation and quantification of oxcarbazepine and its chiral metabolites in beagle dog plasma

A rapid and sensitive assay based on supercritical fluid chromatography-tandem mass spectrometry (SFC-MS/MS) has been developed and validated for the determination of oxcarbazepine (OXC) and its chiral metabolite licarbazine (Lic) in beagle dog plasma using carbamazepine as internal standard. Chiral analysis in a run time of only 3 min was performed on an ACQUITY UPC2 (TM) Trefoil (TM) CEL2 column (3.0 x 150 mm, 2.5 mu m) at 50 degrees C by isocratic elution with a mobile phase of supercritical carbon dioxide (purity >= 99.99%) and methanol (60:40, v/v) at a flow rate of 2.3 mL/min. The assay was linear over the concentration ranges 5-1000 ng/mL for OXC and 0.5-100 ng/mL for the enantiomers of Lic with corresponding lower limits of quantitation of 5 ng/mL and 0.5 ng/mL. Intra- and inter-day precisions were in the range 0.78-14.14% with accuracies in the range -10.80% to 0.42%. The method was successfully applied to a pharmacokinetic study involving a single oral administration of 16 mg/kg OXC as Trileptal(@) tablets to beagle dogs. (C) 2016 Published by Elsevier B.V.

Cationic amphiphilic dendrons with effective antibacterial performance

Summary: Bacterial infections and antibiotic resistance have become a global healthcare crisis. Cationic amphiphilic dendrons designed and synthesized showed excellent antimicrobial activities, depending highly on the length of the hydrophobic alkyl chain. The identified prime candidate exhibited rapid and broad-spectrum bactericidal activity, disrupting the integrity of a pathogen's membrane and promoting wound healing.

Cationic Amphiphilic Dendrons with Anticancer Activity

Summary: This study reports on the potential of cationic amphiphilic dendrons with different hydrophobic alkyl chains and generations for anticancer applications. A specific dendron, C12-G1, showed broad-spectrum antitumor activity with negligible hemolysis. Mechanistic studies revealed that C12-G1 induced cancer cell necrosis through membrane disruption and inhibited tumor growth in a melanoma model.

Toxic aldehyde-scavenging polypeptides mitigate secondary injury after spinal cord injury

Summary: Neuroprotective therapy for traumatic spinal cord injury (SCI) still faces challenges, and the toxic reactive aldehydes produced immediately after SCI promote oxidative stress and inflammation, worsening the secondary injury. To address this, a drug-free polypeptide (PPAH) was designed and synthesized to scavenge toxic aldehydes and mitigate secondary SCI. In vitro and in vivo experiments demonstrated PPAH's ability to scavenge reactive aldehydes, protect against damage in neurocytes, and improve functional recovery in a rat SCI model. PPAH shows promise as an effective therapy for SCI.

Carrier-Free Nanodrug Based on Co-Assembly of Methylprednisolone Dimer and Rutin for Combined Treatment of Spinal Cord Injury

Summary: Spinal cord injury (SCI) is a severe condition that hinders neurological functional recovery and leads to permanent disabilities. Methylprednisolone (MP) is commonly used for SCI treatment, but it has potential side effects. In this study, we developed carrier-free nanoparticles (MP2-TK@RU NPs) that combine MP dimers and rutin to improve SCI treatment. The MP2-TK@RU NPs demonstrated anti-inflammatory and antioxidative properties, promoting locomotor function recovery and neuroprotection in rats with SCI. This carrier-free nanodrug shows promise as a therapeutic strategy for SCI treatment.
