The goal of this work is a systematic optimization of hydrophilic interaction liquid chromatography (HILIC) separation of acidic lipid classes (namely phosphatidic acids-PA, lysophosphatidic acids-LPA, phosphatidylserines-PS and lysophosphatidylserines-LPS) and other lipid classes under mass spectrometry (MS) compatible conditions. The main parameters included in this optimization are the type of stationary phases used in HILIC, pH of the mobile phase, the type and concentration of mobile phase additives. Nine HILIC columns with different chemistries (unmodified silica, modified silica using diol, 2-picolylamine, diethylamine and 1-aminoanthracene and hydride silica) are compared with the emphasis on peak shapes of acidic lipid classes. The optimization of pH is correlated with the theoretical calculation of acidobasic equilibria of studied lipid classes. The final method using the hydride column, pH 4 adjusted by formic acid and the gradient of acetonitrile and 40 mmol/L of aqueous ammonium formate provides good peak shapes for all analyzed lipid classes including acidic lipids. This method is applied for the identification of lipids in real samples of porcine brain and kidney extracts. (C) 2016 Elsevier B.V. All rights reserved.

Summary: A study analyzed data from the Czech Registry of Monoclonal Gammopathies and found that among 83 triple-class exposed relapsed/refractory multiple myeloma patients, the median overall survival was 14.2 months, progression-free survival was 6.2 months, and time to next treatment was 7.2 months. These results indicate a high disease burden in this patient population, consistent with previous studies.

Summary: We analyzed the vaccination rates of patients with multiple myeloma in the INSIGHT MM study and found that influenza and pneumococcal vaccination had an impact on overall survival. Additionally, vaccinated patients had fewer deaths due to infections compared to non-vaccinated patients. Recording vaccination status is important in prospective clinical trials as it may affect survival.

Summary: This retrospective observational study in Europe examined the demographics, clinical characteristics, treatment patterns, and outcomes of 4480 systemic light chain (AL) amyloidosis patients who initiated first-line treatment from 2004-2018. The study found that bortezomib-based regimens were the most commonly used first-line therapy, and only a minority of patients received autologous stem cell transplant. The study highlighted the high early mortality rate and the lack of improvement in survival outcomes for the frail stage IIIb population despite the introduction of new therapies.

Novel Local Off-the-Shelf Immunotherapy for the Treatment of Myeloma Bone Disease

Summary: We propose a novel approach for the local treatment of multiple myeloma bone disease (MBD) using a combination of natural killer cells (NKs) and mesenchymal stem cells (MSCs) within a fibrin scaffold, known as FINM. In vitro studies demonstrated that NKs within FINM exerted a robust anti-tumor activity against MM cells and suppressed osteoclast activity in a 3D model of MBD. The combination of FINM with other therapeutic agents showed a clear therapeutic synergistic effect.

Direct Chiral Supercritical Fluid Chromatography-Mass Spectrometry Analysis of Monoacylglycerol and Diacylglycerol Isomers for the Study of Lipase-Catalyzed Hydrolysis of Triacylglycerols

Summary: A fast and selective method using chiral supercritical fluid chromatography-mass spectrometry (SFC-MS) was developed to study lipase selectivity in the hydrolysis of triacylglycerols (TGs). The method involved the synthesis of enantiomerically pure monoacylglycerol (MG) and diacylglycerol (DG) isomers and their separation using a chiral column and neat methanol as a mobile phase modifier. The results revealed different stereoselectivities for substrates in the lipase-catalyzed hydrolysis.

Summary: Longer-term outcomes of the combination treatment of isatuximab and carfilzomib-dexamethasone (Isa-Kd) were evaluated in a Phase 3 trial. The addition of Isa to Kd significantly prolonged progression-free survival and improved complete response rates and minimal residual disease negativity. The safety profile of Isa-Kd was similar to previous analyses. These findings support Isa-Kd as a standard-of-care treatment for relapsed multiple myeloma patients.

Summary: This study focuses on the sequential separation of trivalent lanthanides using modified C-18 silica-packed supports and reversed-phase HPLC technique. By tuning the column modifiers and separation parameters, the researchers achieved systematic separation of Ln(3+). The results showed that the choice of column modifiers and fine-tuning of the modification procedure were crucial for achieving excellent separation.

Breakthrough curve analysis of phosphorylated hazelnut shell waste in column operation for continuous harvesting of lithium from water

Summary: Biosorption using phosphorylated hazelnut shell waste (FHS) is a promising method for lithium removal and recovery. This study investigated the performance of a continuous-flow packed-bed column system using FHS for ultra-low level lithium removal. The results showed that the lihium biosorption capacity of the FHS column was unaffected by the bed height, but decreased with increasing flow rate.

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

An integrated approach for studying the metabolic profiling of herbal medicine in mice using high-resolution mass spectrometry and metabolomics data processing tools

Summary: Analysis of in vivo exposure to traditional Chinese medicine using mass spectrometry is helpful for identifying effective ingredients and developing new drugs. This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles

Summary: A quantitative analytical method was developed and validated in this study to determine the concentrations of 81 aroma-relevant wine volatiles. The method utilizes stir bar sorptive extraction and multidimensional gas chromatography with flame ionization detector or mass spectrometer. The method demonstrated good linearity, reproducibility, and recovery, and was successfully applied to analyze 20 wine samples.
