There is currently accumulating evidence that endogenous estrogens play a critical role in the development of breast cancer. Estrogens and their metabolites have been studied in both pre-and postmenopausal women with more consistent results shown in the latter population, in part because of large hormonal variations during the menstrual cycle and far fewer studies having been performed in premenopausal women. In this review we describe in detail estrogen metabolism and associated genetic variations, and provide a critical review of the current literature regarding the role of estrogens and their metabolites in breast cancer risk. Leukocyte telomere length, cancer incidence and all-cause mortality among Chinese adults: Singapore Chinese Health Study

Hamed Samavat, Hung N. Luu, Kenneth B. Beckman, Aizhen Jin, Renwei Wang, Woon-Puay Koh, Jian-Min Yuan

Summary: The study revealed that longer telomeres are associated with an increased risk of cancer development, especially for common cancer types like breast, rectal, prostate, pancreatic cancer, and lung adenocarcinoma. Conversely, longer telomeres are linked to a decreased risk of noncancer related death.

Adverse perioperative outcomes among patients undergoing gastrointestinal cancer surgery: Quantifying attributable risk from malnutrition

Erin Kenny, Hamed Samavat, Riva Touger-Decker, J. Scott Parrott, Laura Byham-Gray, David Allen August

Summary: A study revealed a significant association between more than a 10% weight loss prior to gastrointestinal cancer surgery and adverse perioperative outcomes. The contribution of nutrition status markers to surgical outcomes was found to be small, which is a novel finding.

Summary: The study investigated the influence of COMT and UGT1A4 genotypes on changes in liver injury biomarkers in response to long-term, high-dose green tea extract supplementation among postmenopausal women. The results suggest that the UGT1A4 genotype may be an important risk factor for clinically-relevant liver damage in postmenopausal women after high-dose green tea extract supplementation.

Epigenetic inhibition of CTCF by HN1 promotes dedifferentiation and stemness of anaplastic thyroid cancer

Zongfu Pan, Xixuan Lu, Tong Xu, Jinming Chen, Lisha Bao, Ying Li, Yingying Gong, Yulu Che, Xiaozhou Zou, Zhuo Tan, Ping Huang, Minghua Ge

Summary: This study uncovered the emerging role of HN1 in promoting dedifferentiation of anaplastic thyroid cancer (ATC) cells. HN1 negatively regulated the thyroid differentiation markers and had an inhibitory effect on the transcriptional activation of CTCF, thereby influencing the chromatin accessibility of thyroid differentiation genes.

Autophagy machinery in glioblastoma: The prospect of cell death crosstalk and drug resistance with bioinformatics analysis

Yi Qin, Shengjun Xiong, Jun Ren, Gautam Sethi

Summary: Autophagy plays an important regulatory role in glioblastoma, and its dysregulation can lead to drug resistance and radioresistance. It also affects stem cell characteristics, overall growth, and metastasis. Therefore, autophagy is a promising target for glioblastoma therapy.

Elevated 2-oxoglutarate antagonizes DNA damage responses in cholangiocarcinoma chemotherapy through regulating aspartate beta-hydroxylase

Katsuya Nagaoka, Xuewei Bai, Dan Liu, Kevin Cao, Joud Mulla, Chengcheng Ji, Hongze Chen, Muhammad Azhar Nisar, Amalia Bay, William Mueller, Grace Hildebrand, Jin-Song Gao, Shaolei Lu, Hiroko Setoyama, Yasuhito Tanaka, Jack R. Wands, Chiung-Kuei Huang

Summary: This study found that serum 2-OG levels in cholangiocarcinoma patients are associated with the effectiveness of chemotherapy. Patients with progressive disease showed significantly higher levels of serum 2-OG compared to stable disease and partial response patients. The study also revealed that overexpression of ASPH mimics the effects of 2-OG, and knockdown of ASPH improves chemotherapy. Targeting ASPH enhances the effects of chemotherapy by modulating ATM and ATR, two key regulators of DDRs.
