Generation of Premature Termination Codon (PTC)-Harboring Pseudorabies Virus (PRV) via Genetic Code Expansion Technology

This study attempted to develop a live but replication-incompetent Pseudorabies virus (PRV) using genetic code expansion (GCE) technology. Although the premature termination codon (PTC) harboring PRV was successfully rescued, the incorporation efficacy of unnatural amino acids (UAA) was found to be extremely low. Moreover, the establishment of a stable transgenic cell line for PTC virus replication failed, with orthogonal tRNA(Pyl) identified as a limiting factor.
Despite many efforts and diverse approaches, developing an effective herpesvirus vaccine remains a great challenge. Traditional inactivated and live-attenuated vaccines always raise efficacy or safety concerns. This study used Pseudorabies virus (PRV), a swine herpes virus, as a model. We attempted to develop a live but replication-incompetent PRV by genetic code expansion (GCE) technology. Premature termination codon (PTC) harboring PRV was successfully rescued in the presence of orthogonal system MbpylRS/tRNA(Pyl) pair and unnatural amino acids (UAA). However, UAA incorporating efficacy seemed extremely low in our engineered PRV PTC virus. Furthermore, we failed to establish a stable transgenic cell line containing orthogonal translation machinery for PTC virus replication, and we demonstrated that orthogonal tRNA(Pyl) is a key limiting factor. This study is the first to demonstrate that orthogonal translation system-mediated amber codon suppression strategy could precisely control PRV-PTC engineered virus replication. To our knowledge, this is the first reported PTC herpesvirus generated by GCE technology. Our work provides a proof-of-concept for generating UAAs-controlled PRV-PTC virus, which can be used as a safe and effective vaccine.

Effects of short-video use on undergraduates' weight-loss intention: a regulatory mediation model

Summary: This research explores the effects of short video use on college students' weight loss intentions and reveals that short video use can significantly and positively predict weight loss intention. It also finds that body image mediates the relationship between short video usage and weight loss intentions, while self-esteem plays a regulatory effect as the direct path between short video use and weight loss intention as well as having a partial mediation effect of body image. The study highlights the importance of considering self-esteem as another key factor that affects college students' weight loss intentions in addition to short video use and body image.

SNP-based high-density linkage map construction and QTL mapping of black spot disease resistance in Chinese sand pear

Summary: In this study, a high-density genetic linkage map for pear black spot (PBS) resistance was constructed using SLAF-seq technology, and quantitative trait loci (QTL) associated with PBS resistance were identified. Two closely associated markers with PBS resistance were also identified. The results provide valuable information for improving PBS resistance in pear.

Multifunctional NiCo@RGO/SWNTs foam with oriented pore structure for excellent electromagnetic interference shielding

Summary: This study focuses on constructing a lightweight and multifunctional foam material with an oriented pore structure for advanced electromagnetic interference (EMI) shielding. By assembling a carbon-based conductive skeleton consisting of single-walled carbon nanotubes (SWNTs) and reduced graphene oxide (RGO), the material facilitates multilevel reflection of electromagnetic waves in confined space. Furthermore, the introduction of magnetic NiCo nanoparticles enhances the shielding performance by increasing the magnetic loss. The as-prepared foam exhibits exceptional EMI shielding effectiveness (SE) of 105 dB at the X band with an ultralow density of 0.0135 g cm-3. Additionally, the foam also possesses excellent heat insulation and flame-retardant properties. Overall, this lightweight multifunctional foam with an oriented pore structure shows great promise for applications in advanced EMI shielding and other specific areas.

Tension-compression asymmetry and Bauschinger-like effect of AZ31 magnesium alloy bars processed by ambient extrusion

Summary: Ambient extrusion was used to introduce different pre-strains in billets of AZ31 magnesium alloy. The results showed that the pre-strain had a significant effect on tension-compression asymmetry and Bauschinger-like effect, with the strongest effect observed at a pre-strain of approximately 11%.

The materials exhibited a transition from microsphere structure to supramolecular polymer gel, with fluorescence emission enhancement in the gel state. The supramolecular polymer gel material showed sensitive stimulus-response behavior and could be applied to detect food spoilage.

Summary: The production of inclusive, prompt, and non-prompt J/psi was studied for the first time in p-Pb collisions at midrapidity with the ALICE detector at the LHC. The study reconstructed J/psi mesons in the dielectron decay channel and separated prompt and non-prompt contributions. The results showed consistent nuclear modification factors for inclusive, prompt, and non-prompt J/psi.

The use of newly isolated fungal cultures for the selective delignification of bamboo culms

Summary: The screening of fungal species that produce ligninolytic enzymes led to the discovery of Paracremonium sp. LCB1, Clonostachys compactiuscula LCD1 and C. compactiuscula LCN1. These strains exhibited high levels of hemicellulase and ligninolytic enzyme production and low levels of cellulase activity. Co-culturing LCB1 and LCN1 enhanced lignin degradation and this combined fungal approach was found to be effective in bamboo delignification.

Characterization of crack-free AA-7075 fabricated by laser powder bed fusion with addition of Zr50.7Cu28Al12.3Ni9 metallic glass

Summary: The inclusion of 2 wt% Zr50.7Cu28Al12.3Ni9 metallic glass powders successfully eliminated hot cracks in AA-7075 specimens fabricated by laser powder bed fusion. Microstructure analysis revealed significant grain refinement in the AA-7075/MG specimen due to the formation of Al3Zr precipitates. The average grain size reduced from 9.26 μm to 1.6 μm, and the previously coarser columnar grains transformed into finer equiaxed grains with random orientation.

Summary: This paper compares and analyzes the high reliability solutions defined by 3GPP, and discusses the implementation of a high-reliability solution by 5G System (5GS) and the external network. By encouraging 5G service providers to consider high reliability solutions, it can promote more mature and perfect service R & D.
