Summary: The objective of this study was to optimize a thermosensitive in situ gelling formulation to improve the intranasal and nose-to-brain delivery of the antiepileptic drug carbamazepine (CBZ). The formulation consisted of poloxamer 407 (P407), poloxamer 188 (P188), and hydroxypropylmethylcellulose (HPMC) and exhibited suitable properties for nasal delivery. The mucoadhesive properties of the formulation were enhanced by adding different grades of HPMC, and the formulation showed promising mucoadhesive potential. Furthermore, the formulation was able to increase drug solubility, control drug release, and promote drug permeation across barrier membranes.

Summary: Given the threat of extinction to coral reefs, reproductive biotechnologies can help fight against extinction. This study aimed to develop a reliable method to assess oocyte viability for scleractinian corals. The trypan blue (TB) test was found to be unfeasible for assessing oocyte viability due to the high lipid content, while the thiazolyl blue tetrazolium bromide (MTT) assay provided a new technique for assessing coral oocyte viability and mitochondrial activity, which can contribute to reef monitoring and conservation strategies.

Summary: Thiomers, or thiolated polymers, have emerged as avant-garde approaches in anticancer therapy due to their reactivity and properties closely related to their applications. Research has shown that thiol-rich nanoformulations can address major drawbacks in administering various active pharmaceutical ingredients, especially in cancer therapy. This review focuses on synthetic tools available for thiolating polymers, particularly chitosan and hyaluronic acid, and outlines NP formation procedures, highlighting their use in cancer therapy.

Summary: Nanotechnology has found unique applications in drug delivery, cosmetics, fabrics, water treatment, and food packaging. Chitosan, a nanoparticle derived from natural polysaccharide chitin, has gained attention in modern medicine for its unique properties. This review examines the cytotoxicity of chitosan nanoparticles in biomedical research, revealing low cytotoxicity regardless of particle composition or assay method. However, thorough characterization and cytotoxicity assessment are recommended for new chitosan derivatives and compositions before market implementation.

Summary: The correlation between in vivo and in vitro data needs further optimization in order to reduce and replace animal testing in pharmaceutical development. This study investigates the mechanisms and kinetics of drug release from nanocarriers by using experimental methods and mathematical modeling. The findings serve as a significant milestone for the development of reliable in vitro/in silico models to predict drug bioavailability when enabling formulations are used.

Summary: This study extracted and characterized the β-glucans from the edible mushroom Pleurotus eryngii, and found that they contain structurally different polysaccharides. However, these polysaccharides were unable to induce the production of proinflammatory cytokines in LPS-differentiated THP-1 cells, despite being able to activate the human dectin-1a receptor.

Summary: This study aimed to investigate the loading of sildenafil citrate in three commercial transdermal vehicles using 3D full-thickness skin equivalent and compare the results with permeability experiments using porcine skin. The results showed that the results obtained using the 3D skin equivalent were comparable to those obtained using porcine skin, suggesting that the 3D skin model can be a valid alternative for ex-vivo skin absorption experiments.
