Peptides with 6-Aminohexanoic Acid: Synthesis and Evaluation as Plasmin Inhibitors

Fifteen new peptide derivatives of E >-aminocaproic acid (EACA) containing the known fragment -Ala-Phe-Lys- with an affinity for plasmin were synthesised in the present study. The synthesis was carried out a solid phase. The following compounds were synthesised: H-Phe-Lys-EACA-X, H-d-Ala-Phe-Lys-EACA-X, H-Ala-Phe-Lys-EACA-X, H-d-Ala-Phe-EACA-X and H-Ala-Phe-EACA-X, where X = OH, NH2 and NH-(CH2)(5)-NH2. All peptides, except for those containing the sequence H-Ala-Phe-EACA-X, displayed higher inhibitory activity against plasmin than EACA. The most active and selective inhibitor of plasmin was the compound H-d-Ala-Phe-Lys-EACA-NH2 which inhibited the amidolytic activity of plasmin (IC50 = 0.02 mM), with the antifibrinolytic activity weaker than EACA. The resulting peptides did not affect the viability of fibroblast cells, colon cancer cell line DLD-1, breast MCF-7 and MDA-MB-231 cell lines.

Jasmonate Compounds and Their Derivatives in the Regulation of the Neoplastic Processes

Summary: Cancer is a major concern in modern medicine due to the limited efficacy of current therapies. Natural compounds, such as jasmonic acid and its derivatives, are being explored for their potential in cancer prevention and treatment. The anti-tumor activity of jasmonates is attributed to mechanisms such as altering cellular ATP levels, inducing re-differentiation through MAPKs, and promoting apoptosis through reactive oxygen species. Jasmonates can also show synergistic effects when used in combination with other cancer drugs like cisplatin and paclitaxel.

Nitric-Acid Oxidized Single-Walled Carbon Nanohorns as a Potential Material for Bio-Applications-Toxicity and Hemocompatibility Studies

Summary: The study presents the cytotoxicity of single-walled carbon nanohorn (SWCNH) oxidized materials on A549 and HDF cell lines, as well as the hemocompatibility on human and porcine blood samples. Results show correlation between specific surface area of materials and platelet aggregation, and the I-D/I-G ratio from Raman spectra with hemoglobin release from erythrocytes.

Adrenaline May Contribute to Prothrombotic Condition via Augmentation of Platelet Procoagulant Response, Enhancement of Fibrin Formation, and Attenuation of Fibrinolysis

Summary: The study shows that adrenaline can enhance platelet activation and thrombus formation in the presence of subthreshold collagen, leading to increased platelet aggregation and formation of denser platelet-fibrin clots. These effects are associated with a decrease in fibrinolysis rate, which can be reduced by commonly used antiplatelet drugs.

Therapeutic Potential of Jasmonic Acid and Its Derivatives

Summary: Jasmonates, such as jasmonic acid and methyl jasmonate, are natural compounds with broad spectrum of activities, including anti-cancer, anti-inflammatory, and cosmetic effects. Studies on their biological activity in mammals have shown potential for inflammation treatment due to their structural similarities to prostaglandins. Furthermore, jasmonates are being considered as potential anti-cancer drugs when used alone or in combination with other chemotherapy agents, and may also have applications in cosmetics and dermatology.

NSAIDs Induce Proline Dehydrogenase/Proline Oxidase-Dependent and Independent Apoptosis in MCF7 Breast Cancer Cells

Summary: This study found that non-steroidal anti-inflammatory drugs (NSAIDs) induce apoptosis in breast cancer cells through the activation of peroxisome proliferator-activated receptor-gamma (PPAR gamma) and the generation of reactive oxygen species (ROS) via proline dehydrogenase/proline oxidase (PRODH/POX). The effect of NSAIDs on cell apoptosis is independent of cyclooxygenase-2 (COX-2) inhibition. Additionally, the study showed that the role of PRODH/POX in NSAID-induced apoptosis is more pronounced in breast cancer cells with PRODH/POX knockout. The findings suggest a potential therapeutic strategy for cancer treatment using NSAIDs.

Adenylate kinase immobilized on graphene oxide impairs progression of human lung carcinoma epithelial cells through adenosinergic pathway

Summary: Purinergic signaling has emerged as a crucial target for anti-cancer therapies, and the nanobiocatalytic system of AK-GO has been developed to modify the purinergic signaling components in lung carcinoma cells. The immobilized adenylate kinase effectively controls nucleotide balance and decreases adenosine concentration, leading to reduced proliferation and migration ability of cancer cells. The developed AK-GO nanobiocatalytic system shows good compatibility for biomedical applications.

Nitrogen plasma modification boosts up the hemocompatibility of new PVDF-carbon nanohorns composite materials with potential cardiological and circulatory system implants application

Summary: This study aims to design new materials with high blood compatibility and conducts hemocompatibility studies using poly(vinylidene fluoride) (PVDF) composites. Plasma-surface modifications are used to enhance hemocompatibility, and the optimal samples are selected through an optimization procedure. The results show that nitrogen plasma treatment is the best method to improve blood compatibility, with low platelet adhesion, no cytotoxicity, and excellent performance in thromboelastometry and hemolysis tests. The new surfaces have potential applications in cardiological and circulatory system implants and other blood-related biomaterials.
