Summary: Diabetic cardiomyopathy is a major cause of death worldwide, with oxidative stress playing a key role in its pathophysiology. NRF2 is a transcription factor that regulates antioxidant gene expression and detoxifying enzymes, helping to maintain cardiac homeostasis and counteract oxidative stress. Targeting the NRF2 signaling pathway may be a potential strategy for preventing and treating DCM and other cardiac diseases.

Summary: Pre-treatment with curcumin significantly improves the survival of CECs under oxidative stress by reducing ROS production and increasing nuclear Nrf2 expression, as well as enhancing the production of antioxidant enzymes. The activation of the Keap1/Nrf2/ARE pathway by curcumin is crucial for CECs to improve their anti-oxidative capacity.

Summary: The accumulation of senescent cells in aging tissues contributes to age-associated diseases. Inhibiting the anti-apoptotic machinery of senescent cells through BCL-2 protein family inhibitors can eliminate these cells. Combination therapy with BCL-2 inhibitors shows synergistic effects in eliminating senescent cells and overcoming resistance to monotherapy.

Summary: Keap1-Nrf2 is a crucial signaling cascade for promoting or preventing carcinogenesis. Nutraceuticals, as dietary agents with health benefits, show great potential as chemoprevention agents for cancer. This review systematically analyzes the role of 12 different nutraceuticals in modulating the Nrf2/Keap1 signaling mechanism. Although there is considerable preclinical evidence, further high-quality research is needed to validate the potential role of these nutraceuticals in Keap1-Nrf2 modulation.

Summary: Arterial hypertension is a prevalent disease characterized by increased incidence and heterogeneous pathophysiology. Oxidative stress plays an important role in its development, with excessive levels causing vascular cell damage and promoting hypertension. Transcription factors such as Nrf2 mediate antioxidant response pathways and provide protection.

Summary: In this study, the cytotoxicity and mechanisms of action of PM2.5 and TSP on human alveolar epithelial A549 cells were investigated, revealing that TSP was more toxic than PM2.5. PM2.5 induced oxidative stress while low-concentration TSP increased hemeoxygenase-1 levels. Both PM2.5 and TSP activated the protective Kelch-like ECH-associated protein 1/Nrf2 pathway in A549 cells, suggesting a potential key role of Nrf2 in detoxifying environmental airborne particles.

Summary: The study demonstrates that curcumin can protect the skin from UV damage by regulating the Nrf2 pathway. UV irradiation leads to cell apoptosis and oxidative stress, which can be reversed by adding curcumin. Additionally, curcumin induces Nrf2 nuclear accumulation in Hacat cells to provide protection against UV exposure-induced photodamage.

Summary: The Nrf2/ARE pathway plays a critical role in protecting against cellular stress and inflammation in diseases like ulcerative colitis. Both Keap1-dependent and Keap1-independent cascades have been proposed to positively affect the activation of this pathway. Nutritional compounds have the potential to modulate the activation of Nrf2/ARE pathway and may be used for therapeutic applications in UC.

Summary: The study demonstrated that PBA protected cardiomyocytes against OGD/R and LAD-induced MI/RI by suppressing NLRP3 inflammation activation and inducing the Nrf2 signaling pathway. PBA targeted Keap1 by selectively covalently binding to conserved cysteine residues, inhibiting Nrf2 ubiquitination and activating antioxidant enzymes.
