Summary: The secreted phospholipase A2 (PLA2G10) plays an important role in mouse fertilization by enhancing acrosome reaction and secretion, improving fertilization rate, and binding to PLA2R1 on gametes. It is shown that the catalytic activity and PLA2R1 binding of PLA2G10 both contribute to its profertility effect, and the study provides new insights into the complexity of PLA2G10's action in optimizing fertilization.

Summary: Fertilization is a complex process that involves multiple steps. The IZUMO1 protein has been shown to be essential for sperm-egg fusion in mice, but its role in other species is largely unknown. In this study, Izumo1 knockout rats were generated using CRISPR/Cas9 technology and found to be infertile. Unlike in mice, the knockout rat spermatozoa were unable to bind to the egg plasma membrane. Further investigation revealed a decrease in the number of acrosome-reacted sperm bound to the egg plasma membrane in the knockout mice. However, the binding of acrosome-reacted sperm to the egg plasma membrane was not affected in mice lacking other recently discovered fusion-indispensable genes. These findings suggest that IZUMO1 is required for sperm-oolemma binding prior to fusion, at least in rat.

Summary: Gamete fusion, mediated by specialized proteins called fusogens, is the climax of fertilization in sexually reproductive organisms. The identification of HAP2/GCS1 as the fusogen in flowering plants and protists opens the possibility for a similar fusogen in vertebrate fertilization. HAP2/GCS1 proteins share structural similarities with other exoplasmic fusogens, forming the Fusexin superfamily. However, HAP2/GCS1 or other fusexins have not been identified in vertebrates or fungi, leading to questions about their loss during evolution or the difficulty in their identification.

Summary: In this study, we found that porcine beta-defensin 129 (pBD129) is widely present in accessory sex glands and coats the surface of sperm. Inhibiting pBD129 on the sperm surface resulted in increased acrosome integrity, tyrosine phosphorylation levels, and Ca2+ influx, leading to a reduced in vitro fertilization cleavage rate. Furthermore, treatment with recombinant pBD129 restored sperm motility in semen contaminated with Escherichia coli.

Summary: The CatSper cation channel is essential for sperm capacitation and male fertility. The study identifies C2CD6 as a subunit of the CatSper complex and demonstrates its role in the formation of calcium signaling nanodomains on flagellar membranes. Deficiency of C2CD6 leads to male sterility due to depleted CatSper nanodomains and impaired sperm function. C2CD6 may facilitate the incorporation of CatSper complex into the flagellar plasma membrane and function as a calcium sensor.
