These exosomal miRNAs can be delivered to tumor cells via exosomes and directly target genes like FOXP4 and EZH2, reinforcing their tumor inhibitory effects. Overexpression of miR-101-3p and miR-423-5p has been shown to inhibit tumorigenesis in vivo in a xenograft mouse model, indicating a potential therapeutic approach for the treatment of children with medulloblastoma.

Summary: In this study, circROBO1 was found to be significantly upregulated in hepatocellular carcinoma and associated with poor prognosis. Functional experiments revealed that circROBO1 promotes HCC progression by regulating the miR-130a-5p/CCNT2 axis. Additionally, nanoparticles loaded with si-circROBO1 were developed and shown to inhibit HCC cell proliferation without apparent toxicity to major organs in vivo. These findings provide new insights into the biological functions of circROBO1 and suggest it as a potential therapeutic target for HCC treatment.

Summary: Schizophrenia is a severe neuropsychiatric disorder with unknown precise pathogenesis. This study shows that there is an increased level of Interferon beta (IFN-beta) in the serum of schizophrenia patients, which is positively correlated with HERV-W ENV. In addition, a decrease in serum long intergenic non-protein coding RNA 1930 (linc01930) is negatively correlated with HERV-W ENV and IFN-beta. HERV-W ENV promotes IFN-beta expression and neuronal apoptosis by suppressing linc01930, and the cGAS/STING pathway is involved. The monoclonal antibody GNbAC1 targeting HERV-W ENV may be a potential therapeutic choice for schizophrenia.

Summary: Schizophrenia affects 1% of the worldwide population and is characterized by dysregulation of homeostasis in the endoplasmic reticulum (ER). Recent studies suggest a connection between ER stress, unfolded protein response (UPR), and schizophrenia. This research investigates the molecular mechanism linking ER stress and endogenous retrovirus group W member 1 envelope (ERVW-1) in schizophrenia. The findings demonstrate that ERVW-1 induces ER stress by suppressing GANAB expression, leading to upregulation of ATF6 and XBP1 expression and contributing to the development of schizophrenia.

Summary: In this study, the researchers investigated the effects of ERVWE1 on 5-HT neuronal plasticity and found that ERVWE1 enhanced HTR1B expression, thereby activating the ERK-ELK1-Arc pathway and reducing the complexity and spine density of 5-HT neurons. Furthermore, ERVWE1 reduced m6A levels through ALKBH5 demethylation. ALKBH5 mediated the observed alterations in 5-HT neuronal plasticity induced by ERVWE1. These findings suggest that ERVWE1 may contribute to aberrant synaptic plasticity in schizophrenia.
