Summary: This study investigated the effects of the generalist parasitoid Exorista japonica on the biological traits of its lepidopteran host, Bombyx mori. It was found that parasitism by E. japonica shortened the molting duration and decreased the emergence rate of the host, while also affecting the hormone levels and gene expression related to molting and pupation. These findings highlight a novel target mechanism employed by tachinid parasitoids and provide new insights into host/parasitoid relationships.

Summary: 20E is involved in larval molting, metamorphosis, and innate immune response in insects. It regulates AMP genes through the IMD pathway in Drosophila, but its involvement in other immune pathways is unknown. This study shows that BmMD-2A is regulated by 20E and activates downstream immune effectors to combat bacterial infections during larval metamorphosis in Bombyx mori.

Summary: This study identified the ps homologue gene in B. mori and found that PS plays an essential role in maintaining the dynamic balance of apoptosis and proliferation during metamorphosis. Loss-of-function analysis of Bmps using the binary transgenic CRISPR/cas9 system revealed delayed developmental time and abnormal appendages in Bmps-depleted animals, along with affected apoptosis and proliferation related pathways and the Hippo pathway.

Summary: This study identified the upstream region of Bombyx mori ERR (BmERR) as having promoter activity and found that 20-hydroxyecdysone up-regulated its transcriptional activity. Several putative ecdysone response elements were predicted, and an EcRE-like sequence was identified. The study also revealed the interference of BmERR and BmEcR mRNA affected the expression of BmEcR and BmUSP in embryonic cells.

Summary: This study used a genome-wide CRISPR/Cas9-based library and large-scale screening to analyze the key genes involved in the silkworm 20E signaling pathway and their mechanisms of action. Functional annotation revealed that 20E regulates key proteins in processes mainly occurring in the cytoplasm and nucleus. Pathway enrichment analysis showed that 20E can activate phosphorylation, may affect innate immunity, interfere with intracellular nutrition and energy metabolism, and ultimately lead to cell apoptosis. These findings provide a panoramic overview of the silkworm's response to 20E signaling, highlighting the utility of genome-wide CRISPR mutant libraries in deciphering hormone signaling pathways and metamorphosis regulation in insects.

Summary: The matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) play crucial roles in innate immunity and metamorphosis in silkworms. High-activity MMPs and low-activity TIMP may enhance infection by B. mori nucleopolyhedrovirus, affecting silkworm development and causing tissue dysplasia. These findings provide insights into the conserved functions of MMPs and TIMP in human organogenesis and immunoregulation.

Summary: In this study, RNA-seq analysis of the silkworm midgut after BmCPV1 infection identified 2520 differentially expressed genes (DEGs). Gene Ontology (GO) functional annotation showed that the DEGs predominantly functioned in binding, cell, and cellular processes. Kyoto Encyclopedia of Genes and Genomes (KEGG) functional annotation revealed that the DEGs were mainly distributed in global and overview metabolism maps, translation, and signal transduction. Experimental validation confirmed that BmPGRP-S5 acts as a proviral host factor for BmCPV1 infection.

Summary: Diapause is a major developmental switch in insects evolved as a survival strategy under unfavorable environmental conditions. In the silkworm Bombyx mori, diapause is transgenerationally induced as a maternal effect, with progeny diapause determined by the mother's embryonic environmental temperature. The molecular mechanisms of diapause induction involve a hierarchical pathway regulated by gamma-aminobutyric acid (GABA)ergic and corazonin signaling systems.
