Postural freezing relates to startle potentiation in a human fear-conditioning paradigm

Freezing in response to impending threat is a core defensive mechanism in humans. Research has shown that human freezing is highly sensitive to fear conditioning and intensifies with the proximity of threat. This animal-like freezing response aids in active preparation for unexpected attacks and captures real-life expressions of anxiety.
Freezing to impending threat is a core defensive response. It has been studied primarily using fear conditioning in non-human animals, thwarting advances in translational human anxiety research that has used other indices, such as skin conductance responses. Here we examine postural freezing as a human conditioning index for translational anxiety research. We employed a mixed cued/contextual fear-conditioning paradigm where one context signals the occurrence of the US upon the presentation of the CS, and another context signals that the CS is not followed by the US. Critically, during the following generalization phase, the CS is presented in a third and novel context. We show that human freezing is highly sensitive to fear conditioning, generalizes to ambiguous contexts, and amplifies with threat imminence. Intriguingly, stronger parasympathetically driven freezing under threat, but not sympathetically mediated skin conductance, predicts subsequent startle magnitude. These results demonstrate that humans show fear-conditioned animal-like freezing responses, known to aid in active preparation for unexpected attack, and that freezing captures real-life anxiety expression. Conditioned freezing offers a promising new, non-invasive, and continuous, readout for human fear conditioning, paving the way for future translational studies into human fear and anxiety.

Acute-stress-induced change in salience network coupling prospectively predicts post-trauma symptom development

Summary: There are individual differences in how acute stress affects the brain's neurocognitive networks. Changes in the connectivity strength of these networks upon acute stress may predict the development of post-traumatic stress disorder (PTSD) symptoms. A study on police students found that weakened connectivity between the salience network (SN) and default mode network (DMN) was associated with increased perceived-stress level, while increased coupling between the overall SN and anterior cerebellum was related to higher levels of clinician-rated PTSD symptoms.

Oscillatory brain responses to emotional stimuli are effects related to events rather than states

Summary: Emotional cues can enhance attention and information processing. Electrophysiological brain research suggests that increased gamma band activity and decreased alpha band activity over posterior brain areas are associated with attention allocation. This study investigated whether the modulation of brain oscillations occurs in a stimulus-induced manner or leads to prolonged state-like changes. The results showed decreased alpha and increased gamma power in response to unpleasant pictures compared to pleasant pictures, supporting a stimulus-induced effect of alpha and gamma power. The findings also suggest event-related attention toward unpleasant pictures and are discussed in relation to previous EEG research and behavioral research on threat-induced freezing-like response.

Summary: Limited convergence exists in neuroimaging investigations of subcortical brain region volumes in social anxiety disorder (SAD). Variations in methodological approaches and sample selection based on age and clinical characteristics may contribute to inconsistent findings. A global mega-analysis initiated by the ENIGMA-Anxiety Working Group revealed subtle volumetric alterations in subcortical regions in SAD patients. Differences were found in putamen and pallidum volumes, with additional factors such as comorbid anxiety disorders and age of SAD onset influencing these volumetric differences.

Dropout from psychological treatment for borderline personality disorder: a multilevel survival meta-analysis

Summary: Dropout is a significant problem in psychotherapy for borderline personality disorder (BPD). This study found that dropout rates were highest in the first quarter of treatment. Schema therapy had the lowest overall dropout rate, while mentalization-based treatment had the lowest dropout rate in the first two quarters. Individual therapy had the lowest dropout rate, while group therapy had the highest rate, with combined formats in-between. Other variables such as age or substance-use exclusion criteria were not associated with dropout.

Trauma-induced human glucocorticoid receptor expression increases predict subsequent HPA-axis blunting in a prospective longitudinal design

Summary: This study provides the first longitudinal evidence of a dose-response relationship between trauma and human GR mRNA expression changes, indicating that increased traumatic events during police training lead to stronger GR mRNA expression. A neurocognitive theory of flexible emotion control: The role of the lateral frontal pole in emotion regulation

The Flexible Emotion Control Theory (FECT) integrates insights from decision and affective sciences to explain how individuals can adapt their emotion-regulatory behavior. FECT proposes that rapid switching between emotional control strategies requires evaluating both current and alternative options, and involves specific prefrontal structures such as the frontal pole.

Summary: Functional neurological disorder (FND) is often overlooked and misunderstood by clinicians, despite being a common and accurately diagnosable condition based on clinical signs. People with FND face discrimination from healthcare providers, researchers, and the public, reflecting the neglect of disorders primarily affecting women. We argue that FND is a feminist issue, drawing on historical and contemporary perspectives from clinical, research, and social domains. We call for equal attention and support for FND in medical education, research, and clinical services to ensure adequate care for those affected.

Reappraisal is less effective than distraction in downregulation of neural responses to physical threats-An event-related potential investigation

Summary: Evolutionary threats (ETs) elicit stronger fear responses and specific phobias compared to modern threats (MTs). The processing of ET relies on lower-order neural fear circuits, making it less susceptible to higher-order cognitive emotion regulation. In a study involving ERP, participants passively viewed high- and low-threat pictures and successfully downregulated responses using cognitive reappraisal or distraction strategies. ET triggered stronger attention processing compared to MT, and distraction was more effective in early and stronger downregulation of responses.

Developmental shift in testosterone influence on prefrontal emotion control

Summary: A paradoxical effect of testosterone is observed in social emotional behavior between adolescents and adults. While high testosterone levels are associated with increased emotion control in the prefrontal cortex during adolescence, this neuro-endocrine relationship is reversed in adulthood. This study investigated whether this functional transition also occurs in human adolescents and young adults. The findings suggest that testosterone-dependent maturation of the prefrontal-amygdala circuit supporting emotion control changes during the transition from middle adolescence into young adulthood, with a decrease in prefrontal cortex engagement and an increase in testosterone-modulated amygdala reactivity.

Anxious individuals shift emotion control from lateral frontal pole to dorsolateral prefrontal cortex

Summary: It is not well understood why anxious individuals fail to control emotional behaviour. This study shows that highly anxious individuals have an overexcitable lateral frontopolar cortex (FPl) and do not recruit this region during emotional action control. Their FPl is overexcitable and receives stronger amygdalofugal projections, but fails to be recruited during emotional action control, leading to a functional anatomical shift to other prefrontal areas.

No Evidence for a Causal Effect of Exogenous Testosterone on Risky Decision-Making in Women: An Experiment and Meta-Analysis
