Donor clonal hematopoiesis (CH) has a significant impact on clinical outcomes and graft alloimmune function in transplant recipients. DNMT3A-CH is associated with improved recipient survival and an augmented network of inflammatory cytokines in recipients. Different mutations may contribute to donor cell leukemia (DCL) in allogeneic hematopoietic cell transplantation.
PURPOSE Clonal hematopoiesis (CH) can be transmitted from a donor to a recipient during allogeneic hematopoietic cell transplantation. Exclusion of candidate donors with CH is controversial since its impact on recipient outcomes and graft alloimmune function is uncertain. PATIENTS AND METHODS We performed targeted error-corrected sequencing on samples from 1,727 donors age 40 years or older and assessed the effect of donor CH on recipient clinical outcomes. We measured long-term engraftment of 102 donor clones and cytokine levels in 256 recipients at 3 and 12 months after transplant. RESULTS CH was present in 22.5% of donors, with DNMT3A (14.6%) and TET2 (5.2%) mutations being most common; 85% of donor clones showed long-term engraftment in recipients after transplantation, including clones with a variant allele fraction < 0.01. DNMT3A-CH with a variant allele fraction 0.01, but not smaller clones, was associated with improved recipient overall (hazard ratio [HR], 0.79; P = .042) and progression-free survival (HR, 0.72; P= .003) after adjustment for significant clinical variables. In patients who received calcineurin-based graft-versus-host disease prophylaxis, donor DNMT3A-CH was associated with reduced relapse (subdistribution HR, 0.59; P = .014), increased chronic graft-versus-host disease (subdistribution HR, 1.36; P = .042), and higher interleukin-12p70 levels in recipients. No recipient of sole DNMT3A or TET2-CH developed donor cell leukemia (DCL). In seven of eight cases, DCL evolved from donor CH with rare TP53 or splicing factor mutations or from donors carrying germline DDX41 mutations. CONCLUSION Donor CH is closely associated with clinical outcomes in transplant recipients, with differential impact on graft alloimmune function and potential for leukemic transformation related to mutated gene and somatic clonal abundance. Donor DNMT3A-CH is associated with improved recipient survival because of reduced relapse risk and with an augmented network of inflammatory cytokines in recipients. Risk of DCL in allogeneic hematopoietic cell transplantation is driven by somatic myelodysplastic syndrome-associated mutations or germline predisposition in donors. Cesium manganese halide perovskite-analogue nanocrystals with highly efficient energy conversion for flexible multifunctional fibers

Summary: In recent years, all-inorganic perovskite materials have been widely applied in various fields. In this study, novel zero-dimensional lead-free Cs2Mn(Cl/Br)(4)center dot 2H(2)O perovskite-analogue nanocrystals (PA NCs) were synthesized under aerobic conditions. The introduction of lead into manganese halide perovskite-analogue nanocrystals resulted in enhanced fluorescence properties. The Pb2+-doped Cs2Mn(Cl/Br)(4)center dot 2H(2)O PA NCs showed excellent stability and strong photoluminescence, providing new design ideas for potential optoelectronic devices.

Nanopore-Based Fingerprint Immunoassay Based on Rolling Circle Amplification and DNA Fragmentation

Summary: In recent years, nanopore-based sequencers have shown great potential as robust tools in genomics applications. However, challenges remain in using nanopores for sensitive and quantitative diagnostic tools. One major limitation is the lack of sensitivity to detect disease biomarkers at low concentrations, while another limitation is the absence of unique nanopore signals for different analytes. To address these challenges, a strategy has been developed that utilizes immunocapture, amplification, and fragmentation techniques to release multiple DNA reporter molecules for nanopore detection. This approach allows for the identification and quantification of biomarker analytes. Future improvements can include integration with a nanopore array and microfluidics-based chemistry to enhance detection sensitivity, enable multiplexed biomarker detection, and reduce the size and cost of diagnostic devices.

Disulfide Bonds Are Not Necessary for Intrinsic TNSALP Activity

Summary: Recent developments in single-molecule enzymology have revealed subpopulations in enzyme ensembles. Tissue-nonspecific alkaline phosphatase (TNSALP), a model enzyme for these studies, has disulfide bonds critical for its dimerization. Mutations in these bonds have been found in patients with hypophosphatasia, a rare disease affecting bone and tooth mineralization. However, this study shows that these mutations do not significantly affect the enzymatic function of TNSALP, suggesting that the symptoms of hypophosphatasia may not be primarily due to impaired enzyme function, but rather reduced enzyme expression and trafficking.

Summary: The study found that clonal hematopoiesis of indeterminate potential (CHIP) is associated with an increased risk of liver inflammation and chronic liver disease progression. Furthermore, experiments showed that mice transplanted with Tet2-deficient hematopoietic cells exhibited more severe liver inflammation and fibrosis. These findings suggest that CHIP contributes to liver inflammation and the development of chronic liver disease through an aberrant inflammatory response.
