Summary: This review highlights the author's research on catalytic reactions using isoxazolidin-5-ones as substrates. Asymmetric catalysis has been effective for C-C bond formation at the carbonyl alpha-carbon, leading to the development of catalytic asymmetric allylation and direct Mannich-type reactions. The resulting products can be further converted into quaternary beta(2,2)-amino acids, showcasing the potential utility of isoxazolidin-5-ones in various chemical sciences.

Summary: The enantioselective addition of isoxazolidin-5-ones to the α-carbon of allenoates has been achieved using a novel spirobiindane-based quaternary ammonium salt catalyst under classical liquid-solid phase-transfer conditions. This method provides unprecedented highly functionalized β(2,2)-amino acid derivatives with good enantioselectivities and high yields. Further manipulations of the products have also been conducted.

Summary: The palladium-catalyzed addition of arylboronic acid to 2-cyano-(N-aryl)-acetamide or ethyl-2-cyanoacetate followed by subsequent reaction transforms them into biologically significant derivatives. The reaction conditions are robust enough to prevent hydrolysis of the ester/amide moiety during arylation, and can also convert the unactivated nitrile moiety in acetonitrile to the corresponding acetophenone derivative.

Summary: This study developed an efficient method for synthesizing 8-membered benzo[b]oxocines with high stereocontrol and yield through a sequential catalytic reaction. The chiral iridium catalyst was found to control enantioselectivity, while the chiral bifunctional squaramide catalyst controlled diastereoselectivity. The strategy was also demonstrated to be applicable for synthesizing other nitrogen heterocycles with high enantiomeric enrichment.

Summary: Type 2 diabetes mellitus, nonalcoholic fatty liver disease, cardio-cerebrovascular diseases, hyperuricemia and gout, and metabolic-related sexual dysfunction are metabolic diseases that affect human health. The findings from cell or animal models in previous research are challenging to apply clinically due to various differences. Traditional Chinese medicine has been used for over 2,200 years to maintain or improve cardiometabolic health, and our group has collected and studied the classic dietary treatments from ancient literature for metabolic diseases. The exact composition and active ingredients of these treatments are not standardized, and further research is needed.
