A new HIF-1α/RANTES-driven pathway to hepatocellular carcinoma mediated by germline haploinsufficiency of SART1/HAF in mice

The hypoxia-inducible factor (HIF), HIF-1, is a central regulator of the response to low oxygen or inflammatory stress and plays an essential role in survival and function of immune cells. However, the mechanisms regulating nonhypoxic induction of HIF-1 remain unclear. Here, we assess the impact of germline heterozygosity of a novel, oxygen-independent ubiquitin ligase for HIF-1: hypoxia-associated factor (HAF; encoded by SART1). SART1(-/-) mice were embryonic lethal, whereas male SART1(+/-) mice spontaneously recapitulated key features of nonalcoholic steatohepatitis (NASH)-driven hepatocellular carcinoma (HCC), including steatosis, fibrosis, and inflammatory cytokine production. Male, but not female, SART1(+/-) mice showed significant up-regulation of HIF-1 in circulating and liver-infiltrating immune cells, but not in hepatocytes, before development of malignancy. Additionally, Kupffer cells derived from male, but not female, SART1(+/-) mice produced increased levels of the HIF-1-dependent chemokine, regulated on activation, normal T-cell expressed and secreted (RANTES), compared to wild type. This was associated with increased liver-neutrophilic infiltration, whereas infiltration of lymphocytes and macrophages were not significantly different. Neutralization of circulating RANTES decreased liver neutrophilic infiltration and attenuated HCC tumor initiation/growth in SART1(+/-) mice. Conclusion: This work establishes a new tumor-suppressor role for HAF in immune cell function by preventing inappropriate HIF-1 activation in male mice and identifies RANTES as a novel therapeutic target for NASH and NASH-driven HCC. TACE versus TARE for patients with hepatocellular carcinoma: Overall and individual patient level meta analysis

Summary: This study conducted an overall and individual patient data (IPD) meta-analysis of transarterial radioembolization (TARE) and transarterial chemoembolization (TACE) for the treatment of hepatocellular carcinoma (HCC). The results showed that TARE had a significantly longer time to progression (TTP) compared to TACE, but there was no significant difference in overall survival (OS) between the two modalities. The limitations of the current data include inconsistent length of follow-up and response criteria. Therefore, prospective studies comparing these treatments are needed.

ISCA2 inhibition decreases HIF and induces ferroptosis in clear cell renal carcinoma

Summary: This study found that targeting ISCA2 protein can reduce HIF-1/2 alpha protein levels and inhibit the development of clear cell renal cell carcinoma (ccRCC) by inducing ferroptosis. ISCA2 inhibition not only decreases HIF-2 alpha protein levels by blocking IRE-dependent translation, but can also reduce HIF-1 alpha translation at higher concentrations through unknown mechanisms. Additionally, ISCA2 inhibition triggers the iron starvation response, leading to iron/metals overload and cell death.

The results showed a steady incidence rate of MANEC-P over the last two decades, with a poor prognosis and being the 5th leading cause of pancreatic cancer-related death in the US.

Summary: This study aimed to investigate the prevalence, severity, and contributing factors of non-alcoholic fatty liver disease (NAFLD) in Ghanaian individuals with type 2 diabetes mellitus (T2DM). The results showed a high prevalence of NAFLD (51.4%) among the T2DM participants in Ghana, with 11.6% having significant liver fibrosis. Obesity was found to be an independent predictor of NAFLD in individuals with T2DM in Ghana.

Summary: The study reveals that targeting the NAMPT/NAD+ pathway with FK866 can inhibit the growth of CCA cells and alter their mitochondrial metabolism. Additionally, FK866 enhances the anticancer effects of cisplatin in vitro. These findings suggest that NAMPT/NAD+ pathway may be a potential therapeutic target for CCA, and FK866 may serve as a useful medication in combination with cisplatin.

Analysis of N-linked Glycan Alterations in Tissue and Serum Reveals Promising Biomarkers for Intrahepatic Cholangiocarcinoma

Summary: There is a need to identify prognostic biomarkers for iCCA, and N-glycosylation alterations have shown potential for diagnostic strategies in various cancers. This study characterized N-glycan modifications in iCCA tissue and serum, and discovered that bisected fucosylated N-glycan structures were specific to iCCA tumor regions. These modifications were significantly upregulated in iCCA tissue and serum compared to HCC and bile duct disease. An algorithm based on these N-glycan modifications quadrupled the sensitivity of iCCA detection compared to the current biomarker CA19-9.
