Summary: In this study, a reduction-sensitive nitric oxide donor conjugate of furoxans-oxaliplatin was synthesized and a stroma-cell-accessible bioinspired lipoprotein system (S-LFO) was designed to facilitate CTL infiltration in tumors for anticancer immunotherapy. The S-LFO treatment promoted tumor vessel normalization, eliminated components of tumor stroma, increased the number of CD3(+)CD8(+) T cells, and facilitated their trafficking to cancer cell regions in tumors. Additionally, the combination of S-LFO with an antiprogrammed death ligand-1 showed therapeutic benefits of retarded tumor growth and extended survivals in three murine tumor models.

These findings contribute to a better understanding of the mechanisms of response and chemoresistance in pancreatic cancer, and may help to stratify patients for different treatment approaches.

Summary: This study investigates the impact of cGAS-STING expression and the presence of cancer-associated fibroblasts (CAFs) on immune cell infiltration in pancreatic ductal adenocarcinoma (PDAC). The results show that PDAC cases with co-expression of cGAS and STING have favorable survival and exhibit cytotoxic CD8 + T cell infiltrations. However, the presence of CAFs impedes immune cell infiltration.

The results suggest that more severe fibrosis, acinar atrophy, and chronic inflammation in the pancreatic stroma were associated with worse DSS outcomes for PDAC patients.

Summary: In this study, the combined antitumor activity of FAP-targeted and CLDN18.2-targeted CAR-T cells against PDAC was explored. The results showed that prior infusion of FAP-targeted CAR-T cells could eliminate CAFs and enhance the anti-PDAC efficacy of subsequently infused CLDN18.2-targeted CAR-T cells. Moreover, FAP-targeted CAR-T cells could suppress the recruitment of MDSCs and promote the survival of CD8(+) T cells and CAR-T cells in tumor tissue.
