Adverse Effects of Selected Markers on the Metabolic and Endocrine Profiles of Obese Women With and Without PCOS

The study found that PCOS results in a significant elevation of VEGF in obese females, but does not affect levels of kisspeptin and Vit D. It also leads to an increase in several lipid and hormonal abnormalities in obese females. PCOS influences the relationship between Kisspeptin and VEGF, as well as parameters such as glucose, LH or FSH, and LH/FSH ratio in obese females, while not affecting the relationship of Vit D with other parameters.
The aim of the present study, is to investigate the influence of obesity, with and without polycystic ovarian syndrome (PCOS), on the levels of kisspeptin, vitamin D (Vit D), and vascular endothelial growth factor (VEGF) and to explore the relationship between these parameters and endocrine and metabolic variables. The study group included 126 obese Saudi females. Of these 63 were suffering from PCOS while the rest were normo-ovulatory obese women (non-PCOS obese). In the obese PCOS, VEGF was almost four times as high as in the non-PCOS obese, while kisspeptin and Vit D did not differ. A highly significant elevation was recorded in the waist/hip (WHR), cholesterol, LDL-C, fasting glucose, LH, LH/FSH ratio, estradiol (E2), and testosterone, while hip circumference, leptin, progesterone, and sex hormone binding globulin (SHBG) were lower in the obese PCOS subjects. BMI, HDL-C, ghrelin, insulin, and FSH levels did not differ significantly between the two groups. The obese PCOS had the same level of insulin resistance as the non-PCOS group, as judged by QUICK Index. Correlation studies showed a significant negative correlation between kisspeptin and glucose and LH levels, and a positive correlation with LH/FSH ratio in obese PCOS while in the non-PCOS obese, the kisspeptin correlated positively with glucose, and there was no correlation with LH or LH/FSH. VEGF negatively correlated with FSH and positively with LH/FSH ratio in the non-PCOS obese but this was lost in the obese PCOS. PCOS had no effect on the correlation between Vit D and all studied parameters. Multiple regression analysis showed triglyceride as predictor variable for kisspeptin as a dependent variable, while, leptin is a predictor variable for VEGF as a dependent variable. ROC studies showed the highest sensitivity and specificity for VEGF (AOC=1.00), followed by LH/FSH ratio (AOC=0.979). In conclusion, our study shows that PCOS results in significant elevation of VEGF in obese females, while kisspeptin and Vit D levels are not affected. It also leads to elevation in several of the lipid and hormonal abnormalities in the obese females. In addition, PCOS influences relationship between Kisspeptin and VEGF and some parameters such as glucose, LH or FSH and LH/FSH ratio in obese females, but does not affect Vit D relationship with other parameter.

Summary: A series of benzopyran derivatives 4, 5, and 6a-i were synthesized through a cyclocondensation reaction. The synthesized compounds were characterized and evaluated for their inhibitory potential on the alpha-amylase enzyme. The results showed potent alpha-amylase inhibitory activity, with compound 6c exhibiting the highest activity compared to the standard acarbose.

Summary: The study aims to assess the multifaceted effects of the polycyclic aromatic hydrocarbon phenanthrene on the physiology of two bivalve species, Mytilus galloprovinciallis and Ruditapes decussatus, that are commonly consumed as seafood. The results showed that phenanthrene significantly reduced the bivalves' ability to tolerate desiccation and their Median Lethal Time, and also inhibited the activity of acetylcholinesterase. This study suggests that these bivalve species can be used as bioindicators to assess the environmental impact of phenanthrene in marine ecosystems. Rating: 8/10.

Ephedra alata Subsp. Alenda as a Novel Source of Bioactive Phytochemicals: Characterization Based on the Mass Spectrometry and Profiling of Antioxidant and Anti-Inflammatory Properties

Summary: The aim of this study was to analyze the phytochemical content of Ephedra alata pulp extract (EAP) and investigate its antioxidant and anti-inflammatory capacities. High-performance liquid chromatography-electrospray ionization-quadrupole-time-of-flight mass spectrometry (HPLC-ESI-QTOF/MS) was used for phytochemical analysis, and various in vitro antioxidant and anti-inflammatory tests were conducted. The results showed that EAP contains 42 metabolites, including flavonoids, sphingolipids, fatty acids, ephedrine derivatives, and amino acid derivatives. EAP exhibited strong antioxidant activity, as well as notable anti-inflammatory effects by inhibiting COX-1 and COX-2, preventing protein denaturation, and protecting membrane stabilization.

Prenatal Exposure to Acrylamide Differently Affected the Sex Ratio, Aromatase and Apoptosis in Female Adult Offspring of Two Subsequent Generations

Summary: In this study, the effects of acrylamide (ACR) exposure during pregnancy on the ovary of female adult offspring of two subsequent generations were investigated. The results showed that ACR exposure increased the number of antral follicles and corpora lutea in the first generation females (AF1) but led to an increase in the number of degenerating follicles and cysts in the second generation females (AF2). Additionally, ACR exposure reduced the levels of estradiol-producing enzyme CYP19A gene and induced excessive apoptosis. Overall, the ovarian conditions of AF1 females resembled the early perimenopausal stage, while those of AF2 females resembled the late perimenopausal stage in women.

Summary: Experimental studies have found that toluene has toxic effects on female reproductive processes. This in vitro study aimed to investigate the potential of ginkgo, fennel, and flaxseed extracts to modify the effects of toluene on ovarian hormone release. The extracts were found to reduce progesterone release and increase oxytocin release, while the release of prostaglandin F was inhibited by ginkgo and stimulated by fennel and flaxseed. Furthermore, fennel and flaxseed were able to prevent the adverse effects of toluene on ovarian hormones, while ginkgo enhanced these effects.

Fibromyalgia Rapid Screening Tool (FiRST): Arabic Translation and Cross-Cultural Adaptation and Validation

Summary: The present study aimed to analyze the polyphenolic profile of the ethyl acetate extract obtained from the bark of Tetraclinis articulata and evaluate its cytotoxic activity. The extract showed significant cytotoxic effect against HeLa cells. In silico analysis confirmed the binding energy and interaction mode of the major constituents in the extract. This study provides the first report on the polyphenolic composition and cytotoxic activity of the extract from T. articulata bark.

Can flaxseed, chia or puncture vine affect mare ovarian cell functions and prevent the toxic effect of the environmental contaminant toluene?

Summary: The aim of this in vitro study was to examine the potential effect of functional food plant extracts on mare ovarian cell functions and their response to the environmental contaminant toluene. The study found that toluene reduced cell proliferation, viability, and the release of certain hormones, but not oxytocin. Flaxseed and puncture vine extracts mitigated toluene's harmful effects, while chia extract had no effect. Toluene was able to modify the inhibitory effects of flaxseed and other plant extracts on hormone release. This study provides important insights into the effects of functional food plant extracts and toluene on mare ovarian cell functions.

Involvement of Autophagy and Oxidative Stress-Mediated DNA Hypomethylation in Transgenerational Nephrotoxicity Induced in Rats by the Mycotoxin Fumonisin B1

Summary: This study evaluated the transgenerational effects of Fumonisin B1 (FB1) exposure on the structure and function of kidneys in offspring. These findings highlight the nephrotoxicity and transgenerational effects of early-life FB1 exposure.
