Summary: Chronic and intermittent hyperglycemia affect insulin signaling in visceral pre/adipocytes by upregulating analyzed PI3K/AKT pathway molecules. Both mRNA and protein expression levels are more dependent on stage-specific events than the length of the period of high glucose exposure. Additionally, changes in miR expression seem to be involved in the regulation of PI3K/AKT expression in response to hyperglycemic stimulation.

Summary: Bone marrow adipose tissue (BMAT) is a significant component of the bone marrow space in healthy adults, but its expansion is associated with aging, obesity, anorexia nervosa, and irradiation. While traditionally viewed as a negative component, recent studies have revealed that BMAT can serve as an energy reservoir and also plays a role in regulating bone formation and hematopoiesis. This review summarizes the unique characteristics of BMAT and provides an updated understanding of its physiological roles based on a newly established mouse model.

Summary: This study analyzed the morphological and immunophenotypic changes of bone marrow adipocytes (BMAds) in different types of bone marrow cancers. The results showed that BMAds were significantly reduced in both bone marrow metastasis and myeloproliferative neoplasia with grade-3 myelofibrosis, along with a decrease in cell diameter and area. Additionally, the expression of certain markers in BMAds differed in these two types of cancers.

Summary: This study reveals that osteocytes regulate the differentiation of BMSCs into osteoblasts or adipocytes by secreting neuropeptide Y (NPY), with increased NPY expression associated with aging and osteoporosis. Deletion of NPY in osteocytes leads to high bone mass phenotype and attenuated bone-fat imbalance. The study suggests a new mode of neuronal control of bone metabolism through ANS-induced regulation of osteocyte NPY.

Summary: During obesity and high fat-diet feeding, there is an increase in pro-inflammatory macrophages in adipose tissue as well as invasive Ly6C(high) monocytes produced by the bone marrow. This study explores the relationship between bone marrow adipocytes and Ly6C(high) monocytes, and finds that the whitening of bone marrow adipocytes precedes the rise of adipose tissue macrophages during high fat-diet feeding in mice. Ex vivo treatment of bone marrow cells with conditioned media from bone marrow adipocytes or white adipocytes also favors the presence of Ly6C(high) monocytes. Furthermore, there is a metabolic reprogramming and mitochondrial fission in monocytes before the shift towards Ly6C(high) phenotype.

Summary: This study established comprehensive maps of sympathetic adrenergic and sensory peptidergic axons within and around the femur and tibia in mice, defining three distinct patterns of periosteal innervation. The results revealed a gradient of bone marrow axon density that increases along the length of the tibia and identified key regions of interest for neuroskeletal studies. This information serves as a guide for future studies and was compiled as a resource for the field as part of the NIH SPARC consortium.

Studies have shown the highly patterned innervation of bone, and the importance of balancing local and systemic neural functions to promote optimal skeletal repair while managing pain. A better understanding of the bidirectional pathways between the PNS and bone has important implications for skeletal development and regeneration.
