Summary: ACTH stimulation during adrenal venous sampling for primary aldosteronism can improve successful catheterization rates but may lead to decreased lateralization rates and potentially underestimate surgically remediable disease. Studies over the past 5 years have shown approximately 25% discordance in lateralization rates between unstimulated and stimulated sampling. The role of ACTH should primarily be limited to enhancing selectivity during adrenal venous sampling.

Summary: Diffuse midline glioma (DMG) is a devastating pediatric brain tumor with an urgent need for new treatment options. LIN28B RNA binding protein is overexpressed in DMG and suppresses the let-7 family of microRNAs, which in turn suppress a multitude of oncogenes. This review summarizes the role of the LIN28B-let-7-oncogene axis in different glioma subtypes and recommends future research focusing on DMG as a potential therapeutic target.

Summary: Increased activity of the insulin-like growth factor (IGF) axis is associated with the development and progression of colorectal cancer (CRC). The let-7 family of microRNAs (miRNAs) has been shown to target genes involved in the IGF signaling pathway and function as tumor suppressors. This study found that multiple let-7 miRNAs were down-regulated in CRC tissues, and that specific dysregulation of let-7 miRNAs was observed in CRC with perineural invasion. These findings suggest a relationship between the IGF axis, let-7 miRNAs, and CRC, as well as an association between the expression of these miRNAs and perineural invasion.

Summary: Insomnia, especially when associated with objective sleep disturbance, was found to be related to activation of the hypothalamic-pituitary-adrenal axis. Men with insomnia demonstrated shorter total sleep time and lower sleep efficiency on polysomnography, as well as lower levels of adrenocorticotropic hormone and cortisol after exposure to stress, suggesting a disrupted hypothalamic-pituitary-adrenal axis function in this population.
