Summary: This study demonstrates that the natural compound tectorigenin (TG) enhances GLUT4 expression, glucose uptake, and insulin responsiveness by activating AMPK/MEF2 signaling. TG-mediated enhancement of AMPK signaling and GLUT4 expression in skeletal muscle improves insulin resistance and type 2 diabetes. This study suggests that TG could be a potential drug candidate for the treatment of insulin resistance and its related diseases and provides new insights into PKA signaling in glucose metabolism in skeletal muscle.

Summary: This study investigates the effect of serpentine on insulin regulation of blood glucose through insulin receptors. The results show that serpentine can bind to insulin receptors, enhance insulin signaling pathway, and increase glucose uptake in cells. In a diabetic mouse model, serpentine prolongs the hypoglycemic effect of insulin, reduces the use of exogenous insulin, and inhibits endogenous insulin secretion. Therefore, serpentine may serve as an insulin sensitizer to assist in lowering blood glucose.

In this study, treatment with SGLT2i alone did not have significant effects on insulin resistance, kidney function, or blood pressure. However, when combined with CR, these interventions resulted in more profound improvements without causing chronic hypoglycemia.

Summary: The dynamic interaction between lipid droplets (LDs) and mitochondria plays a crucial role in the transfer of long-chain fatty acids (LCFAs) from LDs to mitochondria for oxidation in skeletal muscle during energy stress. The tethering complex comprising Rab8a and PLIN5 is identified as the mediator of LD-mitochondrion interaction. Activation of AMPK increases active GTP-bound Rab8a, promoting LD-mitochondrion interaction by binding to PLIN5 under starvation. The Rab8a-PLIN5 tethering complex recruits adipose triglyceride lipase (ATGL) to facilitate LCFA mobilization from LDs to mitochondria for oxidation. Rab8a deficiency impairs fatty acid utilization and reduces endurance during exercise in mice. These findings shed light on the regulatory mechanisms underlying the beneficial effects of exercise on lipid homeostasis control.

Summary: Emerging evidence suggests that the transcription factor ERRα plays a vital role in the development of pancreatitis. Pancreatitis is associated with an increase in ERRα gene expression, and its inhibition impairs the progression of the disease. Mechanistically, ERRα induces the expression of VDAC1, which modulates mitochondrial Ca2+ and ROS levels. Inhibition of the ERRα-VDAC1 axis alleviates mitochondrial dysfunction and suppresses pancreatitis progression. The therapeutic blockade of ERRα-VDAC1 pathway shows promise in mitigating pancreatitis in mouse models and human hereditary pancreatitis. These findings emphasize the importance of ERRα in pancreatitis and suggest its potential as a therapeutic target.
