Signaling pathways involved in cardiac energy metabolism

Various signaling pathways coordinate energy metabolism and contractile function in the heart. Myocardial uptake of long-chain fatty acids largely occurs by facilitated diffusion, involving the membrane-associated protein, CD36. Glucose uptake, the rate-limiting step in glucose utilization, is mediated predominantly by the glucose transporter protein, GLUT4. Insulin and contraction-mediated AMPK signaling each are implicated in tightly regulating these myocardial gate-keepers' of energy balance, that is, CD36 and GLUT4. The insulin and AMPK signaling cascades are complex and their cross-talk is only beginning to be understood. Moreover, transcriptional regulation of the CD36 and GLUT4 is significantly understudied. This review focuses on recent advances on the role of these signaling pathways and transcription factors involved in the regulation of CD36 and GLUT4.

Summary: The study reveals that PDK4 plays a crucial role in promoting rapid mitochondrial fragmentation in response to ETC toxins, and overexpression of PDK4 can induce mitochondrial fission even in the absence of mitochondrial stress. The investigation also identifies SEPT2 as a key effector molecule for PDK4 to regulate mitochondrial fission through DRP1 at the interface of cellular bioenergetics and mitochondrial dynamics.

Summary: Emerging evidence suggests that the transcription factor ERRα plays a vital role in the development of pancreatitis. Pancreatitis is associated with an increase in ERRα gene expression, and its inhibition impairs the progression of the disease. Mechanistically, ERRα induces the expression of VDAC1, which modulates mitochondrial Ca2+ and ROS levels. Inhibition of the ERRα-VDAC1 axis alleviates mitochondrial dysfunction and suppresses pancreatitis progression. The therapeutic blockade of ERRα-VDAC1 pathway shows promise in mitigating pancreatitis in mouse models and human hereditary pancreatitis. These findings emphasize the importance of ERRα in pancreatitis and suggest its potential as a therapeutic target.
