Rhabdomyolysis induced by antiepileptic drugs: characteristics, treatment and prognosis

Introduction: Rhabdomyolysis syndrome refers to a variety of factors that affect the striated muscle cell membrane, the membrane channels and its energy supply. Most cases of rhabdomyolysis are due to direct trauma. However, infection, toxins, drugs, muscle ischemia, electrolyte imbalance, metabolic diseases, genetic diseases and abnormal body temperature can also lead to rhabdomyolysis. Epilepsy is one of the most common chronic neurological diseases. The primary long-term treatment is antiepileptic drugs (AEDs), which may cause rhabdomyolysis. This article summarizes the characteristics, treatment methods and prognosis of patients with rhabdomyolysis that is induced by antiepileptic drugs. Areas covered: This review is based on PubMed, EMBASE and MEDLINE searches of the literature using the keywords epilepsy, antiepileptic drugs,status epilepticus,rhabdomyolysis, and antiepileptic drugs and rhabdomyolysis syndrome as well as extensive personal clinical experience with various antiepileptic drugs. Potential relationships between antiepileptic drugs and rhabdomyolysis are discussed. Expert opinion: Worldwide, there are approximately 50 million epilepsy patients, most of whom are treated with drugs. Reports have indicated that the majority of antiepileptic drugs on the market can cause rhabdomyolysis. Although rhabdomyolysis induced by antiepileptic drugs is a rare condition with a low incidence, this condition has serious consequences and merits attention from clinicians.

Idebenone Protects against Atherosclerosis in Apolipoprotein E-Deficient Mice Via Activation of the SIRT3-SOD2-mtROS Pathway

Summary: This study demonstrated that idebenone acts as a mitochondrial protective agent by inhibiting NLPR3 activation via the SIRT3-SOD2-mtROS pathway, significantly reducing plaque burden and plaque size while stabilizing atherosclerotic plaques. Up-regulation and down-regulation of proteins were found in idebenone-treated mice, with SIRT3, SOD2, and NLRP3 expression significantly regulated both in vivo and in vitro. Furthermore, idebenone protected against endothelial cell damage and inhibited the production of mitochondrial reactive oxygen species in cholesterol-treated HUVECs.

Summary: This study found that the drug edaravone, by activating the mTORC1 signaling pathway, promotes the maturation of oligodendrocyte progenitor cells and enhances remyelination in NMOSD. Edaravone demonstrated significant remyelination ability in experiments and has potential therapeutic effects for treating NMOSD lesion repair.

Combining idebenone and rosuvastatin prevents atherosclerosis by suppressing oxidative stress and NLRP3 inflammasome activation

Summary: This study aimed to investigate the anti-atherosclerotic effect of the combination of idebenone and statins. The results showed that both idebenone and statins reduced plaque burden and increased plaque stability, with the combined treatment showing even better effects. Additionally, the combination therapy also reduced oxidative stress and cell apoptosis. In conclusion, combining idebenone and statins appears to be a promising strategy for further preventing atherosclerosis.

Pleiotrophin ameliorates age-induced adult hippocampal neurogenesis decline and cognitive dysfunction

Summary: This study reveals that cognitive impairment is associated with a decline in adult hippocampal neurogenesis (AHN). Pleiotrophin (PTN) expression decreases with aging, leading to impaired AHN and poor learning and memory. The study shows that PTN interacts with protein tyrosine phosphatase receptor type Z1 (PTPRZ1) to promote neural stem and progenitor cell (NSPC) proliferation and differentiation through AKT signaling. Overexpression of PTN or pharmacological activation of AKT signaling in aging mice restores AHN and improves memory deficits.

CHI3L1 signaling impairs hippocampal neurogenesis and cognitive function in autoimmune-mediated neuroinflammation

Summary: Chitinase-3-like protein 1 (CHI3L1) is a reliable biomarker for inflammatory central nervous system (CNS) conditions and is associated with neurodegeneration and autoimmune disorders. This study found that elevated CHI3L1 expression correlates with cognitive impairment in patients with neuromyelitis optica (NMO). CHI3L1 inhibits the proliferation and neuronal differentiation of neural stem cells, leading to decreased hippocampal neurogenesis and impaired learning behaviors. Blocking the CHI3L1/CRTH2/beta-catenin cascade restores neurogenesis and improves cognitive deficits, suggesting potential therapeutic development in neuroinflammatory disorders.
