Small leucine-rich repeat proteoglycans in corneal inflammation and wound healing

The small leucine rich repeat proteoglycans are major components of the cornea. Lumican, keratocan, decorin, biglycan and osteoglycin are present throughout the adult corneal stroma, and fibromodulin in the peripheral limbal area. In the cornea literature these proteoglycan have been reviewed as structural, collagen fibril-regulating proteins of the cornea. However, these proteoglycans are members of the leucine-rich-repeat superfamily, and share structural similarities with pathogen recognition toll-like receptors. Emerging studies are showing that these have a range of interactions with cell surface receptors, chemokines, growth factors and pathogen associated molecular patterns and are able to regulate host immune response, inflammation and wound healing. This review discusses what is known about their innate immune-related role directly in the cornea, and studies outside the field that find interesting links with innate immune and wound healing responses that are likely to be relevant to the ocular surface. In addition, the review discusses phenotypes of mice with targeted deletion of proteoglycan genes and genetic variants associated with human pathologies. (C) 2016 Elsevier Ltd. All rights reserved.

Summary: The impact of SARS-CoV-2 and COVID-19 susceptibility is influenced by age and health status, with a challenge in inducing effective immune responses in elderly populations. While the stalk domain of spike protein plays a minor role in viral neutralization, mutations in the receptor-binding domain may reduce vaccine efficacy across ages. A proposal for chimeric vaccines combining multiple proteins aims at addressing these challenges.

Summary: This review focuses on the emerging immune regulatory functions of small leucine-rich proteoglycans (SLRP) such as lumican, keratocan, fibromodulin, biglycan, and decorin. These SLRPs have been extensively studied as structural components in the extracellular matrix, but their immune regulatory roles in inflammation and disease are not well understood. By investigating the leucine-rich repeat (LRR) motifs of SLRP core proteins and their relationships with other pathogen recognition receptors, this review aims to provide molecular clarity on the immune regulatory functions of SLRPs.

Single cell RNA-seq of human cornea organoids identifies cell fates of a developing immature cornea

Characterization of neural damage and neuroinflammation in Pax6 small-eye mice

Summary: Aniridia is a panocular condition characterized by the loss of iris, caused mainly by mutations in the PAX6 gene. This study assessed the effects of Pax6-haploinsufficiency on retinal morphology and vision in Pax6Sey mice. The results showed elevated intraocular pressure and declining visual acuity in Pax6Sey mice, along with local retinal damage and a possible neuroinflammatory response.

Regulated cell death pathways in the sodium iodate model: Insights and implications for AMD

Summary: The sodium iodate (NaIO3) model is a valuable tool in studying dry AMD and evaluating the retinal structure. Through extensive research, it has been found that the death of RPE cells caused by NaIO3 injection is closely related to vision loss in patients with dry AMD. Therefore, studying NaIO3 can provide insights into the mechanisms of RPE and photoreceptor cell death and contribute to understanding RPE degeneration in AMD.

A new model of axon degeneration in the mouse optic nerve using repeat intraocular pressure challenge

Summary: We present a new experimental model for inducing retinal ganglion cell (RGC) dysfunction and degeneration in mice. This model allows for the reliable induction of RGC dysfunction and loss, with the resilience of the optic nerve being dependent on the age of the animal. The time-defined nature of the functional and structural changes observed in this model will facilitate the study of glial responses and the evaluation of neuroprotective treatments after intraocular pressure (IOP) injury.
