Co-delivery of paclitaxel and STAT3 siRNA by a multifunctional nanocomplex for targeted treatment of metastatic breast cancer

Kaipei Luo, Yi Gao, Shaoping Yin, Yawen Yao, Hua Yu, Guangji Wang, Juan Li

Summary: Metastasis is a major cause of mortality in breast cancer patients, and the signal transducer and activator of transcription 3 (STAT3) is closely related to cancer metastasis. In this study, a multifunctional nanocomplex co-loaded with paclitaxel (PTX) and STAT3 siRNA was developed, showing superior efficacy in inhibiting tumor growth and suppressing metastasis by silencing STAT3 expression. The nanocomplex provides a promising platform for targeted treatment of metastatic cancer through chemo-gene combined therapy.

ROS-responsive fluorinated polyethyleneimine vector to co-deliver shMTHFD2 and shGPX4 plasmids induces ferroptosis and apoptosis for cancer therapy

Suleixin Yang, Peng Hua, Chengwei He, Hua Yu, Dan Shao, Zhi Shi, Meiwan Chen, Ka Hong Wong

Summary: In this study, a ferroptosis/apoptosis combinational therapy was achieved by depleting GSH and downregulating GPX4 to disrupt redox homeostasis and amplify ferroptosis-related oxidation effect. ROS-responsive serum-resistant nanoparticles were used to co-deliver shGPX4 and shMTHFD2 plasmids, leading to efficient transfection, lysosomal escape, and induction of ferroptosis and apoptosis in tumor cells.

Summary: In this study, the similarity and difference between Camellia nitidissima Chi (CC), C. euphlebia Merr. ex Sealy (CE), and C. insularis (CI) were comprehensively compared in terms of chemistry and pharmacology. A statistical model combining untargeted metabolomics and fingerprint analyses was developed for chemical pattern recognition and discrimination of the three yellow camellias species. The results showed that CC, CE, and CI could be distinguished based on specific chemical combinations and that they exhibited differences in anti-inflammation/anti-oxidation activities.
