Metabolomics analysis reveals a modified amino acid metabolism that correlates with altered oxygen homeostasis in COVID-19 patients

Using gas chromatography coupled to mass spectrometry, we identified significant changes in serum metabolites associated with severe and mild COVID-19 patients, particularly in altered amino acid catabolism. The study found that three alpha-hydroxyl acids of amino acid origin increased with disease severity, correlating with changes in oxygen saturation levels and clinical markers of lung damage. The conversion of alpha-keto acids to alpha-hydroxyl acids is suggested to play a role in maintaining NAD recycling in patients with altered oxygen levels during SARS-CoV-2 infection, highlighting the potential importance of amino acid supplementation.
We identified the main changes in serum metabolites associated with severe (n=46) and mild (n=19) COVID-19 patients by gas chromatography coupled to mass spectrometry. The modified metabolic profiles were associated to an altered amino acid catabolism in hypoxic conditions. Noteworthy, three alpha -hydroxyl acids of amino acid origin increased with disease severity and correlated with altered oxygen saturation levels and clinical markers of lung damage. We hypothesize that the enzymatic conversion of alpha -keto-acids to alpha- hydroxyl-acids helps to maintain NAD recycling in patients with altered oxygen levels, highlighting the potential relevance of amino acid supplementation during SARS-CoV-2 infection.

Summary: In this study, a murine experimental model of immune complex-mediated brain vasculitis induced by cell wall antigens of Mycobacterium tuberculosis (Mtb) was established. The results showed that high levels of immune complexes were formed and deposited in the brain, lung, and kidneys, leading to vascular inflammation and meningitis. This suggests a significant role of immune complexes in the pathogenesis of tuberculosis.

Summary: This study investigated the benzylation reaction of curcumin and tetrahydrocurcumin with benzyl bromide and potassium carbonate in acetone. It was found that curcumin undergoes O-alkylation first, followed by C-1 substitution, while THC undergoes these reactions in reverse order. The antioxidant and cytotoxic activities of the benzylated derivatives were studied, and the C-dibenzylated curcuminoid showed superior activity compared to the tribenzylated and tetrabenzylated derivatives. DFT calculations and docking calculations on a DNA fragment were conducted to explain the experimental results and the preferences for nucleophilic attack.

Summary: This study aimed to investigate the features associated with the persistence of post-COVID-19 syndrome. A cohort study of 102 COVID-19 patients was conducted, and it was found that obesity, pro-inflammatory state, and increased cellular senescence are independently associated with the persistence of post-COVID-19 syndrome. These findings have important implications for understanding and preventing post-COVID-19 syndrome.

Salivary IgA subtypes as novel disease biomarkers in systemic lupus erythematosus

Summary: This study investigated the levels of IgA in the saliva of SLE patients and found that total IgA and IgA1 levels were significantly elevated in saliva of SLE patients. Furthermore, salivary IgA levels, particularly IgA2, positively correlated with anti-dsDNA IgG antibody titers. Additionally, the presence of salivary anti-nucleosome IgA antibodies in SLE patients, not previously reported elsewhere, was also detected. According to the results, characterization of IgA in saliva could be a potentially helpful tool in the clinical care of SLE patients.

Ball-Milling Preparation of the Drug-Drug Solid Form of Pioglitazone-Rosuvastatin at Different Molar Ratios: Characterization and Intrinsic Dissolution Rates Evaluation

Summary: Ball-milling using neat grinding or liquid-assisted grinding with different solvents allowed the formation of various solid forms of pioglitazone hydrochloride (PGZ·HCl) and rosuvastatin calcium (RSV). By adjusting the molar ratios of the reactants, coamorphous forms with different stoichiometries were obtained. In addition, mixtures of solid forms coexisted with the coamorphous forms when the drug ratio was not stoichiometric. The dissolution profiles of these solid forms were evaluated, and a synchronized release of both drugs was observed. The coamorphous 2:1 form showed an improved dissolution rate compared to pure PGZ·HCl, which may be attributed to pH changes caused by the degradation processes of RSV.

Fungal Infections in the ICU during the COVID-19 Pandemic in Mexico

Summary: This study describes the prevalence, related factors, and outcomes of Invasive Fungal Infections (IFI) in critically ill patients with COVID-19. The overall prevalence of IFI was 9.3%, with 5.6% being COVID-19-associated pulmonary aspergillosis (CAPA) and 2.5% being invasive candidiasis (IC). Factors related to IFI included higher SOFA scores and empiric antibiotic use for COVID-19. CAPA was the most frequent type of IFI.

Summary: A coamorphous salt in a 1:1 ratio of Fluvastatin sodium (FLV) and Pioglitazone hydrochloride (PGZ center dot HCl) can be prepared by ball milling through neat grinding (NG), while the salt-cocrystal continuum is preferably formed by liquid-assisted grinding (LAG) using ethanol (EtOH). Attempts to prepare the coamorphous salt from the salt-cocrystal continuum by neat grinding were unsuccessful. Ball milling by NG or LAG can access a great variety of solid forms (PGZ center dot HCl-FLV 1:1): coamorphous with hexane; physical mixture with ethyl acetate; salt-cocrystal continuum with EtOH; and immiscible components with water.

A novel use of an oxime molecule as supramolecular tecton. Synthesis of a Pt (II) 2D network

Summary: To test the hypothesis, we synthesized and determined the crystal structure of the compound [PtCl3(4-HoxpyH)], 1, which shows that the ligand HoxpyH coordinates through the nitrogen atom of the oxime group and forms the expected H-bonds. In the crystal, six molecules interact through hydrogen bonds and O center dot center dot center dot Cl interactions, forming rings that extend into two-dimensional structures. These 2D structures are stacked on top of each other through Pt center dot center dot center dot pyH and Pt-Cl center dot center dot center dot pyH interactions, without interpenetrating or forming channels.

Summary: We analyzed antimicrobial resistance (AMR) data of clinical isolates from 43 centers in Mexico, including Escherichia coli, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, Enterococcus faecium, and Staphylococcus aureus. The study investigated the site of care, patient age, and clinical specimen type to determine AMR patterns. The findings revealed that ICU patients had the highest AMR in E. coli, K. pneumoniae, and P. aeruginosa isolates, while non-ICU patients had higher AMR in A. baumannii isolates. The oldest age group showed the highest AMR in E. coli, E. faecium, and S. aureus, while the 19-59 age group had the highest AMR in A. baumannii and P. aeruginosa. Additionally, higher AMR was observed in E. coli, K. pneumoniae, and P. aeruginosa isolates from blood specimens, and the most frequently detected carbapenemase-encoding gene in E. coli was blaNDM (84%).

Summary: This study aimed to identify factors associated with 90-day all-cause mortality in patients with CR-GNB infections. The results showed that age, immunosuppression, and septic shock at diagnosis were independently associated with death within 90 days after CR-GNB infection. Additionally, receiving antibiogram-guided appropriate treatment was independently associated with a lower risk of death. These findings have important implications for clinical practice and addressing the problem of carbapenem resistance.
