Bisphenols are endocrine-disrupting chemicals that have been linked to cancer progression, particularly prostate cancer. They are found in the urine of the vast majority of the population and could potentially impact the normal physiology and cancer development of the bladder.
Bisphenols are endocrine-disrupting chemicals found in a broad range of products that can modulate hormonal signalling pathways and various other biological functions. These compounds can bind steroid receptors, e.g. estrogen and androgen receptors, expressed by numerous cells and tissues, including the prostate and the bladder, with the potential to alter their homeostasis and normal physiological functions. In the past years, exposure to bisphenols was linked to cancer progression and metastasis. As such, recent pieces of evidence suggest that endocrine-disrupting chemicals can lead to the development of prostate cancer. Moreover, bisphenols are found in the urine of the wide majority of the population. They could potentially affect the bladder's normal physiology and cancer development, even if the bladder is not recognized as a hormone-sensitive tissue. This review will focus on prostate and bladder malignancies, two urological cancers that share standard carcinogenic processes. The description of the underlying mechanisms involved in cell toxicity, and the possible roles of bisphenols in the development of prostate and bladder cancer, could help establish the putative roles of bisphenols on public health.

Impacts of endocrine-disrupting chemicals on prostate function and cancer

Aurelie Lacouture, Camille Lafront, Cindy Peillex, Martin Pelletier, Etienne Audet-Walsh

Summary: EDCs can modulate the functions of sex-steroid receptors, indirectly impact androgen and estrogen pathways, and have multiple cellular targets beyond sex-steroid receptors, leading to various potential effects. This article reviews the association between prostate cancer and the endocrine-disrupting functions of four prominent EDC families, suggesting common guidelines for studying EDCs in the context of endocrine diseases.

Comprehensive overview of the available pharmacotherapy for the treatment of non-neurogenic overactive bladder in children

Sophie Ramsay, Elisabeth Lapointe, Stephane Bolduc

Summary: Overactive bladder (OAB) is a chronic condition highly prevalent in children, and conservative measures should be the first line of treatment. When conservative management fails, pharmacological options are considered, although only two agents are currently approved for pediatric use. Treatment for OAB in children requires a structured, stepwise approach, active involvement of children and their families, and close monitoring of side effects and medication adherence.

Bladder cancer cell lines adapt their aggressiveness profile to oxygen tension

Stephane Chabaud, Eve Pellerin, Christophe Caneparo, Cassandra Ringuette-Goulet, Frederic Pouliot, Stephane Bolduc

Summary: The hypoxic condition during tumor growth has a significant impact on the survival and migration properties of bladder cancer cells. Low-grade bladder cancer cells tend to become more aggressive under hypoxic culture conditions, while high-grade cells tend to exhibit less aggressive characteristics. Hypoxia greatly alters the properties of bladder cancer cells, including matrix metalloproteinases expression and metabolic changes. Studying bladder cancer cells in hypoxic environments is vital for translating in vitro findings to an in vivo context.

Cancer-Associated Fibroblasts in a 3D Engineered Tissue Model Induce Tumor-like Matrix Stiffening and EMT Transition

Martial Millet, Enola Bollmann, Cassandra Ringuette Goulet, Genevieve Bernard, Stephane Chabaud, Marc-Etienne Huot, Frederic Pouliot, Stephane Bolduc, Francois Bordeleau

Summary: A study on the physical properties of tumors demonstrates that a 3D self-assembly bladder model constructed with cancer-associated fibroblasts can effectively recapitulate the biological and mechanical features of tumors. The model exhibited increased extracellular matrix content, remodeling, and stiffness in the stroma assembled by cancer-associated fibroblasts. Additionally, normal urothelial cells seeded on the tumor model displayed tumor-like responses, including increased cell proliferation, cell infiltration, and features of epithelial-to-mesenchymal transition.

UPEC Colonic-Virulence and Urovirulence Are Blunted by Proanthocyanidins-Rich Cranberry Extract Microbial Metabolites in a Gut Model and a 3D Tissue-Engineered Urothelium

Charlene Roussel, Stephane Chabaud, Jacob Lessard-Lord, Valentina Cattero, Felix-Antoine Pellerin, Perrine Feutry, Valerie Bochard, Stephane Bolduc, Yves Desjardins

Summary: Uropathogenic Escherichia coli (UPEC) are the primary cause of recurrent urinary tract infections. The poor understanding of UPEC ecology-pathophysiology from its reservoir-the gut, to its infection site-the urothelium, partly explains the inadequate and abusive use of antibiotics to treat UTI, which leads to a dramatic upsurge in antibiotic-resistance cases. In this study, the effects of cranberry proanthocyanidins (PAC)-rich extract microbial metabolites on UPEC virulence and fitness were evaluated. The results demonstrated that PAC-rich cranberry extract microbial metabolites significantly reduce the activation of UPEC virulence genes in the gut reservoir, suggesting their potential in preventing UTI.
