A mild and one-pot activation approach of activated carbon using boric acid as the activator for the adsorption of tetracyclines antibiotics in water was studied. Boric acid activated carbon (BAC) from bioresource showed higher removal efficiency than biochar and had a maximum adsorption capacity of 173.9 mg/g for TCs. The interactions between BAC and TCs were identified as hydrogen bond interaction, pi-pi interaction, and electrostatic interaction, influenced by the surface charge of BAC and TCs' ions species in different pH environments. Adsorption kinetics, isotherm models, and thermodynamics were thoroughly studied, demonstrating a spontaneous and favorable process.
A mild and one-pot activation approach of activated carbon was found. The feasibility of boric acid as the activated reagent which was used for the adsorption of four tetracyclines antibiotics (TCs) in water. Boric acid activated carbon (BAC) from bioresource has a much higher removal efficiency than currently reported biochar. The maximum adsorption capacity of BAC is 173.9 mg/g for TCs. BAC is an ecofriendly, nontoxic, and low-cost absorbent from sawdust waste. BAC and TCs could keep coalescing at least 55 days on the surface without stable release. BAC was fully characterized by using scanning electron microscopy, X-ray photoelectron spectroscopy, X-ray diffraction, Raman, zeta potential, and Brunauer-Emmett-Teller analysis; the large surface area and rich pore structure were proved. The interaction between BAC and TCs are hydrogen bond interaction, pi-pi interaction, and electrostatic interaction. These interactions are also related to the surface charge of BAC and the TCs' species of ions in different pH. Furthermore, the adsorption kinetics and adsorption isotherm of BAC were studied thoroughly. The pseudo-first-order, pseudo-second-order, intra-particle diffusion, Elovich Langmuir, Freundlich, and Dubinin-Radushkevich models were fitted and the physical adsorption process was proved. After the study on adsorption thermodynamics, adsorption exhibits a spontaneous and favorable process.

Post-Translational Modifications of G Protein-Coupled Receptors Revealed by Proteomics and Structural Biology

Bingjie Zhang, Shanshan Li, Wenqing Shui

Summary: G protein-coupled receptors (GPCRs) are a large family of proteins that play significant roles in cellular and physiological responses. Post-translational modifications (PTMs) of GPCRs, such as glycosylation, phosphorylation, ubiquitination and palmitoylation, have important regulatory functions in receptor folding, biosynthesis, trafficking, dimerization and signaling. This review provides an overview of PTMs in GPCRs, focusing on their types, locations, cross talk and dynamic regulation, which can shed light on the molecular basis of GPCRs and contribute to structure-based drug discovery.

Summary: This study reveals that the articular microenvironment in osteoarthritis (OA) impairs the therapeutic effects of human umbilical cord-derived mesenchymal stromal cells (hUC-MSCs) due to the activation of the aryl hydrocarbon receptor (AHR) pathway by tryptophan metabolites, leading to compromised chondrogenesis and chondroprotection of hUC-MSCs.

Summary: NTT-seq is a novel experimental method capable of measuring the presence of multiple histone modifications and protein-DNA binding sites at single-cell resolution. It utilizes recombinant Tn5 transposase fused to secondary nanobodies that specifically bind different immunoglobulin-G antibodies. By applying NTT-seq, high-resolution multimodal maps of chromatin states can be generated and it can be extended to study cell surface protein expression and multimodal chromatin states simultaneously.

Hydrophobic-action-driven removal of six organophosphorus pesticides from tea infusion by modified carbonized bacterial cellulose

Jianhui Zhu, Jingyang Yu, Bingjie Zhang, Changsheng Li, Jianli Wang, Jiawen Ji, Desheng Liu, Rumin Gao, Jianzhang Li, Yongqiang Ma

Summary: The abuse of organophosphorus pesticides in tea planting poses potential health risks to consumers. Reliable techniques are needed to remove these pesticides from tea infusion. This study investigated the modification of carbonized bacterial cellulose (CBC) to improve its adsorption performance. The N-CBC treated with hydrazine hydrate showed the best adsorption effect, with a removal rate 13 times higher than CBC. The study also elucidated the adsorption mechanism and suitable models to describe the process, and confirmed no significant changes in tea infusion quality after N-CBC treatment.

Summary: This study aimed to develop an efficient adsorption method for recovering melanoidins from thermal hydrolyzed sludge (THS) by macroporous resin. XAD761 resin was found to be the most appropriate for melanoidins extraction. The adsorption kinetics followed a pseudo-second-order model and the process was confirmed to be physical, spontaneous, and exothermic.

Affinity selection of double-click triazole libraries for rapid discovery of allosteric modulators for GLP-1 receptor

Ye Xin, Shuo Liu, Yan Liu, Zhen Qian, Hongyue Liu, Bingjie Zhang, Taijie Guo, Garth J. Thompson, Raymond C. Stevens, K. Barry Sharpless, Jiajia Dong, Wenqing Shui

Summary: The recently developed double-click reaction sequence is promising in expanding the number and diversity of 1,2,3-triazole derivatives. This study focused on the design, synthesis, and screening of double-click triazole libraries for drug discovery, using the glucagon-like-peptide-1 receptor as a challenging drug target. The streamlined synthesis of customized triazole libraries on an unprecedented scale led to the identification of positive allosteric modulators with unreported scaffolds. The unexpected binding mode of these modulators could provide insights into their mechanism of action.

Summary: This study aims to develop a methodology for identifying, isolating, and characterizing the melanoidins formed during thermal hydrolysis of sludge. Melanoidin formation was confirmed using physicochemical indicators and fluorescence analysis. The macroporous resin adsorption method successfully extracted melanoidins with high recovery and selectivity. The extracted melanoidins were found to consist of carbohydrates, proteins, phenols, and exhibited heterocyclic macromolecular structures. This research provides fundamental knowledge on the characteristics and formation pathways of melanoidins during sludge thermal hydrolysis.

Summary: In this study, the conformational ensemble of an activated GPCR-G protein complex is mapped at near-atomic resolution using cross-linking mass spectrometry (CLMS) and integrative structure modeling. The integrative structures reveal heterogeneous conformations for potential alternative active states of the GLP-1 receptor-Gs complex, showing marked differences from the cryo-EM structure. The functional significance of interface residue contacts observed only in the integrative structures is validated through alanine scanning mutagenesis coupled with pharmacological assays.

Sources and budget analysis of ambient formaldehyde in the east-central area of the yangtze River Delta region, China

Dongyang Liu, Ming Wang, Kun Hu, Zhiqiang Liu, Huabin Dong, Bingjie Zhang, Xin Zhao, Wenhao Ji

Summary: This study investigated the sources, production, and destruction pathways of ambient formaldehyde (HCHO) in the east-central area of the Yangtze River Delta (YRD) region. The results suggested that secondary production was the largest contributor to HCHO concentration, especially on non-attainment days of ozone (O3). The main production pathways for HCHO were reactions of alkoxyl radicals with oxygen (O2), while the main destruction pathways were photolysis and reaction with OH radical. Isoprene, trans-2-butene, ethylene, cis-2-butene, 1,2,4-trimethylbenzene, m/p-xylene, styrene, 1,2,3-trimethylbenzene, propene, and 1,2,4-trimethyl-benzene were identified as key precursors for HCHO production.
