Summary: Th17 cells play a crucial role in the pathogenesis of autoimmune disorders, and their differentiation is regulated by lipid metabolism and the activation of ROR?t. Inhibition of lipid biosynthesis suppresses Th17 cell differentiation. Manipulating cellular lipid metabolism provides a novel therapeutic approach for treating autoimmune disorders.

Summary: SENP2 is involved in regulating post-translational modification of Smad4 in Th17 cells and intestinal bowel diseases. Lack of Senp2 exacerbates the severity of experimental colitis and leads to elevated levels of pathogenic Th17 cells and dysbiosis of the intestinal microbiome. SENP2 enzymatic activity is important for deSUMOylation of Smad4 and regulates the pathogenicity of Th17 cells.

Summary: Primary bile acids (BAs), byproducts of cholesterol metabolism, are closely associated with the pathogenesis of cholestatic liver diseases. In this study, a synthesized sulfated product of lithocholic acid (LCA) was found to selectively inhibit the differentiation of Th17 cells, without affecting other types of T cells. This finding provides clues for further exploration of the relationship between bile acid metabolites and T lymphocytes.

Summary: This research reveals that RORyt not only regulates the differentiation of TH17 cells, but also plays a role in their effector function. By mutating K256R, which prevents ubiquitination of RORyt, the distinct functions of RORyt in TH17 differentiation and effector function can be separated.

Summary: Our study found that there was an enhanced level of 27-OHC in the serum of individuals with mild cognitive impairment (MCI). The balance of Th17-related cytokines was altered, with increased levels of IL-17A, IL-12p70, IL-23, GM-CSF, MIP-3 alpha, and TNF-alpha, but decreased levels of IL-13, IL-28A, and TGF-beta 1. Further experiments showed that 27-OHC induced higher immunogenicity, leading to an increase in Th17 cells and a decrease in Treg cells. The amyloidosis was aggravated, cognitive deficits occurred, and the learning and memory performance were worsened in mice treated with 27-OHC. However, by inhibiting ROR gamma t in 27-OHC-loaded mice, the balance of Th17/Treg cells was restored, the amyloidosis was decreased, and the learning and memory decline was improved.

Summary: Using structure-based virtual screening, this study identified a compound called DDC, which is abundant in plants and food, as a potential immunomodulator. DDC inhibits the activity of ROR gamma t, a transcription factor involved in Th17 cell differentiation, thus regulating Th17/Treg balance and showing promise in the treatment of inflammatory diseases like multiple sclerosis, inflammatory bowel disease, and organ transplant rejection.

Summary: Topical application of BAC in C57BL/6 mice induced an increase in CD4(+)CD69(+), CD4(+)IFN-gamma(+), and CD4(+)IL-17(+) cells, leading to corneal barrier dysfunction, goblet cell loss, and decreased tear production. Adoptive transfer of CD4(+) T cells from BAC-treated mice into nude mice resulted in ocular surface manifestations similar to direct topical BAC treatment, suggesting a role of CD4(+) T cells in BAC-induced ocular surface disorder.

Summary: Thermal metamaterials provide rich control of heat transport by breaking the Onsager reciprocity and introducing thermal convection, leading to a regime beyond effective heat conduction. A continuous switch from thermal cloaking to thermal concentration is demonstrated in a liquid-solid hybrid thermal metamaterial with external tuning. This switch is achieved by tuning the liquid flow, resulting in a topology transition in the virtual space of the thermotic transformation. These findings illustrate the extraordinary heat transport in complex multicomponent thermal metamaterials and pave the way toward an unprecedented regime of heat manipulation.

Summary: The study reported a 21-year-old Chinese man with late-onset glutaric aciduria type II (GA II) who presented with muscle weakness, vomiting, and severe pain. Muscle biopsy revealed lipid storage myopathy, and urine organic acid analyses showed an increase in glycolic acid, consistent with GA II. Whole-exome sequencing identified two novel compound heterozygous missense mutations in the ETFDH gene, expanding the list of mutations associated with GA II. Considering the treatability of this disease, GA II should be considered in patients with muscular symptoms and acute metabolism decompensation.

Summary: We investigated the phenotypic values of panicle elongation length (PEL) in 353 rice accessions across six environments. We identified and validated three significant quantitative trait loci (QTL) associated with PEL, namely qPEL4, qPEL6, and qPEL9, of which qPEL9 is a novel QTL. The allele PEL9(GG) was found to increase the PEL and outcrossing rate in the female parents of hybrid rice, with its frequency gradually increasing with latitude in the Northern Hemisphere. These findings will help improve the PEL of female parents in hybrid rice.

Summary: In this study, square-root higher-order topological insulator phases were discovered in an acoustic metamaterial with a decorated rectangular lattice. The boundary-dependent topological edge and corner states in both the parent and square-root acoustic metamaterials were explored using acoustic pump-probe techniques. Various simulations were used to validate their higher-order topologies and spectral connections. This work not only confirms the existence of square-root higher-order topology in rectangular lattice systems experimentally but also paves the way for versatile higher-order topological phenomena in photonic, acoustic, and mechanical metamaterials.

Summary: Controllable negative photoconductance (NPC) detectors based on organic-gated carbon nanotube field-effect transistors (OG-CNT FETs) are reported, with the strong influence of light-induced electrostatic doping on the nonconventional photoresponse demonstrated. The optimized OG-CNT FETs exhibit high responsivity of 72.6 A W-1 at 880 nm, along with improved response/recovery times of 7 and 5 ms. Reversible switching between NPC and positive photoconductance (PPC) is observed under the same light illumination, offering a new approach for achieving bidirectional photoresponses and paving the way for multifunctional optoelectronic systems.

Summary: This paper proposes an adversarial learning paradigm to enhance the intelligence and robustness of driving policy for signalized intersections with dense traffic flow. It introduces a static path planner to generate trackable candidate paths for different intersection topologies and builds a constrained optimal control problem considering the uncertainty of the driving environment. Adversarial policy gradient is used to solve the problem by introducing disturbances while the driving policy learns to handle the situation through competition. Simulation results show that the trained policy can handle signal lights flexibly and achieve smooth and efficient passing with a humanoid paradigm, while APG greatly improves the resistance to abnormal behaviors, ensuring a high level of safety for autonomous vehicles.

They experimentally realize the hybrid topological photonic crystal and verify its unconventional dual-band gap topological edge states.
