The application of antimicrobial photodynamic inactivation on methicillin-resistant S. aureus and ESBL-producing K. pneumoniae using porphyrin photosensitizer in combination with silver nanoparticles

This study investigated the synergistic antibacterial effect of the photosensitizer TMPyP and silver nanoparticles (AgNPs) in antimicrobial photodynamic inactivation, showing promising results against MRSA and ESBL-KP strains. By determining the optimal combination of TMPyP and AgNPs, it was concluded that a treatment including silver nanoparticles could be a promising approach to treat bacterial infection.
As resistance of bacterial strains to antibiotics is a major problem, there is a need to look for alternative treatments. One option is antimicrobial photodynamic inactivation (aPDI). The pathogenic cells are targeted by a nontoxic photosensitizer while the surrounding healthy tissue is relatively unaffected. The photosensitizer is activated by light of t appropriate wavelength resulting in the generation of reactive oxygen species that are cytotoxic for the pathogens. In this work, the photosensitizer TMPyP and silver nanoparticles (AgNPs) were investigated for their synergistic antibacterial effect. We tested these two substances on two bacterial strains, methicillin-resistant Staphylococcus aureus 4591 (MRSA) and extended-spectrum beta-lactamases-producing Klebsiella pneumoniae 2486 (ESBL-KP), to compare their effectiveness. The bacteria were first incubated with TMPyP for 45 min or 5 h, then irradiated with a LED source with the total fluence of 10 or 20 J/cm(2) and then placed in a microbiological growth medium supplemented with AgNPs. To accomplish the synergistic effect, the optimal combination of TMPyP and AgNPs was estimated as 1.56 +/- 25 mu M for TMPyP and 3.38 mg/l for AgNPs in case of MRSA and 1.56 +/- 50 mu M for TMPyP and 3.38 mg/l for AgNPs in case of ESBL-KP at 45 min incubation with TMPyP and fluence of 10 J/cm(2). Longer incubation and/or longer irradiation led to a decrease in the maximum values of the photosensitizer concentration to produce the synergistic effect. From this work it can be concluded that the combination of antimicrobial photodynamic inactivation with a treatment including silver nanoparticles could be a promising approach to treat bacterial infection.

Infectious complications of induction treatment for acute myeloid leukaemia using the 7+3 protocol without antibiotic prophylaxis-15 years of experience of one clinical site

Summary: This study retrospectively analyzed the incidence of infectious complications in AML patients without routine antibiotic prophylaxis in a clinical site in Czech Republic over a 15-year period. The results showed that infectious complications were common, but infectious mortality was comparable to patients with prophylaxis. Additionally, achieving complete remission had a significant effect on the duration of febrile episodes and overall survival, while infection or fever of unknown origin at diagnosis increased the colonization of multi-drug resistant bacterial strains and prolonged antibiotic therapy.

Restoration of antibacterial activity of inactive antibiotics via combined treatment with a cyanographene/Ag nanohybrid

Summary: This study reports an effective way of enhancing and restoring the antibacterial activity of inactive antibiotics by using a cyanographene/Ag nanohybrid. The combined treatment showed synergistic and partial synergistic effects against multiresistant bacterial strains, indicating the potential of the nanohybrid in overcoming antibiotic resistance. The study also revealed that the interaction between the antibiotic's mode of action and the bacterial resistance mechanism strongly influenced the efficacy of the combined treatment.

Pulmonary Complications after COVID-19

Summary: According to the study, COVID-19 patients hospitalized 29 days or more after infection most commonly had respiratory tract infections, with pneumonia being the most frequent complication. Patients with infectious diseases were hospitalized sooner after testing positive for PCR and had more non-respiratory complications. The overall in-hospital mortality rate was 5.1%.

Analysis of Bacterial Pathogens Causing Complicating HAP in Patients with Secondary Peritonitis

The study emphasizes the importance of initial antibiotic therapy effective against enterobacteria, ESBL and AmpC β-lactamases producing strains, Pseudomonas aeruginosa, and Enterococcus faecalis in patients with secondary peritonitis complicated by HAP. Monitoring the respiratory tract bacterial microflora in these patients is also highlighted as important.

Lipophosphonoxins-A Novel Group of Broad Spectrum Antibacterial Compounds
