ReMOT Control Delivery of CRISPR-Cas9 Ribonucleoprotein Complex to Induce Germline Mutagenesis in the Disease Vector Mosquitoes Culex pipiens pallens (Diptera: Culicidae)

This study demonstrates the successful application of ReMOT Control technique in Culex pipiens pallens mosquitoes, providing basic parameters for gene editing and facilitating research in mosquito biology and control.
The wide distribution of Culex (Cx.) pipiens complex mosquitoes makes it difficult to prevent the transmission of mosquito-borne diseases in humans. Gene editing using CRISPR/Cas9 is an effective technique with the potential to solve the growing problem of mosquito-borne diseases. This study uses the ReMOT Control technique in Culex pipiens pallens (L.) to produce genetically modified mosquitoes. A microinjection system was established by injecting 60 adult female mosquitoes-14 mu l injection mixture was required, and no precipitation occurred with <= 1 mu l of endosomal release reagents (chloroquine or saponin). The efficiency of delivery of the P2C-enhanced green fluorescent protein-Cas9 (P2C-EGFP-Cas9) ribonucleoprotein complex into the ovary was 100% when injected at 24 h post-bloodmeal (the peak of vitellogenesis). Using this method for KMO knockout, we found that gene editing in the ovary could also occur when P2C-Cas9 RNP complex was injected into the hemolymph of adult Cx. pipiens pallens by ReMOT Control. In the chloroquine group, of the 2,251 G(0) progeny screened, 9 individuals showed with white and mosaic eye phenotypes. In the saponin group, of the 2,462 G(0) progeny screened, 8 mutant individuals were observed. Sequencing results showed 13 bp deletions, further confirming the fact that gene editing occurred. In conclusion, the successful application of ReMOT Control in Cx. pipiens pallens not only provides the basic parameters (injection parameters and injection time) for this method but also facilitates the study of mosquito biology and control.

This study aimed to explore the value of intraoperative neurophysiological monitoring (IONM) of penile sensory-evoked potential (PSEP) for standardizing SDN procedures. The results showed that IONM may improve short-term therapeutic effectiveness and reduce complications in SDN surgeries.

A novel NGS-based diagnostic algorithm for classifying multifocal lung adenocarcinomas in pN0M0 patients

Summary: In this study, NGS analysis was used to diagnose MLA patients with MPLAs and IPMs. By incorporating a predefined NGS criterion, accurate classification of IPMs was achieved. A refined algorithm was developed that combined NGS and histologic results to correctly diagnose MPLAs and IPMs. This NGS-based algorithm showed promising potential in complementing traditional CHA-based MLA diagnosis and helping determine patient staging and treatment.

Rare mutation-dominant compound EGFR-positive NSCLC is associated with enriched kinase domain-resided variants of uncertain significance and poor clinical outcomes

Summary: This retrospective study analyzed next-generation sequencing (NGS) data from 1025 non-small cell lung cancer (NSCLC) patients with compound epidermal growth factor receptor (EGFR) mutations. Different subtypes of compound EGFR mutations were associated with distinct clinical features, genetic architectures, and prognostic outcomes. Rare mutation-dominant subtypes with variations of uncertain significance (VUSs) residing in the kinase domain had poor clinical outcomes and implied disruptions in kinase domain regulation/function.

Potential dependent mechanism of the composition and electrochemical property of oxide films of Ti-6Al-3Nb-2Zr-1Mo

Summary: The potential-dependent mechanism of oxide film formation on Ti-6Al-3Nb-2Zr-1Mo in 0.5 M H2SO4 was investigated within a potential range of -0.80-5.00 V. The thicknesses of oxide films and the valence states of Ti, Al, Nb, and Mo were found to increase with increasing potential to 3.00 V, while surface roughness showed an opposite trend. The results indicated that higher potentials in the passive region led to better protection and uniformity of the oxide film. A schematic considering the effect of alloying elements within oxide films formed at different applied potentials on Ti-6Al-3Nb-2Zr-1Mo was established.

Comprehensive profiling of EGFR mutation subtypes reveals genomic-clinical associations in non-small-cell lung cancer patients on first-generation EGFR inhibitors

Summary: In this study, the genomic profiles of NSCLC patients with cEGFR and uEGFR mutations were analyzed using targeted next-generation sequencing. The results showed that uEGFR-positive patients were more likely to have co-occurring genetic alterations in the Hippo pathway and a higher TMB. Smoking-related mutations were also enriched in uEGFR-positive patients. Subgroup analyses revealed potential prognostic biomarkers and significant differences in mutational profiles among patients with different EGFR sub-type mutations.

The relationship between the distribution of facial erythema and skin type in rosacea patients: a cross-sectional analysis

Summary: This study found that individuals with rosacea have different facial erythema distribution patterns, which may be related to skin types. The erythema distribution pattern of rosacea was divided into peace signs, wing shapes, and neither of the two patterns. The peace sign pattern was more common in individuals with darker skin tones, while the wing shape pattern was associated with the lateral parts of the cheeks. This has important implications for understanding the pathogenesis and treatment of rosacea.

Mutations in the MET tyrosine kinase domain and resistance to tyrosine kinase inhibitors in non-small-cell lung cancer

Summary: This study found that MET tyrosine kinase domain (TKD) mutations were detected in both baseline and patients treated with TKIs in non-small-cell lung cancer (NSCLC) patients. MET-H1094Y may be associated with acquired resistance after recurrent EGFR-TKIs treatment. Preliminary data suggests that EGFR-mutated NSCLC patients who acquired MET-V1092I or MET-H1094Y may benefit from combinatorial therapy with EGFR-TKI and MET-TKI, providing new insights into personalized medical treatment.
