Physical activity is associated with reduced cancer risk and improved quality of life for patients; sedentary behavior and obesity are linked to cancer development; exercise training can help mitigate side effects of cancer treatment, but personalized exercise plans are necessary.
Physically active behavior has been associated with a reduced risk of developing certain types of cancer and improved psychological conditions for patients by reducing anxiety and depression, in turn improving the quality of life of cancer patients. On the other hand, the correlations between inactivity, sedentary behavior, and overweight and obesity with the risk of development and progression of various cancers are well studied, mainly in middle-aged and elderly subjects. In this article, we have revised the evidence on the effects of physical activity on the expression and release of the adipose-tissue-derived mediators of low-grade chronic inflammation, i.e., adipokines, as well as the adipokine-mediated impacts of physical activity on tumor development, growth, and metastasis. Importantly, exercise training may be effective in mitigating the side effects related to anti-cancer treatment, thereby underlining the importance of encouraging cancer patients to engage in moderate-intensity activities. However, the strong need to customize and adapt exercises to a patient's abilities is apparent. Besides the preventive effects of physically active behavior against the adipokine-stimulated cancer risk, it remains poorly understood how physical activity, through its actions as an adipokine, can actually influence the onset and development of metastases.

Micro-RNA: A Future Approach to Personalized Diagnosis of Bone Diseases

Giovanni Lombardi, Edgard Delvin

Summary: Osteoporosis is a prevalent bone disease and circulating microRNAs have shown potential as diagnostic and prognostic markers. However, the lack of standardization and harmonization of methods limits their application.

Impact of 12-Week Moderate-Intensity Aerobic Training on Inflammasome Complex Activation in Elderly Women

Marta Gomarasca, Katarzyna Micielska, Martina Faraldi, Marta Flis, Silvia Perego, Giuseppe Banfi, Ewa Ziemann, Giovanni Lombardi

Summary: Exercise can alleviate the chronic low-grade inflammatory status in elderly individuals by inhibiting the activation of the inflammasome and downstream effectors. Aerobic training reduces the expression of inflammasome and Toll-like receptors and decreases the secretion of pro-inflammatory cytokines. However, participants experience an acute inflammatory response at the end of the training.

Changes in selected exerkines concentration post folk-dance training are accompanied by glucose homeostasis and physical performance improvement in older adults

Ewa Aleksandra Rodziewicz-Flis, Malgorzata Kawa, Jan Jacek Kaczor, Marzena Szaro-Truchan, Damian Jozef Flis, Giovanni Lombardi, Ewa Ziemann

Summary: The study aimed to evaluate the impact of folk-dance and balance training on physical performance, insulin resistance, and blood pressure in older adults. Both training programs effectively improved physical performance and blood pressure, accompanied by changes in selected exerkines. Folk-dance training also enhanced insulin sensitivity.

Long non-coding RNAs in bone metastasis: progresses and perspectives as potential diagnostic and prognostic biomarkers

Paola Maroni, Marta Gomarasca, Giovanni Lombardi

Summary: Long non-coding RNAs (lncRNAs) have potential as biomarkers for early cancer diagnosis, prognosis, and therapeutic targets. Bone metastasis is a common progression of advanced cancers, particularly breast, prostate, and lung cancers. Understanding the pathophysiology of bone metastasis and improving clinical management are challenging but important areas of research.

Osteocalcin modulates parathyroid cell function in human parathyroid tumors

Chiara Verdelli, Giulia Stefania Tavanti, Irene Forno, Valentina Vaira, Riccardo Maggiore, Leonardo Vicentini, Paolo Dalino Ciaramella, Francesca Perticone, Giovanni Lombardi, Sabrina Corbetta

Summary: This study found that osteocalcin (OC), a bone matrix protein, can modulate the function of parathyroid tumor cells by activating the calcium sensing receptor (CASR). OC reduces the activity of pERK/ERK and increases the levels of active beta-catenin through CASR activation. It also affects the expression of related genes to influence the physiological functions of parathyroid tumor cells.

Exploring cellular senescence in the musculoskeletal system: Any insights for biomarkers discovery?

Laura Gerosa, Amir Mohammad Malvandi, Marco Malavolta, Mauro Provinciali, Giovanni Lombardi

Summary: The locomotor system, consisting of skeletal muscles and bones, plays a vital role in maintaining function and preventing frailty. As individuals age, chronic disorders of the locomotor system may arise, negatively impacting bone and muscle function. Senescent cells, which accumulate in muscle tissue with advancing age or under pathological conditions, impair muscle regeneration and contribute to muscle weakness. Additionally, senescence in the bone microenvironment affects bone turnover, increasing the risk of osteoporosis. Understanding the mechanisms underlying cellular senescence in musculoskeletal tissues may aid in developing effective biomarkers for early detection of tissue flaws.

Metabolism of vitamin D is not affected by sport activity

Anna Ksiazek, Aleksandra Zagrodna, Giovanni Lombardi, Malgorzata Slowinska-Lisowska

Summary: The main objective of this study was to assess 25-(OH)DF, vitamin D metabolites, and VMRs in inactive men and athletes. The results showed that regular long-term physical activity had no effect on the concentration of vitamin D metabolites at rest. Furthermore, free vitamin D did not correlate more strongly with vitamin D metabolites and VMRs compared to total.

Summary: Novel markers of vitamin D status, such as free 25-(OH)D and the vitamin D metabolite ratio, show seasonal rhythms in athletes, while markers of psychophysical stress and free 25-(OH)D do not exhibit clear rhythmicity. Additionally, strong correlations exist between vitamin D metabolites in different training periods.

Effects of running a marathon on sclerostin and parathyroid hormone concentration in males aged over 50

Aleksandra Zagrodna, Anna Ksiazek, Malgorzata Slowinska-Lisowska, Jan Chmura, Piotr Ponikowski, Giovanni Lombardi

Summary: The aim of this study was to investigate the impact of running a marathon on sclerostin and intact PTH concentration in middle-aged males. The results showed that the levels of serum sclerostin and iPTH increased immediately after the marathon race but returned to baseline after 7 days. It was suggested that running a marathon affected skeletal metabolism by inhibiting bone formation through sclerostin expression and inducing long-lasting PTH elevation to maintain adequate calcium levels for neuromuscular activity and muscle contraction.

Evaluating safety risks of whole-body cryotherapy/cryostimulation (WBC): a scoping review from an international consortium

Fabien D. Legrand, Benoit Dugue, Joe Costello, Chris Bleakley, Elzbieta Miller, James R. Broatch, Guillaume Polidori, Anna Lubkowska, Julien Louis, Giovanni Lombardi, Francois Bieuzen, Paolo Capodaglio

Summary: Whole-body cryotherapy is a non-pharmacological treatment that influences inflammatory events and has various benefits, such as improved sleep quality, faster neuromuscular recovery, and chronic pain relief. It may also have positive effects on mental health and cognitive functions. There have been concerns about its safety, but these concerns are often based on misunderstandings regarding the differences between whole-body cryotherapy and partial-body cryotherapy. This article reviews the literature and concludes that the safety risks associated with whole-body cryotherapy are within acceptable limits and can be prevented by following existing recommendations and guidelines.
