Summary: The study aims to verify the International Chronic Ocular Graft-Versus-Host Disease (ICCGVHD) Group diagnostic criteria and establish an easy-to-use and reliable diagnosis model for quick identification of chronic oGVHD. Through observations and examinations of 180 patients who underwent allogeneic hematopoietic stem cell transplantation, significant differences were found in five complaints and six ophthalmic examinations between patients with chronic oGVHD and the control group. A three-variable-based nomogram was developed for the identification of chronic oGVHD, which showed excellent performance in the validation cohorts.

Summary: Combining XBJ with a reduced dose of CsA showed superior efficacy in protecting gut microbiota, attenuating aGVHD progression, and improving survival rates in a murine model. This regimen also reduced levels of proinflammatory cytokines and reversed dysbiosis at various levels in the gut microbiota of aGVHD mice. The combination treatment appeared to enhance intestinal tissue integrity and reduce intestinal permeability in aGVHD mice, potentially offering a novel strategy to manage aGVHD and improve quality of life for allo-HSCT recipients.

Summary: This study performed molecular and functional characterization of candidate cytokine genes in allogeneic hematopoietic cell transplantation (HCT) and found that recipients carrying the -1082GG IL10 gene promoter variant had a lower incidence of GVHD. This is the first study to report genome-to-cellular evidence for a non-HLA donor immunogenetic variant that appears to be protective against GVHD.

Comprehensive and sustained multidisciplinary efforts are crucial for successful management of cGVHD.

Summary: In this study, it was found in a mouse model that gentamicin (GM) significantly suppressed systemic cGVHD phenotypes and ocular manifestations, and reduced inflammatory cell infiltration and fibrosis in cGVHD-targeted organs. Although GM maintained higher levels of regulatory T cells, there were fewer Th17 cells and IL-6-producing macrophages in cGVHD-targeted organs. These findings suggest that orally administered GM may have positive effects in a cGVHD mouse model.

Summary: Ocular graft-versus-host disease (GVHD) is a serious complication after allogeneic hematopoietic stem cell transplantation. It greatly impacts the quality of life of affected individuals due to impaired visual function, pain, and other symptoms. However, the diagnosis and treatment of ocular GVHD pose challenges for hematologists and ophthalmologists, as there are no unified international criteria. This review explores the complex pathogenesis of ocular GVHD, providing a comprehensive summary of the disease's pathogenic mechanisms, related tear biomarkers, and clinical characteristics. Novel therapies based on these mechanisms are also discussed, offering new insights into ocular GVHD treatment.

The results showed that plasma alpha-ketoglutaric acid levels were consistently elevated before and at the onset of cGVHD, and late aGVHD had a unique metabolomic pattern at an early stage compared to cGVHD.
