Parametric Estimation of Reference Signal Intensity for Semi-Quantification of Tau Deposition: A Flortaucipir and [18F]-APN-1607 Study

Huiwei Zhang, Min Wang, Jiaying Lu, Weiqi Bao, Ling Li, Jiehui Jiang, Chuantao Zuo

Summary: The PERSI-WM method, when used as a reference region, improves the accuracy of measuring Tau retention in AD patients compared to the traditional method of using the cerebellum as a reference in both cohorts. This method shows superior diagnostic discrimination capabilities and can enhance the efficacy of semi-quantification of multiple tau tracers in PET imaging, indicating its potential for large-scale clinical application.

The impact of probable rapid eye movement sleep behavior disorder on Parkinson's disease: A dual-tracer PET imaging study

Qian Xu, Chengfeng Jiang, Jingjie Ge, Jiaying Lu, Ling Li, Huan Yu, Jianjun Wu, Jian Wang, Ping Wu, Chuantao Zuo

Summary: This study aimed to compare differences between PD patients with and without probable RBD and the influence of timing of RBD onset. The results showed that PD patients with probable RBD have worse dopaminergic dysfunction and higher metabolic activity in specific brain regions. Furthermore, RBD preceding PD onset may indicate a steeper disease decline.

Dopamine transporter imaging in progressive supranuclear palsy: Severe but nonspecific to subtypes

Qi-Si Chen, Xin-Yi Li, Ling Li, Jia-Ying Lu, Yi-Min Sun, Feng-Tao Liu, Chuan-Tao Zuo, Jian Wang

Summary: This study compared the dopaminergic lesion patterns of PSP with MSA-P and PD, and found that the severity of DAT loss in the striatum was more severe in PSP patients compared to PD and MSA-P patients, especially in the caudate. Among different subtypes of PSP, no significant difference was detected.

Summary: This study aims to provide a potential auxiliary tool for the differential diagnosis of parkinsonism using deep learning to decode the information in dopamine transporter (DAT) imaging. By employing a 3D deep convolutional neural network, this study successfully achieved a high accuracy rate in differentiating idiopathic Parkinson's disease (IPD), multiple system atrophy (MSA), and progressive supranuclear palsy (PSP). The saliency map and deep-learning-guided radiomics features provided more specific guidance for the diagnosis and quantitative analysis of parkinsonism.

Image-level trajectory inference of tau pathology using variational autoencoder for Flortaucipir PET

Summary: This study used a data-driven approach to identify the tau trajectory and quantify tau progression through a continuous latent space learned by VAE. The inferred tau trajectory in line with Braak staging showed tau first deposits in the parahippocampal and amygdala before spreading to other specific brain regions.

Hoeglinger, Claudio Bassetti, Yihui Guan, Wolfgang H. Oertel, Wolfgang Weber, Axel Rominger, Jian Wang, Chuantao Zuo, Kuangyu Shi

Summary: This study developed metabolic imaging indices based on deep learning to support the differential diagnosis of early idiopathic Parkinson disease (IPD) and atypical parkinsonian syndromes. The proposed indices showed high sensitivity and specificity in the blind-test cohort and were robust when dealing with discrepancies between populations and imaging acquisitions.

Jiaying Lu, Jingjie Ge, Keliang Chen, Yimin Sun, Fengtao Liu, Huan Yu, Qian Xu, Ling Li, Zizhao Ju, Huamei Lin, Yihui Guan, Qihao Guo, Jian Wang, Chuantao Zuo, Ping Wu

Summary: This study investigates the relationship between dementia with Lewy bodies (DLB) and Parkinson's disease dementia (PDD) and finds highly similar metabolic patterns between the two. The metabolic patterns of DLB, PDD, and Parkinson's disease (PD) can be utilized for disease diagnosis and progression monitoring.

Summary: This study systematically investigated the localization and magnitude of F-18-Florzolotau uptake in patients with the parkinsonian subtype of multiple system atrophy (MSA-P). The results showed elevated F-18-Florzolotau uptake in the putamen, globus pallidus, and dentate in MSA-P patients, which was not observed in patients with Parkinson's disease (PD). This increased signal was significantly associated with the core symptoms of MSA-P.
