Exopolysaccharide Producing Bifidobacterium animalis subsp. lactis Strains Modify the Intestinal Microbiota and the Plasmatic Cytokine Levels of BALB/c Mice According to the Type of Polymer Synthesized

Bacteria-host interactions are mediated by different microbial associated molecular patterns which are most often surface structures such as, among others, exopolysaccharides (EPSs). In this work, the capability of two isogenic EPS-producing Bifidobacterium animalis subsp. lactis strains to modulate the gut microbiota of healthy mice, was assessed. Each strain produces a different type of polymer; the ropy strain S89L synthesized a rhamnose-rich, high-molecular weight EPS in highest abundance than the non-ropy DMS10140 one. BALB/c mice were orally fed for 10 days with milk-bifidobacterial suspensions and followed afterward for 7 post-intervention days (wash-out period). The colonic content of mice was collected in several sampling points to perform a metataxonomic analysis. In addition, the influence of specific microbial clades, apparently stimulated by the ropy and non-ropy strains, on mouse plasmatic cytokine levels was investigated through hierarchical association testing. Analysis of 16S rRNA gene sequences showed that the abundance of Firmicutes phylum significantly increased 7 days after cessing the treatment with both strains. The relative abundance of Alloprevotella genus also rose, but after shorter post-treatment times (3 days for both DMS10140 and S89L strains). Some bacterial clades were specifically modulated by one or another strain. As such, the non-ropy DMS10140 strain exerted a significant influence on Intestinomonas genus, which increased after 4 post-administration days. On the other hand, feeding with the ropy strain S89L led to an increase in sequences of Faecalibaculum genus at 4 post-treatment days, while the abundance of Erysipelotrichaceae and Lactobacillaceae families increased for prolonged times. Association testing revealed that several lactobacilli and bifidobacterial significantly stimulated by ropy S89L strain were positively associated with the levels of certain cytokines, including IL-5 and IL-27. These results highlight relevant changes in mice gut microbiota produced after administration of the ropy S89L strain that were associated to a potential immune modulation effect.

Fermentation of chickpea flour with selected lactic acid bacteria for improving its nutritional and functional properties

Summary: This study aims to improve the nutri-functional quality of chickpea flour through fermentation with selected lactic acid bacteria. The results show that fermentation with LAB significantly increased LAB population, acidity, total phenolic contents, and removed antinutritional factors. Fermented doughs were used to make crackers with higher TPC and antioxidant activity. Fermentation is a better method than other processing methods for enhancing the properties of chickpea flour.

The gut microbiota, a key to understanding the health implications of micro(nano)plastics and their biodegradation

Summary: Plastic debris has a global impact on the environment, plants, animals, and human health, with a particular focus on micro(nano)plastics (MNPs). MNPs are frequently found in the food chain, leading to increased human intake. They have been detected in various types of consumed foods, drinking water, and human feces. The gastrointestinal tract, especially the gut, constantly interacts with MNPs as oral ingestion becomes the main source of exposure. Although the consequences of human exposure to MNPs remain unclear, studies have shown that MNPs of different types and sizes can affect gut intestinal bacteria, leading to dysbiosis, frequent pathogen outbreaks, and metabolic disorders. Furthermore, the presence of micro- and nano-plastic particles in animal tissues raises the question of whether human gut bacteria play a role in the bio-transformation of ingested MNPs. This article discusses these issues and reveals the complex interplay between MNPs and the human gut microbiome, highlighting the need for further research on the biological consequences of this interaction.

Immunomagnetic Capture of Faecalibacterium prausnitzii Selectively Modifies the Fecal Microbiota and Its Immunomodulatory Profile

Evaluation of a Shotgun Metagenomics Approach for Detection of ESBL- and/or Carbapenemase-Producing Enterobacterales in Culture Negative Patients Recovered from Acute Leukemia

Summary: Patients diagnosed with acute leukemia often have a weakened immune system, making infections a major concern. The results showed no presence of these bacteria, indicating successful decolonization and positive implications for future clinical management. Further studies are needed to validate the utility of metagenomics in routine screening of multidrug-resistant bacteria in onco-hematological patients.

Arabinoxylan-based substrate preferences and predicted metabolic properties of Bifidobacterium longum subspecies as a basis to design differential media

Summary: Arabinoxylan (AX) and arabinoxylo-oligosaccharides (AXOS) are prebiotics that promote beneficial species in the human gut, such as bifidobacteria. Bifidobacterium longum and B. longum subsp. longum have the highest capability to metabolize complex AX substrates, while B. longum subsp. infantis also possesses some machinery related to AXOS metabolization. A differential carbohydrate free medium (CFM) supplemented with AX and AXOS was developed to selectively isolate novel probiotic bifidobacteria that can metabolize structurally complex AX-derived prebiotics.

PARP-1 Expression Influences Cancer Stem Cell Phenotype in Colorectal Cancer Depending on p53

Summary: Poly(ADP-ribose) polymerase-1 (PARP-1) is involved in various physiological processes. Elevated PARP-1 expression is found in several tumors, associated with stemness and tumorigenesis. In colorectal cancer patients, PARP-1 expression correlated with differentiation grade, with a significant association only in tumors with wild-type p53. Additionally, PARP-1 and cancer stem cell markers were positively correlated in these tumors. In vitro studies showed that PARP-1 regulates cancer stem cell phenotype depending on p53 status, with overexpression increasing marker expression and sphere forming ability in wild-type p53 cells, while reducing these features in mutated p53 cells.
