Detection of urinary miRNAs for diagnosis of clear cell renal cell carcinoma

The lack of symptoms at the early stages of clear cell renal cell carcinoma (ccRCC) allows the tumour to metastasize, leading to a dramatic reduction in patient survival. Therefore, we studied and set up a method based on urinary microRNAs (miRNAs) for the diagnosis of ccRCC. First, miRNA expression in ccRCC specimens and kidney tissues from healthy subjects (HSs) was investigated through analysis of data banks and validated by comparing expression of miRNAs in ccRCC and adjacent non-cancerous kidney tissue specimens by RT-qPCR. Subsequently, we developed an algorithm to establish which miRNAs are more likely to be found in the urine of ccRCC patients that indicated miR-122, miR-1271, and miR-15b as potential interesting markers. The evaluation of their levels and three internal controls in the urine of 13 patients and 14 HSs resulted in the development of a score (7p-urinary score) to evaluate the presence of ccRCC in patients. The resulting area under the Receiver Operating Characteristic (ROC) curve, sensitivity, and specificity were equal to 0.96, 100% (95% CI 75-100%), and 86% (95% CI 57-98%), respectively. In conclusion, our study provides a proof of concept that combining the expression values of some urinary miRNAs might be useful in the diagnosis of ccRCC.

A previously developed algorithm based on the level of specific small RNAs in urine showed promising results in predicting the presence of kidney cancer. This study further validated the algorithm's effectiveness and demonstrated its potential application in noninvasive early diagnosis of ccRCC.

Association of GILZ with MUC2, TLR2, and TLR4 in Inflammatory Bowel Disease

Summary: Ulcerative colitis (UC) and Crohn's Disease (CD) are chronic relapsing inflammatory diseases caused by genetic, environmental, and immune factors. The glucocorticoid-induced leucine zipper (GILZ) plays a role in inflammatory bowel disease (IBD) and is associated with genes MUC2, TLR2, and TLR4 in the mucosal barrier. Direct correlations between GILZ/TLR2 and GILZ/TLR4 were found in healthy subjects (HSs) and UC patients, while an inverse correlation was observed between GILZ and MUC2 in HS and IBD patients, particularly those with active disease.

Overexpression of Potential Markers of Regulatory and Exhausted CD8+ T Cells in the Peripheral Blood Mononuclear Cells of Patients with B-Acute Lymphoblastic Leukemia

Summary: B-acute lymphoblastic leukemia (B-ALL) is a common pediatric cancer. Treg and exhausted CD8(+) T cells play important roles in its development and maintenance. In this study, the expression of 20 Treg/CD8 exhaustion markers was evaluated and their correlation with other factors was analyzed. The results suggest that T cells expressing CD39, CTLA-4, TNFR2, TIGIT, and TIM-3 may promote B-ALL progression and targeted immunotherapy against these markers could be a promising approach for treating B-ALL.

Summary: This study aimed to evaluate the intra- and postoperative complications, as well as the oncological and functional outcomes, of robot-assisted radical prostatectomy with pelvic lymph node dissection in patients with high-risk prostate cancer. The results showed that this technique is safe and effective, with only a few low-grade intra- and postoperative complications.
