Mitochondria-related transcriptional signature is downregulated in adipocytes in obesity: a study of young healthy MZ twins

Low mitochondrial activity in adipose tissue is suggested to be an underlying factor in obesity and its metabolic complications. We aimed to find out whether mitochondrial measures are downregulated in obesity also in isolated adipocytes. We studied young adult monozygotic (MZ) twin pairs discordant (n = 14, intrapair difference Delta BMI >= 3 kg/m(2)) and concordant (n = 5, Delta BMI < 3 kg/m(2)) for BMI, identified from ten birth cohorts of 22- to 36-year-old Finnish twins. Abdominal body fat distribution (MRI), liver fat content (magnetic resonance spectroscopy), insulin sensitivity (OGTT), high-sensitivity C-reactive protein, serum lipids and adipokines were measured. Subcutaneous abdominal adipose tissue biopsies were obtained to analyse the transcriptomics patterns of the isolated adipocytes as well as of the whole adipose tissue. Mitochondrial DNA transcript levels in adipocytes were measured by quantitative real-time PCR. Western blots of oxidative phosphorylation (OXPHOS) protein levels in adipocytes were performed in obese and lean unrelated individuals. The heavier (BMI 29.9 +/- 1.0 kg/m(2)) co-twins of the discordant twin pairs had more subcutaneous, intra-abdominal and liver fat and were more insulin resistant (p < 0.01 for all measures) than the lighter (24.1 +/- 0.9 kg/m(2)) co-twins. Altogether, 2538 genes in adipocytes and 2135 in adipose tissue were significantly differentially expressed (nominal p < 0.05) between the co-twins. Pathway analysis of these transcripts in both isolated adipocytes and adipose tissue revealed that the heavier co-twins displayed reduced expression of genes relating to mitochondrial pathways, a result that was replicated when analysing the pathways behind the most consistently downregulated genes in the heavier co-twins (in at least 12 out of 14 pairs). Consistently upregulated genes in adipocytes were related to inflammation. We confirmed that mitochondrial DNA transcript levels (12S RNA, 16S RNA, COX1, ND5, CYTB), expression of mitochondrial ribosomal protein transcripts and a major mitochondrial regulator PGC-1 alpha (also known as PPARGC1A) were reduced in the heavier co-twins' adipocytes (p < 0.05). OXPHOS protein levels of complexes I and III in adipocytes were lower in obese than in lean individuals. Subcutaneous abdominal adipocytes in obesity show global expressional downregulation of oxidative pathways, mitochondrial transcripts and OXPHOS protein levels and upregulation of inflammatory pathways. Summary: Uterine leiomyomata (UL) are the most common tumours in women and often result in surgical removal of the uterus. This study identified 22 new genetic loci associated with UL risk, in addition to the previously known 50 loci. The identified genes are involved in development, growth, and cellular senescence, particularly in the smooth muscle cell differentiation and proliferation pathways.

Summary: Nicotinamide riboside (NR), a precursor of nicotinamide adenine dinucleotide (NAD(+)), has shown promise in improving obesity-associated mitochondrial dysfunction and metabolic syndrome. However, short-term clinical trials have not yielded positive results. This study aimed to investigate the long-term effects of NR supplementation on mitochondrial biogenesis and metabolic health in humans. The results showed that NR improved NAD(+) metabolism, muscle mitochondrial number, myoblast differentiation, gut microbiota composition, and epigenetic control of gene expression. However, it did not improve adiposity or metabolic health.

Summary: Refractive error is a complex eye condition caused by genetic and environmental factors. Genome-wide association studies have identified common genetic risk factors, but a significant portion of the heritability is still unexplained. By conducting gene-based association tests, we identified 129 unique genes associated with refractive error in a large dataset. Some of the novel candidates, such as PDCD6IP, PER3, and P4HTM, show promise for future functional studies and validation.

Chronotype and mortality-a 37-year follow-up study in Finnish adults

Summary: According to the Finnish Twin Cohort study, there was a small increase in overall mortality for evening types, but this effect was mainly due to smoking and alcohol. Morning types did not have a significant increase in mortality.

New insights into the treatment of obesity

Summary: Obesity is a chronic disease with increasing prevalence worldwide and is associated with various health risks. The treatment of obesity includes behavioral interventions, pharmacotherapy, and bariatric surgery, but long-term weight maintenance is challenging. Recent advances in understanding the pathophysiology of obesity have led to the development of novel and effective therapies, such as semaglutide and tirzepatide, which have shown promising weight loss effects and improvement in cardiometabolic risk factors. These new agents may bridge the gap between traditional treatments and achieve better outcomes in obesity management.

The association between night shift work and breast cancer risk in the Finnish twins cohort

Summary: Using a discordant twin pair design, this study examined the association between night shift work and breast cancer risk, finding that women who worked night shifts had a 1.58-fold higher risk of breast cancer compared to those who worked only during the day. The highest risk was observed among younger women born between 1950 and 1957. Women with longer sleep and night shift work had the greatest risk of breast cancer.

Summary: Previous Mendelian randomization (MR) studies suggest that educational attainment has beneficial effects on adult health outcomes. However, these estimates may be biased due to population stratification, assortative mating, and indirect genetic effects. Using both population and within-sibship MR approaches, this study provides evidence that genetic liability to educational attainment is associated with decreased BMI, cigarette smoking, and SBP, and potentially with mortality.

Self-reported sleep bruxism and mortality in 1990-2020 in a nationwide twin cohort

Summary: This study investigated the association between sleep bruxism and mortality and found that self-reported sleep bruxism is associated with increased risk of mortality. However, this association is no longer significant after adjusting for other risk factors, suggesting that bruxism is actually a behavior rather than a disease.

The mechanism of assortative mating for educational attainment: a study of Finnish and Dutch twins and their spouses

Summary: Assortative mating refers to the non-random mating of individuals based on their similarity or dissimilarity in certain traits. This study analyzed the mechanisms underlying assortative mating for educational attainment in Finland and the Netherlands. The results showed that both phenotypic assortment and social homogamy play important roles in spouse selection, but phenotypic assortment has a greater impact.

Associations of polygenic inheritance of physical activity with aerobic fitness, cardiometabolic risk factors and diseases: the HUNT study

Summary: There may be genetic factors underlying the associations between higher levels of physical activity (PA) and better aerobic fitness as well as a lower risk for cardiometabolic diseases (CMDs). The study found that genotypes predisposing to higher PA were associated with greater self-reported PA, healthier cardiometabolic profile, and lower incidence of hypertensive diseases, stroke, and type 2 diabetes.

Dementia in former amateur and professional contact sports participants: population-based cohort study, systematic review, and meta-analysis

Summary: This study found that retired amateur athletes have a higher risk of dementia compared to the general population, with professional boxers having the highest risk. Retired wrestlers and soccer players also have an increased risk, although lower in magnitude. Furthermore, professional American football players may have a higher risk compared to amateurs.

Genetic and environmental factors underlying parallel changes in body mass index and alcohol consumption: a 36-year longitudinal study of adult twins

Summary: This study examined the genetic and environmental factors underlying simultaneous changes in body weight and alcohol consumption. The results showed that both genetic and environmental factors play a role in the changes of these traits. There were significant correlations between baseline body weight and change in alcohol consumption, suggesting a direct relationship between the two.
