Obesity-Related Changes in High-Density Lipoprotein Metabolism and Function

In obese individuals, atherogenic dyslipidemia is a very common and important factor in the increased risk of cardiovascular disease. Adiposity-associated dyslipidemia is characterized by low high-density lipoprotein cholesterol (HDL-C) levels and an increase in triglyceride-rich lipoproteins. Several factors and mechanisms are involved in lowering HDL-C levels in the obese state and HDL quantity and quality is closely related to adiponectin levels and the bioactive lipid sphingosine-1-phosphate. Recent studies have shown that obesity profoundly alters HDL metabolism, resulting in altered HDL subclass distribution, composition, and function. Importantly, weight loss through gastric bypass surgery and Mediterranean diet, especially when enriched with virgin olive oil, is associated with increased HDL-C levels and significantly improved metrics of HDL function. A thorough understanding of the underlying mechanisms is crucial for a better understanding of the impact of obesity on lipoprotein metabolism and for the development of appropriate therapeutic approaches. The objective of this review article was to summarize the newly identified changes in the metabolism, composition, and function of HDL in obesity and to discuss possible pathophysiological consequences.

Summary: Atherosclerosis is a disease characterized by increased oxidative stress, leading to modifications of proteins and lipids in the blood vessel wall. Myeloperoxidase (MPO), an enzyme mainly found in neutrophils and monocytes, plays a crucial role in this oxidative process. Research has shown that MPO interacts with lesion derived high-density lipoprotein (HDL) and modifies its components, leading to dysfunction of HDL. Understanding the impact of MPO-induced changes on the structure and function of HDL is important for the future use of HDL in diagnostics and therapy.

However, the biological mechanisms underlying these adverse outcomes are not well understood. Preeclampsia Affects Lipid Metabolism and HDL Function in Mothers and Their Offspring

Summary: Preeclampsia is associated with increased cardiovascular risk for both mother and child. This study investigated the effects of preeclampsia on lipid metabolism, HDL composition, and function in mothers and neonates. Preeclampsia was linked to dyslipidemia in mothers, including high triglycerides and low HDL-cholesterol levels. Mothers with preeclampsia showed shifts in HDL subclasses and increased levels of HDL-associated apolipoprotein C-II. Neonates born to mothers with preeclampsia had increased total cholesterol levels and reduced HDL cholesterol efflux capacity. These findings suggest that preeclampsia affects both maternal and neonatal lipoprotein metabolism.

Myeloperoxidase Alters Lung Cancer Cell Function to Benefit Their Survival

Summary: This study shows that myeloperoxidase (MPO), a neutrophil-derived enzyme, promotes the proliferation and inhibits the apoptosis of lung cancer cells. It functions by increasing the phosphorylation of AKT and ERK, and can be found inside the cells and in the nucleus. Blocking MPO uptake or inhibiting its enzymatic activity reduces its effects on cancer cell function. Animal experiments demonstrate that tumor-bearing mice treated with the MPO inhibitor 4-ABAH have reduced tumor burden.

HDL-Related Parameters and COVID-19 Mortality: The Importance of HDL Function

Summary: This study compares the lipoprotein and metabolic profiles of COVID-19-infected patients and non-COVID-19 pneumonia patients, and finds significant differences in HDL-related parameters between the two groups. The cholesterol efflux capacity (CEC) shows the strongest negative association with COVID-19 mortality.
