Assessment of cytotoxicity biomarkers on the microalga Chlamydomonas reinhardtii exposed to emerging and priority pollutants

The study aimed to investigate the effects of different pollutants on microalgae and to identify sensitive biomarkers that can promote a common cytotoxic response. Results showed that all chemicals tested resulted in a decrease in growth rate, vitality, and photosynthetic efficiency, as well as an increase in autofluorescence and alterations in cell size and complexity.
Contamination of aquatic ecosystems linked to anthropogenic activity is currently a major concern; therefore, ecotoxicological studies are needed to assess its effect on organisms. The main objective of this study was to investigate the effects of different pollutants on microalgae in search of sensitive biomarkers that can promote a common cytotoxic response regardless of the contaminant. Cultures of the freshwater microalga Chlamydomonas reinhardtii were exposed for 24 h to four chemicals, three emerging pollutants (benzophenone-3, bisphenol A and oxytetracycline) and one priority substance (atrazine). A cytometric panel was carried out to assess toxicity biomarkers including cellular growth, inherent cell properties, viability, vitality, cytoplasmic membrane potential and ROS levels. Lipid peroxidation, photosynthetic efficiency and transcriptional responses of photo synthesisand oxidative stress-related genes using RT-qPCR were also studied. Some toxicity responses showed a similar pattern; a decrease in growth rate, vitality and photosynthetic efficiency and an increase in autofluorescence and in the number of cells with depolarised cytoplasmic membrane and were found for all chemicals tested. However, ATZ and OTC provoked a decrease in cell size, whereas BP-3 and BPA caused an increase in cell size, intracellular complexity and ROS levels and a decrease in cell viability. Assayed pollutants generally promoted an overexpression of genes related to cellular antioxidant defence system and a subexpression of photosynthesis-related genes. In addition to the traditional growth endpoint, cell vitality, autofluorescence and gene expression of catalase, glutathione peroxidase and Fe-superoxide dismutase were significantly affected for all chemicals tested, showing a common cytotoxic response. Among the tested substances, BP-3 provoked the strongest cytotoxic alterations on this microalga, pointing out that some emerging contaminants could be more harmful to organisms than priority pollutants.

Summary: Standardised microalgal toxicity assays with bisphenol A showed differences in sensitivity among three marine microalgae species, with Phaeodactylum tricornutum being the most affected and Nannochloropsis gaditana being the least affected. Significant differences were observed between single- and multispecies bioassays for some species, but overall, a similar pattern of alterations was observed in both types of toxicity tests.

Modeling the number of people infected with SARS-COV-2 from wastewater viral load in Northwest Spain

Summary: The quantification of SARS-CoV-2 RNA load in wastewater has emerged as a useful tool to monitor COVID-19 outbreaks in communities. This approach was implemented in the metropolitan area of A Coruna, where wastewater analysis helped track the epidemic dynamics. Regression models allowed estimation of the number of infected individuals and served as an effective early warning tool for predicting outbreaks in the municipality.

Cytotoxicity of BP-3 and BP-4: Blockage of extrusion pumps, oxidative damage and programmed cell death on Chlamydomonas reinhardtii

Unravelling joint cytotoxicity of ibuprofen and oxytetracycline on Chlamydomonas reinhardtii using a programmed cell death-related biomarkers panel

Summary: Pharmaceutical active compounds (PhACs), such as ibuprofen (IBU) and oxytetracycline (OTC), are a growing concern due to their ubiquitous presence and harmful impact on aquatic ecosystems. In this study, the cyto- and genotoxic effects of IBU, OTC, and their binary mixture on Chlamydomonas reinhardtii were investigated. The results showed that the binary mixture caused the most severe damages, including decreased cell proliferation and metabolic activity, increased intracellular ROS levels, and depolarization of cytoplasmic and mitochondrial membranes. Additionally, elevated caspase-like and endonuclease activities and a loss of nuclear DNA integrity were observed. The findings suggest that programmed cell death (PCD) is an important toxicological target for understanding the harmful effects of co-exposure to PhACs in aquatic organisms.

Analysis of the effects of BP-3 and BP-4 on the transcriptome of Chlamydomonas reinhardtii: An RNA-Seq approach

Summary: Benzophenones (BPs), widely-used UV-filters, have been found to alter the physiology of the freshwater microalga C. reinhardtii. RNA-Seq analysis revealed that BPs affected cellular metabolic pathways and gene expression. Both BP-3 and BP-4 caused changes in gene expression, with BP-3 having a higher number of differentially expressed genes. The findings suggest that BPs impair photosynthesis and lead to a shift towards heterotrophic metabolism in microalgae.
