Summary: Chromatin modification plays a crucial role in adult hippocampal neurogenesis by regulating gene expression. HDACs remove acetyl groups from histones, creating a non-permissive chromatin state. Understanding the role of individual HDACs in adult hippocampal neurogenesis is important for controlling stem cell proliferation, neuronal regeneration, and developing more specific HDAC therapeutics for neurological disorders.

Summary: Neuroinflammation does not affect NSCs proliferation but induces aberrant neuronal differentiation, leading to dystrophic, non-functional neurons. PtdCho-enriched media can enhance neuronal differentiation under inflammatory stress, increasing the population of healthy normal neurons and ameliorating neuronal damage to modulate neuronal plasticity. These findings contribute to understanding NSCs behavior under inflammatory conditions and offer new avenues for improving neurogenic capacity in the brain.

Summary: The study investigated the role of CDC25C in neural progenitor cell division and differentiation by knocking down the gene in developing rat brains. The results showed that CDC25C is essential for maintaining the proliferative state of neural progenitor cells during cortical development. Knockdown of CDC25C resulted in early cell cycle exit and premature differentiation of neural progenitor cells, revealing a novel role for CDC25C in NPC division and cell fate determination.

Summary: This study investigated the effects of different micrograting and stiffness combinations on neuronal differentiation and maturation. The results showed that the extracellular matrix-modified PAA-ACA gels can support neuronal cell attachment throughout the differentiation and maturation stages. The interaction between micrograting and stiffness significantly increased the proportion of positive neurons and neurite branching and length.

Summary: The study showed that administration of NPC-CM promoted functional recovery in rats with pMCAO, by suppressing inflammatory damage, inducing endogenous neurogenesis, and reducing cerebral infarct and inflammation. Proteome analysis of NPC-CM identified proteins associated with nervous system development, neurogenesis, and angiogenesis, highlighting the potency of NPC-CM in stroke therapy. Transciptome analysis confirmed the importance of inflammatory response during stroke recovery and validated key hub genes in the interaction network.

Summary: Oligodendrocyte precursor cells (OPCs) are non-neuronal brain cells that give rise to oligodendrocytes, glia that myelinate the axons of neurons in the brain. They are not only involved in myelination but also play diverse roles in the nervous system, including blood vessel formation and antigen presentation. Emerging literature suggests that OPCs are essential for establishing and remodeling neural circuits in the brain, and they have specialized features that allow them to integrate activity-dependent and molecular cues for brain wiring. Studying the communication between neurons and glia, including OPCs, is important for understanding both health and disease.

Summary: This study reveals that upregulation of matrix chondroitin sulfate proteoglycans (CSPGs) induced by SCI impedes neurogenesis of neural precursor cells (NPCs) through the activation of LAR and PTPa receptors. Inhibiting these receptors promotes regeneration of motoneurons and spinal interneurons, improves their morphological maturity and synaptic connectivity, and enhances locomotion and sensorimotor integration after SCI.

Summary: Neurogenesis is a complex process that relies on the balanced execution of expression programs involving various molecular players, including transcription factors, chromatin modulators, and noncoding RNAs. In this study, the function of the long noncoding RNA (lncRNA) Malat1 was investigated during neural embryonic stem cell (ESC) differentiation. Deletion of Malat1 inhibited the proliferation of neural progenitor cells (NPCs) and resulted in increased expression of miR-26a and miR-26b. Inactivation of miR-26a/b rescued the proliferative phenotype of Malat1 knockout cells and accelerated neuronal differentiation. This study reveals an unknown interaction between Malat1 and miR-26 in the regulation of NPC proliferation and neuronal differentiation.

Summary: In the Drosophila model, the proneural transcription factor Asense plays a key role in promoting the transition of neuroepithelial cells to neuroblasts during brain development. This novel action of Asense integrates with well-known signaling mechanisms, providing insights into the regulatory network of neurogenesis. The interaction between master regulators of neurogenesis and Notch signaling differs in simple nervous systems compared to complex systems, suggesting potential evolutionary implications.
