Summary: Bisphosphonate-related osteonecrosis of the jaw (BRONJ) is a serious complication in patients with osteoporosis or metastatic bone cancer. The study found that dietary nitrate, abundant in green vegetables, can protect against BRONJ-like lesions in mice. Nitrate intake increased plasma nitric oxide levels, reducing monocyte necrosis and inflammatory cytokine production. The findings suggest that dietary nitrate can inhibit monocyte necroptosis in BRONJ and promote bone remodeling after injury.

Summary: Peroxiredoxin 5 (Prdx5) plays a significant role in regulating bone resorption and bone formation, and its deficiency leads to osteoporosis. Through immunoprecipitation and LC-MS/MS analysis, hnRNPK was identified as an interacting partner of Prdx5, and Prdx5 negatively regulates the expression of osteocalcin mediated by hnRNPK. Additionally, the loss of Prdx5 enhances the expression of osteoclast-related genes during the differentiation of bone marrow-derived macrophages into multinucleated osteoclasts. These findings suggest that Prdx5 plays an important role in maintaining bone homeostasis by regulating bone cell differentiation.

Summary: Bone fracture healing is a complex process involving hematoma formation, tissue formation, bony callus formation, and bone remodeling. Current therapeutic strategies for fracture repair include autografts, allografts, and tissue engineering. This study suggests that DP2, a disaccharide compound, may enhance fracture healing by promoting osteoblast differentiation and earlier mineralization in vitro. DP2 might serve as a potential therapeutic candidate for bone fracture repair.

Summary: Skeletal progenitor cells rely on collagen-binding integrins and discoidin domain receptors (DDR1 and DDR2) as collagen receptors for bone development and regeneration. This study evaluated specific triple helical peptides containing GFOGER and GVMGFO binding domains to stimulate DDR2 and integrin signaling, as well as osteoblast differentiation. The combination of these peptides cooperatively enhanced both DDR2 and focal adhesion kinase (FAK) signaling, leading to increased osteoblast differentiation. This finding suggests that scaffolds containing DDR and integrin-activating peptides could be a new approach for promoting bone regeneration.
